-Cryopreservation is a viable option for conservation of coffee germplasm. However, for this technique to be completely successful, it is of fundamental importance to carry out studies that ensure maintenance of cell integrity before and after immersion in liquid nitrogen (LN). Therefore, the aim in this study was to investigate the water content, cooling rate, and final temperature most suitable for cryopreservation of Coffea arabica L. seeds. The seeds were dried by silica gel to water contents of 5, 10, 15, 20, 30 and 40 % wb, subjected to slow cooling treatments at speeds of -1, -3 and -5 °C min.-1 to final temperatures of -40, -50 and -60 °C and then directly immersed in LN. After storage, seeds were rewarmed at 40 °C for two minutes. The survival rate and viability of the seeds and embryos were evaluated by the tetrazolium and germination tests.Results of the tetrazolium test indicate that embryos excised from cryopreserved seeds are less sensitive to cryopreservation than whole seeds are. The water content of 20% wb and the use of zygotic embryos led to the highest survival rate of the coffee seeds, depending on the cooling rate and the final temperature of precooling.Index terms: silica gel, water content, slow cooling, liquid nitrogen, coffee.Estudos exploratórios para a criopreservação de sementes de Coffea arabica L.RESUMO -A criopreservação é uma opção viável para a conservação de germoplasma de café. Entretanto, para que essa técnica apresente total sucesso é de fundamental importância realizar pesquisas que garantam a manutenção da integridade celular antes e após a imersão em NL. Portanto, o objetivo neste estudo foi investigar o teor de água, velocidade e temperatura final de resfriamento mais adequados para criopreservar sementes de Coffea arabica L. As sementes foram secadas em sílica gel até teores de água de 5, 10, 15, 20, 30 e 40% bu, submetidas a tratamentos de resfriamento lento nas velocidades -1, -3 e -5 ºC min -1 até às temperaturas finais de -40, -50 e -60 ºC e diretamente imersas em NL. Após armazenamento as sementes foram reaquecidas por dois minutos à 40 ºC. A sobrevivência e a viabilidade de sementes e embriões foram avaliadas por meio do teste de tetrazólio e de germinação. Por meio do teste de tetrazólio, observa-se que embriões excisados das sementes criopreservadas são menos sensíveis à criopreservação quando comparados às sementes inteiras. Esse fato se deve, principalmente, ao acúmulo de etileno nos tubos fechados e à relação etileno/citocinina nas folhas. Assim sendo, avaliaram-se o efeito de fontes de citocinina sobre o retardo da senescência foliar em brotações de Annona glabra L. e suas implicações sobre o seu desenvolvimento. Segmentos nodais foram cultivados em tubo de ensaio contendo 15 mL do meio WPM, suplementado com 30g L -1 de sacarose, 500mg L -1 de benomyl e 1g L -1 de carvão ativado. A esse meio adicionaram-se 6-benzilaminopurina, thidiazuron, cinetina e zeatina, todos na concentração de 1mg L -1 . Decorridos 45 dias após a inoculação, plantas foram submetidas à senescência em ambiente escuro, por um período de 9 dias, coletando-se folhas a cada três dias para quantificação de clorofila "a", clorofila "b", carotenóides, proteínas e açúcares solúveis totais. No final das fases de multiplicação e enraizamento, quantificaram-se a matéria seca, a área foliar e o número de folhas que sofreram abscisão nas plantas que não foram submetidas à senescência. Adotou-se o delineamento em blocos casualizados, sendo que cada período de senescência constituiu um bloco, com quatro repetições por tratamento. Os resultados mostraram que cinetina e zeatina, seguidas de thidiazuron e 6-benzilaminopurina, preservam maior teor de clorofilas "a", "b" e de carotenóides durante todo o período de senescência induzida. 6-benzilaminopurina e cinetina promoveram maior retenção da área foliar durante as fases de multiplicação e enraizamento de Annona glabra L. Annona glabra is a tropical species that has significant agronomic potential in terms of furnishing fruits for in natura consumption and for the production of phyto-pharmaceuticals. In vitro cultivation has been considered the most promising form of propagation for this species, although large scale utilization of this technique is currently limited by high rates of leaf abscission, reduced rates of explant multiplication and slow bud growth. The present work evaluated the effects of different cytokinins on mineral accumulation in shoots of A. glabra cultivated in vitro, and their effects on growth and survival of these plants. Buds of A. glabra were cultivated in Wood Plant Medium (WPM) in the presence of 6-benzilaminopurine (BAP), thidiazuron (TDZ), kinetin (KIN), and zeatin (ZEA). KIN and BAP use resulted in the greatest growth, largest accumulation of dry mass and leaf area development, as well as the greatest survival rate during in vitro cultivation of this species. All cytokinins tested stimulated large accumulations of nitrogen and boron in shoots, but diminished levels of calcium as compared to controls.Index terms: Growth regulators, Annonaceae, mineral nutrition. O cultivo in vitro tem sido preconizado como a forma mais adequada de propagação para essa espécie, embora sua utilização em larga escala ainda seja limitada pela elevada taxa de abscisão foliar, reduzida taxa de multiplicação dos explantes e crescimento lento das brotações. Objetivou-se, neste trabalho, avaliar o efeito de citocininas sobre o acúmulo de minerais nas brotações de A. glabra cultivadas in vitro e seus reflexos sobre o crescimento e sobrevivência das plantas nesse tipo de ambiente. Brotações de A. glabra foram cultivadas em meio Wood Plant Medium (WPM), na presença de 6-benzilaminopurina (BAP), thidiazuron (TDZ), cinetina (KIN) e zeatina (ZEA). A utilização de KIN e BAP induziu maior crescimento, maior acúmulo de massa seca, maior desenvolvimento da área foliar e maior taxa de sobrevivência das plantas durante o cultivo in vitro dessa espécie. O Inga vera Will subsp. affinis (DC). T.D. Penn. é uma espécie frutífera nativa do Cerrado, importante na recuperação de matas ciliares degradadas. Entretanto, apresenta sua propagação dificultada pelo fato de suas sementes serem recalcitrantes, ou seja, não tolerarem a perda de água. O objetivo deste trabalho foi estudar aspectos da germinação ex vitro e in vitro de ingazeiro. Para tanto, foram avaliados os efeitos de diferentes substratos: areia, Plantmax® e Areia+ Plantmax®; diferentes concentrações de sais: WPM, WPM/2, MS e MS/2, e diferentes concentrações de GA3 (0, 5, 10, 17 e 20 µM) no meio de cultura. Observou-se que, na germinação ex vitro, o substrato Plantmax® proporcionou maior porcentagem de germinação (82%). Com relação à germinação in vitro, a maior percentagem de germinação foi obtida utilizando-se meio de cultura WPM/2 (96%). A adição de GA3 no meio de cultura não foi estatisticamente significativa, no entanto, a concentração de 20 µM de GA3 proporcionou um aumento na germinabilidade de sementes de ingazeiro.
ABSTRACT. The aim of this study was to evaluate the efficiency of vitrification and droplet vitrification for the cryopreservation of Hancornia speciosa shoot tips. The shoot tips were subjected to four different periods of exposure (15, 30, 45 and 60 min.) to plant vitrification solution 2 (PVS2) before plunging into liquid nitrogen. We evaluated the regrowth of H. speciosa shoot tips that were cryopreserved by the classical vitrification technique and by the droplet vitrification technique. Shoot tips were submitted to different periods of pre-culture (absence, 24 or 48h) in a medium containing 0.3 M sucrose prior to cryopreservation. With a PVS2 exposure period of 60 min., significant differences were observed between the two techniques used in this study with respect to regrowth rates; however, both cryopreservation methods allowed for a shoot tip regrowth rate of over 70%. Use of a preculture step increased the regrowth rate of cryopreserved shoot tips. Shoot tip cryopreservation techniques employing vitrification and droplet vitrification proved to be viable for the long-term storage of mangaba tree samples.Keywords: native species, in vitro conservation, recalcitrant seeds, plant genetic diversity conservation, vitrification, droplet vitrification.Criopreservação de mangabeira (Hancornia speciosa Gomes): um protocolo para armazenamento a longo prazo RESUMO. O objetivo deste trabalho foi avaliar a eficiência da droplet vitrification e vitrificação na criopreservação de ápices caulinares de mangabeira. Foi avaliada a retomada de crescimento de ápices de mangabeira criopreservados conforme a técnica de droplet vitrification utilizando diferentes tempos de tratamento com a solução de vitrificação (15, 30, 45 e 60 min.) e submetidos a diferentes períodos de pré-cultivo (ausência, 24 ou 48h) em meio com 0,3 M de sacarose antes da criopreservação. Além desta técnica, foi avaliado o efeito da técnica clássica de vitrificação utilizando quatro tempos de exposição à solução de vitrificação (15, 30, 45 e 60 min.). Foram observadas diferenças significativas no tempo de exposição para a técnica de vitrificação e droplet vitrification. Para ambas as técnicas o tempo de imersão de 60 minutos promoveu uma retomada de crescimento superior a 70% nos ápices criopreservados. O pré-cultivo promoveu o aumento na sobrevivência dos explantes criopreservados. As técnicas de criopreservação de ápices caulinares por droplet vitrification e vitrificação mostraram-se viáveis para a conservação em tempo prolongado de mangabeira.Palavras-chave: espécie nativa, conservação in vitro, sementes recalcitrantes, conservação da diversidade vegetal, vitrificação, droplet vitrification.
