Parts of the large phosphorylated tegument protein, pp150, of human cytomegalovirus (HCMV) were expressed in bacteria. The resulting fusion proteins were tested in a Western blot (immunoblot) assay for reactivity with a monoclonal antibody against pp150, with a polyspecific rabbit antiserum, and with human reconvalescent-phase sera. Those fusion proteins that performed well in the Western blot assay were used as antigens in enzyme-linked immunosorbent assays (ELISAs) for the detection of antibodies against HCMV. Five different recombinant beta-galactosidase fusion proteins were evaluated by ELISA using 62 seropositive and 38 seronegative human serum samples. Of all the proteins tested, one peptide representing 162 amino acids of pp150 was superior to the others with regard to sensitivity and specificity. All sera known to be positive for antibodies against HCMV were identified by combining the results of the ELISAs with the different pp150 fusion proteins. Therefore, it appears that peptides from a single protein of HCMV might be sufficient to identify HCMV-seropositive individuals by recombinant ELISA.
Zusammenfassung
In der vorliegenden Arbeit wurden die serologischen Verfahren der Immundiffusion (ID) nach Ouchterlony und des ELISA zur Diagnostik der enzootischen Rinderleukose angewandt. Untersuchungsmaterialien waren sowohl Blut‐ als auch Milchserum. Die wichtigsten Ergebnisse sind wie folgt zusammenzufassen:
Die schon bekannte höhere Empfindlichkeit der ELISA‐Methodik gegenüber der ID wurde bestätigt. Bei Untersuchung von Blutserum erfaßte die ID maximal 76,2% der im ELISA gegen Leukoseantigen positiven Rinder.
Auch an Milchserum war die Treffsicherheit der ELISA‐Methodik höher als diejenige der ID an Blutserum. Sie betrug im Einzeltiervergleich unabhängig vom Laktationsstadium 85,1% gegenüber 76,2%, beide wiederum bezogen auf den ELISA‐Test an Blutserum. Die Anwendung der ELISA‐Technik an Milch erwies sich damit der ID an Blut diagnostisch überlegen. Vergleichbare Untersuchungen und Ergebnisse sind in der uns bekannten Literatur bisher nicht publiziert.
Trotz des günstigen Einzeltiervergleichs war die Frage nach der Eignung der ELISA‐Methodik an Milch als bestandsdiagnostisches Verfahren für die staatliche Bekämpfung der Rinderleukose nicht eindeutig abzuklären. Hierzu sind weitere langfristige Untersuchungen an umfangreicherem Material eingeleitet.

Comparative serological studies on blood and milk for diagnosis of enzootic bovine leucosis
In the present paper the immunodiffusion (ID) according to Ouchterlony and the ELISA tests were applied in the diagnosis of enzootic bovine leukosis. Blood sera and milk whey were tested. The following results can be summarized:
The higher sensitivity of ELISA in comparison to ID was confirmed. ID on blood serum detected 76.2 per cent (128 of 168) of the ELISA positive animals in blood serum.
ELISA on milk serum had a higher positive rate than ID on blood serum. ELISA on milk serum detected 85.1 per cent (143 of 168) of the ELISA positive animals with blood serum.
There is an advantage with the ELISA on milk serum when comparing single animals compared with the ID on blood serum. Whether this advantage can be used for herd diagnosis and eradication programs will be investigated in a further study.
