Syngonanthus mucugensis Giul. subsp. mucugensis is an herbaceous plant with significant economic value in the ornamental dry flower business. The restricted occurrence of the municipality Mucugê-BA, Brazil, exclusively associated with extractive exploitation, has considered this species as endangered. The objective of this work was to evaluate the organogenic potential of three different types of S. mucugensis subsp. mucugensis explants to promote the development of an alternative method to the propagation of the genetic resources of this important plant. The morphogenetic capacities of the leaf, stem and root this species was tested using Murashige and Skoog culture medium at half salt concentration and different concentrations of growth of regulators benzylaminopurine - BAP (0.00; 2.22 and 4.44 µM), and naphthalene acetic acid - NAA (0.00; 1.34 and 2.68 µM). The morphoanatomic events that lead to formation of shoots were described. Stems proved to be the best source of explants, showing 58.75% regeneration of shoot by direct organogenesis in the absence of growth regulators, and 32.18 and 47.55% of shoot regeneration by indirect organogenesis in the presence of 2.22 and 4.44 µM BAP, respectively. As for leaves, there was callus formation, but without regenerating shoots. Morphogenesis was not observed when roots were used as explants. The histological analyses showed that shoot regeneration in S. mucugensis subsp. mucugensis occurred both indirectly, by unorganized tissue differentiation, and directly through returning to merismatic activity in differentiated mature cells and preexisting bud proliferation.

A ocorrência restrita ao município de Mucugê-BA, Brasil, associado à exploração extrativista tem levado essa espécie ao risco de extinção. Neste estudo, objetivou-se avaliar o potencial organogênico de três diferentes tipos de explantes de S. mucugensis subsp. mucugensis visando ao desenvolvimento de um método alternativo para a sua propagação. A capacidade morfogênica de caule, folha e raiz foi testada utilizando o meio de cultura Murashige e Skoog com metade da concentração salina e diferentes concentrações dos reguladores de crescimento benzilaminopurina - BAP (0,00; 2,22 e 4,44 µM) e ácido naftaleno acético - ANA (0,00; 1,34 e 2,68 µM). Os eventos morfoanatômicos que levaram a formação dos brotos foram descritos. O caule demonstrou ser a melhor fonte de explante, apresentando 58,75% de regeneração de brotos via organogênese direta, em meio livre de regulador de crescimento; e 32,18 e 47,55% de regeneração de brotos por organogênese indireta na presença de 2,22 e 4,44 µM de BAP, respectivamente. Para folha, foi obtida a formação de calos sem regeneração de brotos, não sendo observada morfogênese, quando se utilizou raiz como explante. Effect of substrate and rooting on acclimatization of in vitro propagated Melocactus glaucescens Buining & BrederooMelocactus glaucescens (Cactaceae) is endemic to Bahia and is included in the IUCN and MMA lists as endangered species. Transference of in vitro cultures to ex vitro environment is critical and can become limiting for the production of micropropagated scions. The main objective of this work was to examine the effect of different substrates and rooting during acclimatization of Melocactus glaucescens. The plants were in vitro propagated and kept in 100 % light, on daily cycles for 75 days. The results suggested that the best substrate should consist of 50 % soil and 50 % sand; plantlets should be at least 5mm in diameter and height before being transferred; and that both steps in vitro rooting and harding could be eliminated from micropropagation of M. glaucescens. Further studies are still needed on desiccation time of shoots above 5 mm so that the step of in vitro rooting can be completely eliminated.

1RESUMO -O gênero Calliandra Benth. (Leguminosae) possui 134 espécies exclusivamente neotropicais, das quais 42 ocorrem na Chapada Diamantina, sendo 32 endêmicas da área. O objetivo deste trabalho foi avaliar o efeito da luz e substrato na germinação e desenvolvimento inicial de C. viscidula e C. hygrophila. As sementes foram submetidas a três tratamentos em laboratório: luz, escuro e luz/escuro (12 h/12 h). Em viveiro foram utilizados três substratos: (I) areia; (II) terra vegetal + areia; (III) vermiculita + terra vegetal + areia, mantidos a 100%; e 70% de luminosidade. As avaliações da germinação foram diárias durante 21 dias, utilizando-se os parâmetros: germinabilidade, índice de velocidade de germinação (índice de velocidade de emergência para viveiro), tempo médio de germinação e frequência relativa. Os parâmetros analisados no desenvolvimento inicial foram: comprimentos da parte aérea e raiz, número de folhas, massa seca, relação parte aérea/sistema radicular e razão do peso da folha. C. viscidula e C. hygrophila foram classificadas como fotoblásticas neutras e apresentaram melhor desenvolvimento a 70% de luminosidade nos substratos III e I e II , respectivamente. The genus Melocactus (“cabeça-de-frade”) comprises 32 species in Brazil, of which M. glaucescens and M. paucispinus are threatened with extinction. The present work evaluated the effects of different concentrations of Murashige & Skoog (MS, MS/2 and MS/4) culture medium and sucrose (15 g L-1 and 30 g L-1) on in vitro seed germination and plant growth of M. glaucescens and the efficiency of sterilization with sodium hypochlorite (NaOCl) on in vitro seed germination and plant growth of M. glaucescens when using seeds and M. glaucescens and M. paucispinus when using apical segment of cladode. In M. glaucescens, the final germination at the different MS and sucrose concentrations varied between 53.5 and 68.1% and the best results for in vitro growth were observed with the lowest mineral salt (MS/2 and MS/4) and sucrose (15 g L-1) concentrations, with lengths of the aerial portion of 9.70 and 10.76 mm, respectively. There was no difference in seed germination and plant growth in chemical and autoclave medium. It is concluded that the use of chemical sterilization with NaOCl at low concentrations of salts (MS/2 and MS/4) and sucrose (15 g L-1) are quite advantageous for producing ornamental plants germinated in vitro and/or apical segment of cladode of M. glaucescens andM. paucispinus, representing a reduction of costs for in vitro cultivation of this species.
