Differential gene expression can be investigated effectively by cDNA arrays. Because tissue homogenates result inevitably in an average expression of a bulk of different cells, we aimed to combine mRNA profiling with cell-type-specific microdissection. Using a polymerase chain reaction (PCR)-based preamplification technique, the expression profile was shown to be preserved. We modified the existing protocol enabling to apply the total amount of extracted RNA from microdissected cells. A mean amplification factor of nearly 1000 allowed to reduce the demand of initial RNA to ϳ10 ng. This technique was used to investigate intrapulmonary arteries from mouse lungs (ϳ500 cell equivalents). Using filters with 1176 spots, three independent experiments showed a high consistency of expression for the preamplified cDNAs. These profiles differed primarily from those of total lung homogenates. Summary Brain tissue from 187 animals of different species was investigated by means of fluorescent antibody test, peroxidase anti‐peroxidase technique, mouse inoculation test and cell culture technique for a diagnosis of rabies. With peroxidase anti‐peroxidase technique the rabies specific reaction comprised inclusion bodies and a granular staining of the cytoplasm of affected cells. A specific positive reaction was found only in neurons, in which pericaryon as well as cell processes were affected. Fluorescent antibody test and peroxidase anti‐peroxidase technique detected 98 % each, mouse inoculation test 95 % and cell culture technique 81 % of the rabies positive animals. In conclusion, peroxidase anti‐peroxidase technique allows a highly reliable diagnosis of rabies when only formalin‐fixed and paraffin‐embedded material is available. Histopathological alterations comprising Negri bodies, inflammatory and degenerative lesions were encountered in 53 % of the rabies positive brains. Zusammenfassung Vergleìch von funf verschiedenen Methoden zur Routinediagnostik der Tollwut Gehirnproben von 187 Tieren verschiedener Spezies wurden mit Hilfe des Immunfluoreszenztests, der Peroxidase‐Anti‐Peroxidase‐Technik, des Mäuseinokulationstests und der Zellkulturtechnik auf Tollwut untersucht. Einschlußkörperchen und eine granuläre Anfärbung des Zytoplasmas betroffener Zellen stellten die beiden spezifischen positiven Reaktionen bei der Peroxidase‐Anti‐Peroxidase‐Technik dar. Eine spezifisch positive Reaktion fand sich ausschließlich in Ganglienzellen, in denen sowohl das Perikaryon als auch die Fortsätze betroffen waren. Die Nachweisrate tollwutpositiver Fälle betrug im Immunfluoreszenztest und bei der Peroxidase‐Anti‐Peroxidase‐Technik jeweils 98%, beim Mäuseinokulationstest 95% und bei der Zellkulturtechnik 81%. Die Peroxidase‐Anti‐Peroxidase‐Technik ermöglicht eine zuverlässige Diagnose der Tollwut in den Fällen, in denen ausschließlich formalinfixiertes und in Paraffin eingebettetes Untersuchungsmaterial zur Verfügung steht. Histopathologische Veränderungen in Form von Negrischen Körperchen sowie entzündlichen und degenerativen Prozessen fanden sich bei 53 % der tollwutpositiven Tiere.
Acute alveolar hypoxia causes pulmonary vasoconstriction that matches lung perfusion to ventilation to optimize gas exchange. Chronic alveolar hypoxia induces pulmonary hypertension, characterized by increased muscularization of the pulmonary vasculature and right ventricular hypertrophy. Elevated erythropoietin (EPO) plasma levels increase hematocrit and blood viscosity and may affect structure and function of the pulmonary circulation. To differentiate between the direct effects of hypoxia and those linked to a hypoxia-induced increase in EPO/hematocrit levels, we investigated the lung vasculature in transgenic mice constitutively over-expressing EPO (termed tg6) upon exposure to normoxia and chronic hypoxia. Despite increased hematocrit levels (approximately 0.86),tg6 mice kept in normoxia did not develop selective right ventricular hypertrophy. The portion of vessels with a diameter of 51-95 microm and >155 microm was increased whereas the portion of small vessels (30-50 microm) was decreased. Pulmonary vascular resistance and the strength of hypoxic vasoconstriction measured in isolated perfused lungs were decreased. Vasoconstrictions induced by the thromboxane mimetic U46619 tended to be reduced. After chronic hypoxia (FiO2 = 0.10, 21 days), vascular resistance and vasoconstrictor responses to acute hypoxia and U46619 were reduced in tg6 mice compared to wildtype controls. Chronic hypoxia increased the degree of pulmonary vascular muscularization in wildtype but not in tg6 mice that already exhibited less muscularization in normoxia. In conclusion, congenital over-expression of EPO exerts an "anti-pulmonary hypertensive" effect, both structurally and functionally, particularly obvious upon chronic hypoxia.
Zusammenfassung In der vorliegenden Arbeit wurden die serologischen Verfahren der Immundiffusion (ID) nach Ouchterlony und des ELISA zur Diagnostik der enzootischen Rinderleukose angewandt. Untersuchungsmaterialien waren sowohl Blut‐ als auch Milchserum. Die wichtigsten Ergebnisse sind wie folgt zusammenzufassen: Die schon bekannte höhere Empfindlichkeit der ELISA‐Methodik gegenüber der ID wurde bestätigt. Bei Untersuchung von Blutserum erfaßte die ID maximal 76,2% der im ELISA gegen Leukoseantigen positiven Rinder. Auch an Milchserum war die Treffsicherheit der ELISA‐Methodik höher als diejenige der ID an Blutserum. Sie betrug im Einzeltiervergleich unabhängig vom Laktationsstadium 85,1% gegenüber 76,2%, beide wiederum bezogen auf den ELISA‐Test an Blutserum. Die Anwendung der ELISA‐Technik an Milch erwies sich damit der ID an Blut diagnostisch überlegen. Vergleichbare Untersuchungen und Ergebnisse sind in der uns bekannten Literatur bisher nicht publiziert. Trotz des günstigen Einzeltiervergleichs war die Frage nach der Eignung der ELISA‐Methodik an Milch als bestandsdiagnostisches Verfahren für die staatliche Bekämpfung der Rinderleukose nicht eindeutig abzuklären. Hierzu sind weitere langfristige Untersuchungen an umfangreicherem Material eingeleitet. Summary Comparative serological studies on blood and milk for diagnosis of enzootic bovine leucosis In the present paper the immunodiffusion (ID) according to Ouchterlony and the ELISA tests were applied in the diagnosis of enzootic bovine leukosis. Blood sera and milk whey were tested. The following results can be summarized: The higher sensitivity of ELISA in comparison to ID was confirmed. ID on blood serum detected 76.2 per cent (128 of 168) of the ELISA positive animals in blood serum. ELISA on milk serum had a higher positive rate than ID on blood serum. ELISA on milk serum detected 85.1 per cent (143 of 168) of the ELISA positive animals with blood serum. There is an advantage with the ELISA on milk serum when comparing single animals compared with the ID on blood serum. Whether this advantage can be used for herd diagnosis and eradication programs will be investigated in a further study. Résumé Recherches sérologiques comparées avec le sang et le lait pour le diagnostic de la leucose enzootique bovine Les tests sérologiques d'immunodiffusion (ID) selon Ouchterlony et ELISA ont été utilisés pour le diagnostic de la leucose enzootique bovine. Le matériel testé fut aussi bien du sérum sanguin que lacté. Les résultats les plus importants peuvent se résumer comme suit: La sensibilité plus forte déjà connue de la méthode ELISA par rapport à l'ID a été confirmée. Lors de recherches sur le sérum sanguin, l'ID n'a détecté que le 76,2% des bovins réagissant positivement à l'antigène de la leucose avec ELISA.
