DNA Barcoding is recommended as a tool for identifying and confirming species within the taxonomy framework. The rbcL gene is the barcode DNA for plant species. Ternate Island is one of clove plantation center in North Maluku. The diversity and productivity of cloves on the Ternate island known since in earlier times. This study has a purpose to amplify the rbcL gene in clove plants collected from clove plantations of communities in Ternate island. Isolation of total DNA carried out with using ZymoBiomic (Zymo Research DNA Extraction) Kit from leaves tissue, then DNA specimen is amplified based on rbcL gene with forwarding sequence rbcLaF 5′-ATG TCA CCA CAA ACA GAG ACT AAA GC-3′ and reverse sequence is rbcLaR 5′-GTA AAA TCA AGT CCA CCR CG-3′. The results showed that the specimen was successfully amplified with an amplicon size of 600 bp. Furthermore, BLASTN analysis results note that the sequence has a similarity of 99% with Syzygium cumini vc. J.R. Abbott 23676 (FLAS) ribulose-1,5-bisphosphate carboxylase large subunit (rbcL) gene, but phylogenetically the precise position of sample could not found because the limitation of rbcL gene.
At present, coconut shell charcoal briquettes are one of the alternative energy sources that are in demand by the export market. The purpose of this research was to produce an alternative briquette product based on local natural materials. Kenari shell is a waste of natural materials or biomass. The characteristics of Kenari shells have the same structure with coconut shell. The pre-design of briquette production of this Kenari shell used pre-treatment process stages, charcoal making, destruction, printing with adhesive variation, drying, and SNI quality of briquette quality analysis. The results of pre-design of bio-briquette production of Kenari shell showed that the application of the method and the basic ingredients of waste could produce the product of charcoal briquettes equal to charcoal briquettes of coconut shell.
Tujuan: Penelitian ini bertujuan melihat best-practices implementasi lesson study dalam memfasilitasi pembelajaran yang berpusat pada peserta pada mata pelajaran IPS Ekonomi. Metode: Studi deskriptif kualitatif ini dilakukan di SMAN 10 Ternate selama bulan Agustus-November 2021 dengan melibatkan subyek sebanyak 36 siswa kelas X IPS1. Pengumpulan data menggunakan teknik observasi dengan menggunakan instrument berupa lembar observasi, rekaman proses pembelajaran, dan catatan harian peneliti yang kemudian dianalisis secara kualitatif. Temuan: hasil penelitian menunjukkan bahwa lesson study mampu memfasilitasi pembelajaran yang berpusat pada peserta didik melalui setiap implementasi pada tahapan designing, action, and reflection. Kesimpulan: Lesson study sebagai model peningkatan kompetensi guru melalui pengkajian praktik pembelajaran memberi kontribusi positif bagi guru dan peserta didik di kelas X IPS1 pada mata pelajaran Ekonomi. Lesson study memberi ruang kepada guru untuk mengembangkan kreatifitasnya dalam melakukan perubahan dan perbaikan. Sementara bagi peserta didik, lesson study memberi ruang hak belajar untuk difasilitasi mengembangkan potensi yang dimilikinya. Hasil penelitian ini menjadi rekomendasi bagi perbaikan kualitas pembelajaran di Sekolah Menengah Atas di Kota Ternate, khususnya pada mata pelajaran IPS.
The nutmeg extract had been recognized as the source of antioxidant and antimicrobial activity. The aims of this study were to obtain endophytic fungi isolated from the mace and seed of Nutmeg (Myristica fragrans Houtt) and to determine antifungal activity of those isolates. Microbiol routine assay were conducted and it consists of three stages, namely isolation of endophytic fungi, identification of fungus and antifungal activity test by using dual culture. Sixteen endophytic fungus isolates were found in this study. Those are identified as Alternaria sp (BPB 1.
The Biochemistry course is a basic and compulsory subject for Biology Education students. This study aims to determine students' level of participation and problem-solving skills through the ability to solve cases of Biomolecules and proteins on Student Worksheets (LKM). This is an Action Research integrated with lesson study with two cycles. The instruments used were student participation rubrics in the case method and LKM problem-solving skills in the Biochemistry course. The subjects in this study were 24 third-semester students of the Biology Education Study Program. Data analysis techniques through descriptive statistics with percentages. The results showed an increase in the percentage from cycle 1 to cycle 2. Student participation in collaborative activities increased from 71.5% to 73.3%, while problem-solving skills increased from 53.5% to 71.7%. Based on the observer transcripts, some students still do not understand the material and cases discussed in lectures in class and the laboratory. This is because students do not actively ask questions and do not have curiosity about the material and case explanations presented by lecturers or fellow students. This study shows that implementing the integrated case method in lesson study can increase student participation activities and problem-solving skills in Biochemistry courses.Abstrak Matakuliah Biokimia merupakan matakuliah dasar dan wajib bagi mahasiswa Pendidikan Biologi. Penelitian ini bertujuan untuk mengetahui tingkat partisipasi dan problem-solving skill mahasiswa melalui kemampuan menyelesaikan kasus-kasus konsep Biomolekul dan protein pada Lembar Kerja Mahasiswa (LKM). Ini adalah penelitian Action Reseach terintegrasi lesson study dengan dua siklus. Instrumen yang digunakan adalah rubrik pertisipasi mahasiswa dalam case method dan LKM problem-solving skill matakuliah Biokimia. Subjek dalam penelitian ini adalah 24 mahasiswa semester 3 prodi Pendidikan Biologi. Teknik analisis data melalui statistik deskriptif dengan persentase. Hasil penelitian menunjukkan peningkatan persentasi dari siklus 1 ke siklus 2. Aktivitas partisipasi mahasiswa dalam kolaborasi dari 71,5% menjadi 73,3%, sedangkan problem-solving skill mahasiswa dari 53,5% menjadi 71,7%. Berdasarkan transkrip observer masih ada mahasiswa yang kurang memahami materi dan kasus yang didiskusikan dalam perkuliahan di kelas maupun di laboratorium. Hal ini karena mahasiswa tidak aktif bertanya dan tidak memiliki rasa ingin tahu terhadap materi dan penjelasan kasus yang disampaikan oleh dosen atau sesama mahasiswa. Penelitian ini menunjukkan bahwa implementasi case method terintergrasi dalam lesson study dapat meningkatkan aktivitas partispasi dan problem-solving skill mahasiswa pada matakuliah Biokimia.
Bamboo is a versatile plant and occupies a special place in the life of indonesian people. Bamboo is also a fairly available and inexpensive raw material for making household appliances and furniture, building materials, pipes for water distribution, musical instruments and religious purposes. In addition, some types of bamboo are ornamental plants and waste filter processors and erosion prevention. Bamboo belongs to the results of non-timber forests and can be used as an alternative to wood. This research aims to find out the diversity of bamboo plants and the form of utilization of bamboo plants in Cobodoe Village, East Tidore Subdistrict. This research is a descriptive study, with survey methods used to describe the diversity and form of utilization of bamboo plants in Cobodoe Village, East Tidore District. The results of this study showed that the diversity of bamboo types in Cobodoe Village was categorized as low with an H' value of 1.03. There are three types of bamboo, namely Spotted bamboo (Bambusa maculate), Bamboo Tui (Melocanna hamilis Rpr.), and Bamboo Fence (Bambusa glaucescens). People in Cobodoe Village use bamboo plants as raw materials for food and household furniture.
