This study investigated Allium subhirsutum L. (AS) anticancer and antioxidant effects and inhibition of tumor angiogenesis in a murine model of skeletal metastases due to inoculation of Walker 256/B cells. Phytochemical composition of AS extract (ASE) was studied by High Resolution-Liquid Chromatography Mass Spectroscopy (HR-LCMS). Total phenolic and flavonoid contents (TPC and TFC) were determined. In vitro, the antioxidant properties were evaluated by reducing power and antiradical activity against DPPH. Cancer cells’ proliferation, apoptosis, metastatic development and angiogenesis were evaluated using Walker 256/B and MatLyLu cells. The p-coumaric acid was the major phenolic acid (1700 µg/g extract). ASE showed high levels of TPC and TFC and proved potent antioxidant effects. ASE inhibited Walker 256/B and MatLyLu cells’ proliferation (Half-maximal inhibitory concentration: IC50 ≃ 150 µg/mL) and induced apoptosis. In silico and in vivo assays confirmed these findings. ASE effectively acts as a chemo-preventive compound, induces apoptosis and attenuates angiogenesis and osteolytic metastases due to Walker 256/B malignant cells.
In this study, we investigate the phytochemical profile, anticancer, and antioxidant activities of Teucrium polium methanolic extract using both in vitro and in silico approaches. The results showed the identification of 29 phytochemical compounds belonging to 13 classes of compounds and 20 tripeptides using High Resolution-Liquid Chromatography Mass Spectrometry (HR-LCMS). 13R-hydroxy-9E,11Z octadecadienoic acid, dihydrosamidin, valtratum, and cepharantine were the main compounds identified. The tested extract showed promising antioxidant activities (ABTS-IC50 = 0.042 mg/mL; 1,1-diphenyl-2-picrylhydrazyl (DPPH)-IC50 = 0.087 mg/mL, β-carotene-IC50 = 0.101 mg/mL and FRAP-IC50 = 0.292 mg/mL). Using both malignant Walker 256/B and MatLyLu cell lines, T. polium methanolic extract showed a dose/time-dependent antitumor activity. The molecular docking approach revealed that most of the identified molecules were specifically binding with human peroxiredoxin 5, human androgen, and human progesterone receptors with high binding affinity scores. The obtained results confirmed that T. polium is a rich source of bioactive molecules with antioxidant and antitumor potential.
The chemical profile of Teucrium polium L. (T. polium) methanolic extract was tested using liquid chromatography coupled with high resolution mass spectrometry (HR-LCMS). Disc diffusion and microdilution assays were used for the antimicrobial activities. Coxsackievirus B-3 (CVB3) and Herpes simplex virus type 2 (HSV-2) were used for the antiviral activities. Chromobacterium violaceum (ATCC 12472 and CV026) and Pseudomonas aeruginosa PAO1 were used as starter strains for the anti-quorum sensing tests. Isoprenoids are the main class of compounds identified, and 13R-hydroxy-9E,11Z-octadecadienoic acid, valtratum, rhoifolin, sericetin diacetate, and dihydrosamidin were the dominant phytoconstituents. The highest mean diameter of growth inhibition zone was recorded for Acinetobacter baumannii (19.33 ± 1.15 mm). The minimal inhibitory concentrations were ranging from 6.25 to 25 mg/mL for bacterial strains, and from 6.25 to 25 mg/mL for Candida species. The 50% cytotoxic concentration on VERO (African Green Monkey Kidney) cell lines was estimated at 209 µg/mL. No antiviral activity was recorded. Additionally, T. polium extract was able to inhibit P. aeruginosa PAO1 motility in a concentration-dependent manner. However, the tested extract was able to inhibit 23.66% of the swarming and 35.25% of swimming capacities of PAO1 at 100 µg/mL. These results highlighted the role of germander as a potent antimicrobial agent that can interfere with the virulence factors controlled by the quorum-sensing systems.
Ticks are blood feeder able to transmit a wide diversity of microbes including pathogens. Therefore, it is of our interest to detect the diversity of microorganisms residing within
ticks using massive sequencing of 16S rDNA. In this study, 200 adult ticks were collected from healthy camels in two localities from Hail province (Saudi Arabia). The analysis showed
high microbial diversity dominated by the two domains (Archaea and Bacteria) associated with Hyalomma dromedarii from both regions. Proteobacteria (61.3%) and Firmicutes (31.2%)
dominated the ticks from the Al Khotha region. While, the microbiome of ticks from the Al Gayed region was dominated by Proteobacteria (81.2%) and Firmicutes (9.2%). Twenty-three
families were identified in the DNA-pool from the Al Gayed region, and was dominated by Pseudomonadaceae (45.37%), and Marinobacteraceae (14.39%) families. Francisellaceae (46%),
Staphylococcaceae (24.26%) dominated the microbiome of the ticks collected from Al Gayed region. Thus, the genera Pseudomonas, Francisella, Proteus, Marinobacter, Glutamicibacter,
Pedobacter, and Staphylococcus are largely distributed in the two identified microbiomes. This study concluded that ticks collected from the studied localities contained a wide range
of microbial communities. These data have a great veterinary and medical importance in near future.
The nanocellulose derived biodegradable plant biomaterial as nano-coating can be used in the medical, biomedical cosmetics, and bioengineering products. Bio-plastic and some synthetic derived materials are edible and naturally biodegradable. The study was conducted to investigate edible nano-biopolymer based nano-coating of capsules and drugs or other definite biomedical materials from corn leaf biomass. Corn leaf biomass was used as an innovative sample to produce edible nano-coating bioplastic for drug and capsule coating and other industrial uses. The data show the negligible water 0.01% absorbed by bio-plastic nanocoating. Odor represented by burning test was under the completely standard based on ASTM. Moreover, data on color coating, tensile strength, pH, cellulose content have been shown under standard value of ASTM (American standard for testing and materials) standard. In addition to that data on the chemical element test like K+, normalCO3−−, Cl-, Na+ exhibited positive data compared to the synthetic plastic in the laboratory using the EN (166)) standardization. Therefore, it can be concluded that both organic (cellulose and starch) based edible nano-coating bioplastic may be used for drug and capsule coating as biomedical and medical components in the pharmaceutical industries.
In this study, the antibacterial activity of selected medicinal plants and their synergistic effect with antibiotics were investigated. Different antibiotic resistant bacterial species were employed including: Escherichia coli (E. coli), Klebsiella pneumoniae (K. pneumonia), Acinetobacter baumannii (A.baumannii), Pseudomonas aeruginosa (P. aeruginosa), Staphylococcus aureus (S. aureus) and Salmonella spp. The results indicated that methanol extracts of most of the tested plants inhibited growth of the tested bacteria, however, the highest inhibition was by Senna followed by Harmal, saffl ower and Guaada, where the inhibition zone diameter was 8 mm, 4 mm, 3 mm, 2 mm and 2 mm, respectively. The synergism between plants extract and antibiotics resulted in inhibition of the tested organisms; generally, the synergism with Harmal extract was more effective in inhibition of antibiotic-resistant bacteria, followed by Senna and fi nally Saffl ower. The outcomes additionally demonstrated that methanolic concentrate of the plant has more inhibitory impact than that of the water extract. Further investigation of the plants extract to seclude and recognize the dynamic fi xings is prescribed.
Environmental factors such as temperature and humidity directly affect the growth and fruit bodies of fungi. We studied the diversity of wood decaying fungal species, which have grown on same substrate in forest as well as laboratory environment. Ten specimens of fruit body of wood-decaying fungi and 24 random pieces of coarse wooden debris were collected from the forest of northwest Arkansas. The samples of coarse woody debris were incubated in laboratory-growth chambers for two months to promote the fungal growth. Fourty-two different species of wood-decay fungal isolates were recovered and identified by internal transcribed spacer (ITS) region sequencing. The isolates from the forest belonged to twenty-two different taxa whereas twenty taxonomic groups were reported from the growth compartments. Remarkably, data observed from two sets did not shared any taxon. These results indicated that environmental growth conditions play crucial role on fungal diversity even if grown on same substrates.
