RESUMOA cagaiteira é uma espécie nativa do Cerrado com elevado potencial frutífero. Como suas sementes apresentam elevada variabilidade, sendo ainda recalcitrantes e dormentes, a germinação in vitro apresenta-se como uma alternativa para a propagação da espécie, além de permitir a obtenção de explantes juvenis para a micropropagação. Com presente trabalho, objetivou-se avaliar o efeito da escarificação e da luminosidade na germinação in vitro de sementes de cagaiteira. Sementes desprovidas do tegumento e sementes intactas foram inoculadas em meio MS e mantidas na ausência e presença de luz. Sementes desprovidas de tegumento germinadas tanto na presença quanto na ausência de luz apresentaram 86,25% e 88,25% de germinação aos 31 e 71 dias da inoculação, respectivamente. Após 150 dias de inoculação, plântulas provenientes de sementes desprovidas de tegumento, germinadas na presença e ausência de luminosidade, apresentaram 10% e 12% de anomalias, respectivamente. Because it seeds show high variability, it is yet considered recalcitrant and dormant, the germination in vitro appears as an alternative for specie propagation, in addition to obtain juvenile explant for micropropagation. The present research aimed to evaluate the effect of scarification and ligth on germination in vitro of cagaiteira seeds. Seeds without teguments and intact seeds were inoculated in MS medium and kept in abscence and presence of radiation. Seeds without tegument germinated in the presence or abscence of ligth showed 86.25% and 88.25% germination at 31 and 71 days of inoculation, respectively. After 150 days of inoculation, plantlets from seeds without tegument germinated in the presence and abscence of ligth, showed 10% and 12% of abnormalities, respectively.
Abstract. Numerous studies have demonstrated that the phytohormone abscisic acid (ABA) plays an important role in many processes during seed development. Abscisic acid prevents precocious germination of the developing seed and regulates the expression of specific genes during development. The availability of maize (Zea mays L.) viviparous mutants, which are defective in ABA synthesis or responsiveness to this phytohormone, allowed the investigation of varying physiological ABA levels on expression patterns of specific ABA-responsive genes. The present work indicated that the expression of several ABA-responsive genes is variable among the different viviparous mutants. Among the genes examined, Em showed the highest correlation between endogenous ABA content and transcript levels. Results from ABA studies strongly support the positive role of this phytohormone in inducing Globulin (Glbl and Glb2) gene expression in maize embryos. In the absence of ABA, no Glbl transcripts are detected within 24 h of embryo germination. On the other hand, induction of Glbl is observed in germinating embryos within 18 h in the presence of ABA. Although Glbl is positively induced by ABA, the endogenous ABA levels obtained from sibling normal and viviparous mutant embryos do not correspond to the increase in Globulin expression in these mutants. Neither Glbl, Glb2, nor Em transcripts are detected in vpl mutant embryos suggesting that a functional Vpl gene product is required for Globulin and Em gene expression. These studies indicate that Globulin gene expression in maize embryos is under the control of different factors which require at least a combination of specific endogenous ABA levels and the presence of a functional Vpl gene product.
RESUMOCattleya walkeriana é uma das mais apreciadas orquídeas do mundo, possuindo grande valor ornamental. A micropropagação é uma importante ferramenta para reprodução dessa espécie. Pelos altos custos de produção através da micropropagação convencional, relacionados à perdas durante aclimatização e alto consumo de energia em salas de crescimento, objetivou-se com este trabalho avaliar estratégias alternativas no cultivo in vitro. Foram testados dois diferentes fatores, sendo eles luz (em sala de crescimento convencional, casa de vegetação com sombreamento de 50%, e casa de vegetação sem sombreamento) e concentrações de sacarose (0, 15 e 30 g.L -1 ). O material se constitui de plântulas previamente germinadas in vitro, em meio MS e cultivadas em sala de crescimento por 90 dias. Luz natural em casa de vegetação, resultou em aumento no número de brotos, na espessura foliar e na freqüência e diâmetro dos estômatos, e ocasionou diminuição do comprimento das plântulas e dos teores de clorofila. Um menor número de raízes e brotações foram formados na concentração de 15 g.L -1 de sacarose. A ausência de sacarose resultou em menores teores de clorofila. Pela análise dos resultados, foi possível concluir que o ambiente de cultivo altera as respostas de plântulas cultivadas in vitro e pode-se recomendar o uso da luz natural e redução nas concentrações de sacarose pela metade para a micropropagação de Cattleya walkeriana, observandose a fase da propagação e os objetivos da mesma. Micropropagation plays an important role in the reproduction of this species. Based on high production costs of the standard micropropagation regarding the losses during acclimatization and high energy consumption in growth chambers, this paper aimed at evaluating alternative strategies for the in vitro cultivation. Two different factors were tested: light (standard growth chamber, greenhouse with 50% shading, and greenhouse without shading) and sucrose concentrations (0, 15, and 30g.L -1 ). The material was composed by plantlets previously germinated in vitro on MS medium and cultivated in a growth chamber for 90 days. Natural light in greenhouse induced an increase in the number of shoots, leaf thickness, stomata frequency and diameter and, on the other hand, caused decrease in the plantlet length and chlorophyll levels. Lower number of roots and shoots were formed at the concentration of 15g.L -1 sucrose. The absence of sucrose resulted in lower chlorophyll levels. Based on these results, it was possible to conclude that the cultivation environment modifies the response of in vitro cultivated plantlets and the natural light and reduction of sucrose concentrations by half can be recommend for the micropropagation of Cattleya walkeriana, taking in consideration the propagation phases and their goals.
Por apresentar dormência em suas sementes, a propagação do pequizeiro necessita de estudos para a obtenção de mudas por via assexuada. Este trabalho teve como objetivo contribuir para o estabelecimento de um protocolo de micropropagação de pequizeiro. Para indução de brotações, os explantes foram inoculados em meio WPM com diferentes concentrações de BAP e ANA. Para a indução de raízes, brotações obtidas in vitro foram transferidas para meio WPM suplementado com diferentes concentrações de AIB e na presença ou ausência de carvão ativado. As plantas obtidas foram transferidas para sacos de polietileno contendo o substrato, cobertas com uma garrafa tipo pet para auxiliar no processo de aclimatização. O melhor tratamento para a indução de brotações foi aquele que utilizou 0,05mg L-1 de ANA combinado com 0,75mg L-1 de BAP, obtendo-se uma média de 6 brotações por explante, induzindo maior número de gemas por explante (17,4), o que proporcionou a maior taxa de multiplicação (8,7). A utilização de 3mg L-1 de AIB proporcionou a indução de raízes em 100% dos explantes, gerando um número médio de 12,87 raízes por brotação. As raízes desenvolvidas no meio de cultivo contendo carvão ativado foram maiores (33,16 mm), com maior número de raízes secundárias (19,53) e com maior taxa de sobrevivência na fase de aclimatização.
RESUMOO murici-pequeno (Byrsonima intermedia A. Juss.) é um arbusto do cerrado cujo chá da casca do caule apresenta atividade adstringente nas diarréias e disenterias. O gênero Byrsonima apresenta taxa de germinação baixa e emergência lenta da plântula, dificultando a propagação sexuada. Objetivou-se com este trabalho obter calos friáveis em explantes foliares de murici-pequeno para estudos futuros em suspensão celular e metabolismo secundário, assim como embriogênese somática. Avaliou-se o efeito de diferentes concentrações da auxina 2,4-D e a sua interação com as citocininas TDZ e BAP na calogênese. Os resultados demonstraram que na ausência de 2,4-D não ocorre formação de calo em explantes foliares. A adição de BAP ou TDZ não influencia o processo de calogênese. Para a indução e proliferação de calo em explantes foliares de murici-pequeno, os resultados recomendam o uso de meio MS, acrescido de 1,0 mg L -1 de 2,4-D, mantendo os explantes em condição de escuro por 30 dias. Juss is a shrub of cerrado whose husk presents medicinal activities in diarrheas and dysenteries. The Byrsonima genus present low germination rate and slow plantlet emergency which makes difficult sexual propagation. The objective of this work was to obtain friable callus in leaf explants for future studies with suspension cells and secondary metabolites as well as somatic embryogenesis. The effect of different concentrations of 2,4-D and its interaction with TDZ and BAP in callus formation was evaluated. The results demonstrated that there is no formation of callus in leaf explants maintained in absence of 2,4-D. The addition of TDZ or BAP had no influence in the calogenesis process. For callus induction and proliferation, the results suggest the use of MS medium supplemented with 1.0 mg L -1 2,4-D maintaining the explants in the dark.
-Subcellular changes are relevant to understand plant organogenesis and embryogenesis in the early stages of cell development. The cytology during cell development in tissue culture is however still poorly characterized. This study aimed to characterize the ultrastructural differences related to callogenesis of anthers, ovaries, leaf and nodal segments of Inga vera Willd. subsp. Affinis (DC.) T.D. Penn. Flower buds, nodal segments and leaves were disinfected and inoculated in test tubes containing MS medium with 3% sucrose and 4.5µM 2.4-D, except for leaf callogenesis, where 9µM of this auxin was used, and for the callogenesis of anthers and ovaries, where the culture medium was enriched with 0.25% activated charcoal and 90µM PVP. After 45 days in culture medium, the anther, ovary, leaf and nodal segment calli were fixed in Karnovisky and prepared for visualization by scanning and transmission electron microscopy. Ultrastructural differences were observed among the callus cells of anthers, ovaries, segments and leaves. There was no evidence of somatic embryo formation in the anther, leaf and nodal segment calli, in spite of some embryogenic characteristics in the cells. The ovary calli, with indications of embryo formation, seem to be the most responsive explant source for embryogenesis. Keywords: Anthers, Ovaries e Nodal segments. RESUMOA mangabeira destaca-se por possuir um grande potencial como planta frutífera. Suas sementes apresentam recalcitrância, dificultando sua propagação, o que torna evidente a necessidade da obtenção de mudas por via assexuada. Neste contexto, a cultura de tecidos apresenta-se como uma alternativa a ser utilizada. Entre os fatores que afetam a morfogênese in vitro, as citocininas merecem destaque, pois influenciam na diferenciação de gemas e no crescimento das brotações. Assim sendo, avaliou-se, neste trabalho, a influência de três diferentes fontes de citocinina (6-benzilaminopurina -BAP, cinetina -CIN e thidiazuron -TDZ) na indução de brotações in vitro de mangabeira, bem como o efeito residual dessa classe de reguladores no enraizamento e na multiplicação em subcultivos sucessivos. Os resultados mostraram que, entre as citocininas testadas, a 6-benzilaminopurina foi a que induziu maior número de brotações (1,98), gemas (19,22) e folhas (18,86) por explante, tendo promovido a formação de brotos de maior comprimento (4,55 cm). As brotações oriundas de meio WPM basal apresentaram maior facilidade de enraizamento. A 6-benzilaminopurina foi responsável pela maior taxa de multiplicação (9,61) de brotações de mangabeira. ABSTRACTThe mangabeira stands out for having a strong potential for fruit production. Its seeds present recalcitrance, making its propagation difficult, which makes much clear the need to obtain seedlings through asexual methods. In this context, the tissue culture presents as an alternative to be used. Among the factors that affect in vitro morphogenesis, the cytokinins should be highlighted as it influences the differentiation of buds and shoot growth. In this context the influence of three different sources of cytokinins (6-benzylaminopurine -BAP, kinetin -CIN and thidiazuron -TDZ) was evaluated on the induction of in vitro mangabeira shoots, as well as the residual effect of this type of regulator on rooting and multiplication in successive subcultures. The results showed that among the tested cytokinins, the 6-benzylaminopurine was the one that induced a higher number of shoots (1.98), buds (19.22) and leaves (18.86) per explant, promoting the formation of shoots with higher length (4.55 cm). The shoots from basal WPM medium rooted easier. The 6-benzylaminopurine was responsible for the highest rate of multiplication (9.61) of mangabeira shoots. Sucessive subcultures decreased the capacity of shoot multiplication.
Expression of genes encoding the major Zea mays embryo globulins was examined in the maize precocious germination viviparous (vp) mutants. Comparison of globulin protein profiles of precociously germinating mutant embryos with those of normally germinating mature embryos revealed substantial differences with respect to the proteins encoded by the Gibl gene. Analysis of GMbl transcript levels in vp/vp embryos suggests that these mutants do not fully switch from a program of embryo maturation to one of germination. These preliminary studies indicate that the vp mutants provide an excellent system for the study of embryo maturation in maize. We also provide evidence for the positive regulation of GibI expression by the plant growth regulator abscisic acid.
