The Araucaria Forests in southern Brazil are part of the Atlantic Rainforest, a key hotspot for global biodiversity. This habitat has experienced extensive losses of vegetation cover due to commercial logging and the intense use of wood resources for construction and furniture manufacturing. The absence of precise taxonomic tools for identifying Araucaria Forest tree species motivated us to test the ability of DNA barcoding to distinguish species exploited for wood resources and its suitability for use as an alternative testing technique for the inspection of illegal timber shipments. We tested three cpDNA regions (matK, trnH-psbA, and rbcL) and nrITS according to criteria determined by The Consortium for the Barcode of Life (CBOL). The efficiency of each marker and selected marker combinations were evaluated for 30 commercially valuable woody species in multiple populations, with a special focus on Lauraceae species. Inter- and intraspecific distances, species discrimination rates, and ability to recover species-specific clusters were evaluated. Among the regions and different combinations, ITS was the most efficient for identifying species based on the ‘best close match’ test; similarly, the trnH-psbA + ITS combination also demonstrated satisfactory results. When combining trnH-psbA + ITS, Maximum Likelihood analysis demonstrated a more resolved topology for internal branches, with 91% of species-specific clusters. DNA barcoding was found to be a practical and rapid method for identifying major threatened woody angiosperms from Araucaria Forests such as Lauraceae species, presenting a high confidence for recognizing members of Ocotea. These molecular tools can assist in screening those botanical families that are most targeted by the timber industry in southern Brazil and detecting certain species protected by Brazilian legislation and could be a useful tool for monitoring wood exploitation.
Este trabalho apresenta o estudo taxonômico das espécies de Ocotea (Aubl.) no estado do Paraná, Sul do Brasil. São fornecidos chave de identificação, descrições, ilustrações e comentários, que incluem distribuição geográfica, habitat, estado de conservação e época de floração e frutificação. O estudo confirmou 31 espécies de Ocotea no estado do Paraná. A Floresta Atlântica tem o maior número de espécies (26) sendo onze endêmicas, seguida pela Floresta com Araucária (14) com uma endêmica, Floresta Estacional (11), Cerrado (7) com duas endêmicas e Campos (1). Duas espécies correm risco de extinção local. Ocotea daphnifolia (Meisn.) Mez e Ocotea velutina (Nees) Rohwer são citadas pela primeira vez no Paraná.
RESUMO O Morro dos Perdidos está localizado no município de Guaratuba, estado do Paraná (25º45'-25º50'S e 49º03'-49º06'O), abrange uma área de 1440 hectares de Floresta Ombrófila Densa, com altitudes entre 767 a 1439 m. Na área, a família Lauraceae está representada por 15 espécies: Cinnamomum hatschbachii; Cryptocarya aschersoniana; Endlicheria paniculata; Nectandra puberula; Ocotea bicolor; O. catharinensis; O. elegans; O. nunesiana; O. odorifera; O. porosa; O. pulchella; O. tristis; O. vaccinioides; Ocotea sp e Persea willdenovii. São apresentados chave, descrições, ilustrações e comentários sobre as espécies.
Resumo Lauraceae está entre as três principais famílias do componente arbóreo das florestas no Paraná, sendo elemento frequente nos estudos fitossociológicos e biogeográficos. Apesar disso, a família continua insuficientemente conhecida no estado. Nossos objetivos foram: reconhecer as espécies de Lauraceae no Paraná, relacionando-as com as unidades fitogeográficas; mapear as regiões com maior riqueza de espécies e concentração de endemismos; identificar áreas de complementaridade e compará-las com as áreas prioritárias para conservação. Para isso, nós utilizamos registros de coleções de 23 herbários nacionais, totalizando 3.332 espécimes analisados, plotando suas coordenadas no mapa fitogeográfico do estado. Constatamos que no Paraná ocorrem 76 espécies, das quais onze são citadas pela primeira vez no estado. A Floresta Ombrófila Densa abrange o maior número de espécies (59 spp.), seguida pela Floresta Ombrófila Mista (31 spp.), Floresta Estacional Semidecidual (23 spp.), Savana (9 spp.) e Estepe (2 spp.). As regiões da Serra do Mar e Vale do Ribeira apresentam a maior riqueza e endemismo. O Parque Estadual das Lauráceas é uma unidade de conservação estratégica, pois abriga dois terços das espécies paranaenses, com sete endêmicas.
