Reactive oxygen species (ROS) have various biological effects and they are non-linear in characteristic. In high oxidative stress, they may cause cytotoxicity, inhibit cell proliferation, and induce cell death in the form of apoptosis/necrosis; while in low or medium oxidative stress, ROS may cause DNA damage, cell mutation, inflammation, cell proliferation, and eventually they may induce carcinogenesis. Antioxidants are compounds with the ability to reduce ROS. Cell line MCF-7 is one of the breast cancer cell lines that is known to have small amount of antioxidant MnSOD compared to the other cell lines. Low antioxidant MnSOD level in breast cancer cell line MCF-7 leads to low concentration of hydrogen peroxide, because antioxidant MnSOD will convert radical superoxide to hydrogen peroxide. The aim of this research was to analyze oxidants and antioxidants profile in breast cancer cell line MCF-7 and their relationship with cell number. Observations were conducted for 5 days. The cell number was counted with tryphan blue method and haematometer. The concentration of radical superoxide was measured with DHE staining using LSCM tipe Olympus Fluoview FV 1000-Ver 1.7. MnSOD activity, hydrogen peroxide concentration, and catalase activity were measured with ELISA. The results showed that the longer of observation, the greater concentration of oxidants and MnSOD activity, but there was no change in catalase activity. Conclusion the increase in cancer cells number is influenced by radical superoxide.
<p class="Default">Peningkatan transportasi di Indonesia cukup signifikan khususnya di wilayah Sumatera Utara. Hal ini tentu akan berdampak negatif bagi pengguna transportasi jika tidak menerapkan standar keselamatan dalam berkendaraan. Menteri Perhubungan RI telah mengeluarkan Peaturan Nomor 26 tahun 2015 tentang Standar Keselamatan Lalu Lintas dan Angkutan Jalan yang bertujuan meminimalisir terjadinya kecelakaan di jalan raya. Adanya peraturan tersebut tentu dapat meminimalisir terjadinya kecelakaan apabila implementasinya diterapkan dengan baik oleh para pengguna kendaraan termasuk transportasi umum seperti bus ALS. Akan tetapi ada beberapa kejadian kecelakaan yang justru melibatkan bus ALS, sehingga perlu dilakukan kajian penelitian mengenai Implementasi Peraturan Menteri Perhubungan Republik Indonesia Nomor 26 tahun 2015 tentang Standar Keselamatan Lalu Lintas dan Angkutan Jalan, dengan studi kasus pada PT. Antar Lintas Sumatera (ALS) Medan. Metode penelitian ini yaitu destriktif kualitatif, yang menggunakan <em>key informan </em>sebagai subyek<strong> </strong>penelitian, untuk menganalisis<strong> </strong>perkembangan informasi maupun<strong> </strong>sumbernya, maka teknik pengambilan<strong> </strong>sampel yang digunakan <em>snowball sampling</em>. Populasi dalam penelitian adalah seluruh manajemen dan pegawai di PT. ALS Medan, sedangkan sampel yang digunakan hanya sebahagian dari pihak manajemen dan pegawai PT. ALS Medan yang dimulai dari arahan <em>key informan</em>. Hasil penelitian menunjukkan bahwa Implementasi Peraturan tersebut sudah diterapkan oleh pengelola jasa angkutan PT. ALS Medan dengan baik dan benar karena keempat variabel yaitu Komunikasi, Sumber Daya, Disposisi, dan Struktur Birokrasi cukup mendukung berjalannya implementasi kebijakan yang ada diperusahaan. Standar Operasional Prosedur (SOP) perusahaan juga sudah mengadopsi pada Peraturan Menteri Perhubungan Republik Indonesia Nomor 26 tahun 2015.</p>
<span id="docs-internal-guid-25a2977b-7fff-96bd-b93a-19bd55e68ea7"><span>In this research we try to solve the recognition problem in differentiating between batik and its imitation. Batik is an Indonesian heritage of process in making traditional textile product that is now endangered by the existence of imitation products. We try to compare two popular CNN model to classify batik products into five classes. The classes are tulis, cap, print warna, print malam, cabut warna. Tulis and cap are genuine batik, and the other three are an imitation. We realize that this problem is go beyond the recognition of fine grained image problem, it is a hard to identify image problem because even the batik experts is having a hard time identifying batik and its imitation if only based on its picture. The two CNN models, inceptionV3 and mobilenetV2 were trained on three types of image. One type is a freely taken image, the other two were taken based on the experts suggestion. The accuracy score shows that the model trained with the suggestion based picture perform better than the one trained with the random picture.</span></span>
Instia spp is one kind of forest plant which have a high economic value for the development of plantation forest, there fore, there is a need a high seed supply. Plant growth regulators and plant mediums are important aspects in vegetative plant propagation especially by shoot cutting. The objective of this study was to evaluate the effect of plant growth regulators and plant mediums. The study was conducted at the Centre of Plantation Forest Research and Development from Juny to December 2004. The experiment was arranged in Complete Randomize Design with 2/actorial applications. The first factor are plant growth regulators: 0, 20 ppm IBA and !AA meanwhile the second factor are plant mediums: soil+ sand ( 1:1), sand+ compass ( 1: 1) and soil + sand+ compass ( 1: 1: 1 ), with 3 replications and 10 cuttings each. The experiment result showed that Instia spp could be propagated by shoot cutting, plant growth regulators gave better effect on the growth of soot cuttings and the fresh and dry weight of cuttings than untreated with plant medium consist of a high organic matter. ABSTRAKMerbau (Instia spp) merupakan jenis tanaman hutan yang memiliki nilai ekonomi tinggi, dalam pembangunan hutan tanaman memerlukan pengadaan bi bit dalam jumlah ban yak. Sal ah satu upaya dalam pengadaan bibit adalah dengan perbanyakan tanaman secara stek pucuk. Zat pengatur tumbuh dan media tanam merupakan aspek penting dalam perbanyakan tanaman dengan cara tersebut. Penelitian yang bertujuan untuk mengetahui pengaruh zat pengatur tumbuh dan media tanam telah dilakukan di Pusat Litbang Rutan Tanaman sejak Juni sampai Desember 2004. Rancangan penelitian yang digunakan adalah Rancangan Acak Lengkap dengan menguji 2 faktor perlakuan, yaitu faktor pertama zat pengatur tumbuh IBA dan IAA dengan konsentrasi O dan 20 ppm, sedangkan faktor kedua adalah media tanam yaitu campuran tanah + pasir (1: 1), pasir + kompos (1: 1) dan tanah + pasir + kompos (1: 1: 1) dengan ulangan sebanyak 3 kali dan setiap ulangan terdiri dari 10 stek. Parameter yang diamati meliputi pertumbuhan, bobot basah dan bobot kering stek dan volume akar stek. Hasil penelitian menunjukkan bahwa merbau dapat diperbanyak secara stek pucuk dan perlakuan zat pengatur tumbuh dapat meningkatkan pertumbuhan dan bobot segar dan bobot kering stek dengan media tanaman yang mengandung bahan organik tinggi.Kata kunci : Biomasa, media tanam, merbau, stek pucuk, zat pengatur tumbuh.
Obat Pahit is a potion that has been long commonly consumed by Lingga Malay society for generations as stamina keeper. The most dominant plants found in the packaging of the Obat Pahit were namely Bauhunia semibifida, Cnestis palala and Penawa Root (3 species). This research aimed to investigate and determine activity of antioxidant contents in Obat Pahit from five Traditional Medicine Practitioners (TMPs) in the district of Lingga. The tested samples were mashed then being soaked into 2 types of solvent: distilled water and methanol, containing HCl 1%. DPPH method was also used in this research. Quantitatively antioxidant activity test of Obat Pahit from the five TMPs by using methanol solvent had extremely highest activity compared to the distilled water solvent. The test, using TLC plate by spraying the extract from three dominant plants with 0.1 mM of DPPH solution, produced a pale-yellow spots at a wavelength of 366 nm. On the other hand, the test using HPLC at wavelengths of 230 nm and 280 nm showed the presence of two dominant secondary metabolites contents: flavonoid and phenolic. IC50 (ppm) of Bauhinia semibifida (6.6247), Penawa Root (5.0124) and Cnestis palala (5.9968) were much lower than IC50 of mangosteen's rind (41.7675), vitamin C (6.6612) and Stimuno drug (8.333). This antioxidant analysis has not been reported previously. This proof contributed greatly to uncovering potentially native natural resources as an indigenous Indonesian drug which is expected to decrease dependence on imported drugs especially imunomodulator, antihypertensive, antidiabet etc. This research would be beneficial and excellent manifestation for the development of natural antioxidant-based medicines from traditional knowledge of Indonesia's local ethnicities.
The target becomes a self-reliant village must be accompanied by strengthening village ability in supplying the basic needs independently. Vegetables are one of the crucial basic needs to achieve a healthy and prosperous society. The demand for vegetables in Sungai Bawang Village, Kuantan Singingi Regency, is constrained by the available land for vegetable farming. Hydroponic planting system offers the technique without soil that can be implemented anywhere. It has added value in providing clean and hygienic vegetables. Hydroponics technology system of Deep Flow Technique able to provide water and oxygen for plants and suitable for leafy vegetables. Hydroponic is appropriate to be implemented in Sungai Bawang village and did not require huge land in applying this technology. The mentoring strategy is carried out by counseling and demonstration of direct planting stages and the practice by the community. This activity gave a positive influence on the community as well as many people who want to apply this technology. This was the first step in increasing the income of the community, especially farmers. And for the community in general, this technology facilitates the people to get vegetables easily at an affordable price.
Melastoma malabathricum L. is a type of plant naturally grows in Kalimantan that has medicinal properties. Ethanolic extract of M. malabathricum L. flower has quercetin and kaempferol contents that have antioxidant and anticancer activities. But the antioxidant and cytotoxicity activities of M. malabathricum L. fruit ethanolic extract were not known. This study measured the quercetin and kaempferol level in M. malabathricum L. fruit ethanolic extract using HPLC MS/MS, antioxidant activity using DPPH method, and cytotoxicity activity using Brine Shrimp Lethality Test (BSLT) method. Results showed the level of quercetin and kaempferol from M. malabathricum L fruit ethanolic extract are 67.78 µg/g and 43.52 µg/g, respectively. Beside, the antioxidant activity with by IC50 was 16.82±0.24 ppm and 7.38±0.41 ppm. The cytotoxicity activities of M. malabathricum L. fruit ethanolic extract and quercetin are shown by the LC50 of 313.44 ppm (95%CI 283.97-344.43) and 37.24 ppm, respectively.
The implementation of licensing services through Online Single Submission (OSS) at the Binjai City Investment Service and One-Stop Integrated Licensing Service (DPMPPTSP) can still be said to be unsatisfactory because it still has problems in licensing services, especially Construction Services Business Permit (IUJK) services. Problems that are often faced include the completion of the Construction Service Business Permit (IUJK) not on time and business actors do not understand the operation of the OSS system, which results in licensing services through the OSS not yet maximized. This study is to determine the quality of licensing services through OSS at the DPMPPTSP Binjai City and to find out the inhibiting factors for the implementation of licensing through OSS at the Binjai City DPMPPTSP. This research method uses a descriptive method with a qualitative approach. The results of this study indicate the quality of licensing services through OSS at DPMPPTSP of Binjai City which is examined from the dimensions of service quality, namely Tangible, Reliability, Responsiveness, Assurance ( guarantee), Empathy (empathy) as a whole is considered good but not optimal. The DPMPPTSP Of Binajai City is expected to have a technical team located at the Dinas and Dinas to conduct socialization regarding the use of the OSS system to the public so that the public is able to register their own permits and not through third parties/brokers.
