RESUMO:A aroeira-do-sertão (Myracrodruon urundeuva Fr. All.) é espécie arbórea que apresenta grande valor econômico devido às propriedades químicas da casca com elevado teor de taninos. Em decorrência da exploração predatória encontra-se inserida na lista oficial de espécies da flora brasileira ameaçadas de extinção na categoria vulnerável. A micropropagação vem sendo colocada como importante forma de multiplicação dessa espécie, na busca de alternativa para a exploração sustentável. Este trabalho foi realizado com o objetivo de induzir calos a partir de segmentos foliares usando 2,4-D e caracterizá-los quanto aos aspectos morfológicos e bioquímicos. Os resultados obtidos indicam que é possível induzir calos a partir de segmentos foliares de aroeirado-sertão utilizando-se 2,4-diclorofenoxiacético (2,4-D). Os calos obtidos foram compactos e não-embriogênicos, apresentando valores médios de 43,32 mg g -1 de açúcares solúveis totais; 23,62 mg g -1 de açúcares redutores; 3,157 mg g -1 de proteína total; 3,147 mg g -1 de aminoácidos livres e 0,914 mg g -1 de compostos fenólicos totais na matéria fresca. All.). "Aroeira-do-sertão" (Myracrodruon urundeuva Fr. All.) is a tree species that has great economic value due to the chemical properties of its bark, which has high tannin content. On account of the predatory exploitation it has been placed in the official list of threatened species of the Brazilian flora, in the vulnerable category. Micropropagation has been considered an important form of multiplication of this species in the search for an alternative for its sustainable exploitation. This study was conducted with the aim of inducing callus from leaf segments using 2,4-D and characterizing them according to their morphological and biochemical aspects. The obtained results indicate that it is possible to induce callus from leaf segments of M. urundeuva using 2,4-dichlorophenoxyacetic (2,4-D). The obtained calluses were compact and non-embryogenic, with average values of 43.32 mg g -1 total soluble sugars, 23.62 mg g -1 reducing sugars, 3.157 mg g -1 total protein, 3.147 mg g -1 free-form amino acids and 0.914 mg g -1 phenolic compounds in the fresh matter.
RESUMO -O trabalho teve por objetivos controlar a abscisão foliar de plântulas de angico utilizando AgNO 3 e CoCl 2 e avaliar o efeito do paclobutrazol sobre o comportamento in vitro das plântulas. As sementes foram desinfestadas e inoculadas em placas de Petri contendo papel germtest, previamente esterilizado e umedecido com água estéril. As placas ficaram no escuro por dois dias até que ocorresse a germinação das sementes e, em seguida, foram transferidas para tubo de ensaio contendo meio WPM. Todo esse material foi mantido em sala de crescimento com temperatura de 25 ± 2 ºC. No primeiro experimento, o meio de cultura foi suplementado com dois tipos de inibidores de etileno (AgNO 3 e CoCl 2 ) em diferentes concentrações (0,0; 5,0; 10,0; 20,0; e 40,0 µM). No segundo, o meio foi suplementado com diferentes concentrações (1,7; 3,4; 6,8; e 13,6 µM) de paclobutrazol. Verificou-se que houve aumento no número de folhas e redução na abscisão foliar com a utilização de 10 e 20 µM (respectivamente), independentemente do inibidor de etileno utilizado. Tanto o AgNO 3 quanto o CoCl 2 contribuíram com acréscimos no número de gemas e no número de brotações. O aumento na concentração de paclobutrazol reduziu o crescimento das plântulas, obtendo-se menores médias das seguintes variáveis: comprimento da parte aérea, número de folhas, número de folhas senescentes e matéria seca da parte aérea na maior concentração (13,6 µM) de paclobutrazol. Esse inibidor de crescimento teve efeito também no sistema radicular, reduzindo o comprimento da raiz e o número de raízes secundárias, no entanto promoveu o engrossamento das raízes.Palavras-chave: Angico, paclobutrazol e AgNO 3 e CoCl 2 . The role of cytokinins in the differentiation of the photosynthetic apparatus in micropropagated plants and their effect on the plant's ability to transition from a heterotrophic to an autotrophic condition during acclimatization was investigated. Annona glabra L. shoots were cultured on woody plant medium supplemented with sucrose and different cytokinins to evaluate leaf tissue for chloroplast development, chloroplast numbers, photosynthetic pigmentation, total photosynthetic potential, and soluble sugar content. Plants were transferred to the rooting medium in the presence or absence of sucrose and then acclimatized. Kinetin and benzyladenine (BAP) stimulated chloroplast differentiation. Inclusion of zeatin in the medium induced the formation of greater numbers of chloroplasts in the leaves, while plants cultivated in the presence of only kinetin and BAP demonstrated greater chlorophyll a and carotenoid content. The use of kinetin and BAP during in vitro culture promoted accumulation of dry matter during the acclimatization phase, especially in plants rooted under autotrophic conditions (without sucrose). Kinetin and BAP promoted development of more leaf area and greater plant survival rates in plant acclimatization on both autotrophic and heterotrophic media. The inhibitory effects of thidiazuron on the differentiation of chloroplasts, accumulation of chlorophyll a, and photosynthetic potential were examined.

Esse fato se deve, principalmente, ao acúmulo de etileno nos tubos fechados e à relação etileno/citocinina nas folhas. Assim sendo, avaliaram-se o efeito de fontes de citocinina sobre o retardo da senescência foliar em brotações de Annona glabra L. e suas implicações sobre o seu desenvolvimento. Segmentos nodais foram cultivados em tubo de ensaio contendo 15 mL do meio WPM, suplementado com 30g L -1 de sacarose, 500mg L -1 de benomyl e 1g L -1 de carvão ativado. A esse meio adicionaram-se 6-benzilaminopurina, thidiazuron, cinetina e zeatina, todos na concentração de 1mg L -1 . Decorridos 45 dias após a inoculação, plantas foram submetidas à senescência em ambiente escuro, por um período de 9 dias, coletando-se folhas a cada três dias para quantificação de clorofila "a", clorofila "b", carotenóides, proteínas e açúcares solúveis totais. No final das fases de multiplicação e enraizamento, quantificaram-se a matéria seca, a área foliar e o número de folhas que sofreram abscisão nas plantas que não foram submetidas à senescência. Adotou-se o delineamento em blocos casualizados, sendo que cada período de senescência constituiu um bloco, com quatro repetições por tratamento. Os resultados mostraram que cinetina e zeatina, seguidas de thidiazuron e 6-benzilaminopurina, preservam maior teor de clorofilas "a", "b" e de carotenóides durante todo o período de senescência induzida. 6-benzilaminopurina e cinetina promoveram maior retenção da área foliar durante as fases de multiplicação e enraizamento de Annona glabra L. CULTIVATIONABSTRACT -The micropropagation of Annonaceae has been limited by early foliar abscission in shoots, which makes the maintenance and development of plants in vitro environment difficult. This is mainly due to ethylene accumulation in closed tubes and the ethylene/cytokinin ratio in leaves. Based in these circumstances it was evaluated the effect of cytokinins sources on foliar senescence delaying in shoots of Annona glabra L. and its implications on its development. Node segments were cultivated in test tubes containing 15mL of WPM medium, supplemented with 30g L -1 of sucrose, 500mg L -1 of benomyl and 1g L -1 of activated charcoal. It was added to this medium 6-benzilaminopurine, thidiazuron, kinetin and zeatin, all of them in the concentration of 1mg L -1 .
