A comparative analysis of caecum and crop microbiota of chick, grower and adult stages of Indian indigenous chickens was conducted to investigate the role of the microbiota of the gastrointestinal tract, which play an important role in host performance, health and immunity. High-throughput Illumina sequencing was performed for V3, V4 and V4-V6 hypervariable regions of the 16S rRNA gene. M5RNA and M5NR databases under MG-RAST were used for metagenomic datasets annotation. In the crop, Firmicutes (~78%) and Proteobacteria (~16%) were the predominant phyla whereas in the caecum, Firmicutes (~50%), Bacteroidetes (~29%) and Actinobacteria (~10%) were predominant. The Shannon-Wiener diversity index suggested that sample richness and diversity increased as the chicken aged. For the first time, the presence of Lactobacillus species such as L. frumenti, L. antri, L. mucosae in the chicken crop along with Kineococcus radiotolerans, Desulfohalobium retbaense and L. jensenii in the caecum are reported. Many of these bacterial species have been found to be involved in immune response modulation and disease prevention in pigs and humans. The gut microbiome of the indigenous chicken was enriched with microbes having probiotic potential which might be essential for their adaptability.

The study aimed to investigate the effect of feed supplements, viz Lactobacillus plantarum LGFCP4 (laboratory isolate from GIT of Guinea fowl), Lactobacillus acidophilus (NCDC, Karnal) and in-feed antibiotic bacitracin methylene disalicylate (BMD) on growth performance, FCR, carcass traits and immune organs weight, intestinal histomorphometry and gastrointestinal microflora population in broiler chickens. In a completely randomized design, CARIBRO-Dhanraja broiler chicks (n = 160) were used with four treatment groups. During the entire experimental duration of 35 days, treatment groups were provided with different dietary treatments (T1 - basal diet (negative control), T2 - antibiotic growth promoter BMD 20 g/100 kg feed (positive control), T3 - 1 × 10 cfu of L. acidophilus/gm-fermented feed +MOS 1 g/kg feed and T4 - 1 × 10 cfu of laboratory-isolated L. plantarum LGFCP4/gm-fermented feed+ MOS 1 g/kg feed. After 35 days of experimental period, no significant results have been observed in different growth performance traits among treatment groups. Cut-up parts and edible organs' weight remained unaffected by dietary supplementation, whereas weight of immune organs were significantly higher (p < 0.05) in L. plantarum LGFCP4-supplemented group. At the end of feeding trial, significantly (p < 0.05) lower E. coli count was observed in crop of T4 birds, while in ileum, T2 and T3 showed lower count. In caeca, T2 group showed lowest E. coli count. Salmonella count in crop and ileum was significantly (p < 0.05) low in T3 and T4, while in caeca, T2 group showed lowest count. In terms of histomorphometry, duodenal villous height (VH), crypt depth (CD) and VH:CD ratio were higher for T3 and T4 and lowest values were obtained for T2 group. The results of the study showed that L. plantarum LGFCP4 isolated from GIT of guinea fowl can effectively replace in-feed antibiotic growth promoters in broiler diets by altering intestinal villi morphology and improving the gut health by reducing the pathogenic microbial load.

This study investigated the role of dietary prebiotic mannan-oligosaccharides (MOS), and probiotic Bifidobacterium bifidum (BFD) in lipid metabolism, deposition, and consequent health indices in broiler chicken. The supplementation of 0.2% MOS along with either 106 or 107 CFU BFD/g feed resulted in downregulation of Acetyl-CoA carboxylase, fatty acid synthase, sterolregulatory element binding protein-1, and apolipoprotein B100; and up-regulation of peroxisome proliferator activated receptor-α AMP-activated protein kinase α-1, and stearoyl CoA (∆9) desaturase-1 hepatic expression in broiler chicken. The birds supplemented with 0.2% MOS along with either 106 or 107 CFU BFD/g feed depicted lower body fat percentage, palmitic acid, stearic acid, and saturated fatty acid contents, whereas, higher palmitoleic acid, oleic acid, and MUFA contents were observed. The ∆9-desaturase indices of chicken meat have shown higher values; and elongase index (only thigh) and thioesterase index have shown lower values in birds supplemented with 0.2% MOS along with either 106 or 107 CFU BFD/g feed. The meat health indices such as Polyunsaturated fatty acids (PUFA)/Saturated fatty acids (SFA) ratio, Mono-saturated fatty acids (MUFA)/SFA ratio, unsaturated fatty acids (UFA)/SFA ratio, hypocholesterolemic/hypercholesterolemic fatty acid ratio, saturation index, atherogenic index, thrombogenic index, and hypercholesterolemic fatty acid content were positively improved in birds supplemented with 0.2% MOS along with either 106 or 107 CFU BFD/g feed. Similarly, the birds supplemented with 0.2% MOS along with either 106 or 107 CFU BFD/g feed have shown lower serum triglyceride and total cholesterol levels along with higher high density levels and improved serum health indices cardiac risk ratio, atherogenic coefficient, and, atherogenic index of plasma.

A total of 32 Lactobacillus isolates, 8 each from the crop (LGFCP1-LGFCP8), proventriculus (LGFP9-LGFP16), ileum (LGFI17-LGFI24) and caeca (LGFCM25-LGFCM32) were isolated from 25 adult guinea fowl (Pearl variety), 22-28 weeks of age, and characterised morphologically, physiologically, biochemically and by molecular methods. Isolates were screened for their probiotic quality using range of in vitro tests: aggregation test, cell surface hydrophobicity, resistance to bile salts and acidic conditions, enzymatic tests and coaggregation and antagonistic test. Based on in vitro test results and a novel scoring method, the two best isolates were selected and partial 16S rRNA sequencing was done. BLAST (Basic Local Alignment Search Tool) analysis of sequence of isolate LGFCP4 showed 99% genetic identity with Lactobacillus plantarum and LGFP16 with Lactobacillus reuteri. The study shows that these two microbial agents may be suitable as potential probiotic candidates in guinea fowl, as well as in a feed supplement for other poultry species.

Aim: The present experiment was conducted to study the effect of different tropical stress conditions on haemato-biological traits in various broiler strains during 3 to 4 weeks of age. Introgressing some important major genes likes Naked neck (Na) and Frizzle (F) into broiler germplasm may substantially improve the heat tolerance. Materials and Methods:The experiment was designed to evaluate three indigenously developed broilers viz. CARIBRO-Tropicana (Naked neck and Frizzle gene bearing), CARIBRO-Mritunjai (Naked neck gene bearing) and CARIBRO-Vishal (Normal plumaged) under different THI (i.e. 72, 85 and 91) for 4 hours daily for 7 days. Total 324 broiler chicks (i.e. 36 chicks in each group) of 3 weeks of age were used in this study. Results:The m-RNA expression of HSP-70 gene was observed highest in frizzle plumaged birds and lowest in normal plumaged birds. The CARIBRO-Vishal showed highest stress as compared to other group. Higher the THI more severe was the effect on the traits. During the 7 day of exposure trial birds of all the genetic group exhibited the phenomenon of acclimatization as reveled by the averages of various traits at different days into the exposure. The mRNA expression analysis of HSP-70 in liver revealed higher expression levels at later days during the exposure trials which indicated the phenomenon of memory (stress memory) and acquired thermotolerance. Conclusion:Among the three genetic groups, CARIBRO-Tropicana exhibited highest means for HSP-70 production as well as tolerated the heat stress in a better way; therefore CARIBRO-Tropicana was adjudged to be the best genetic group for production under tropical climate.

A field experiment was carried out during three consecutive years 2008 to 2009 and 2010 to 2011 at two locations; one at University research centre and another at farmers' fields in Meerut and Muzaffarnagar district of western Uttar Pradesh, India. The objective of the study was to compare the conventional planting methods (22.5 cm) and broadcasting method of wheat sowing with drill planting method [15.0, 17.5, 20.0 cm and paired row (15:25 cm)] for grain yield and its parameters. The experiment comprised of six (15, 17.5, 20, 22.5 and 15:25 cm paired spaced rows and broad casting) treatments arranged in randomized complete block design with three replications in a net plot size of 13 × 8 m. The results over the years of the study revealed that, the germination were statistically at par in drill sowing at 17.5 cm apart rows and broadcasting. Better plant height was noted in drill planting with 17.5, 20 cm rows and 15:25 cm paired rows. However, number of spikelets spike -1 and number of grains spike -1 were statistically similar in drilling at 17.5, 20, and 15:25 cm paired apart rows. Similarly, 1000 grain weight was recorded in drill sowing at 20 cm and 15:25 cm paired rows. The maximum grain yield was obtained through 15:25 cm paired rows drill planting method and it was statistically at par with drill planting method where row spacing was 20 cm. Whereas, drill-planting techniques with row spacing 15 cm was inferior to broadcast method. It may be concluded that, 15:25 cm paired rows and 20 cm drill planting method is suitable for wheat sowing in sandy loam soils of irrigated areas of western Uttar Pradesh.
