RESUMO O objetivo deste trabalho foi verificar as atividades antibacteriana e indutora de resistência de extratos de Pycnoporus sanguineus para controle do crestamento bacteriano comum, causado por Xanthomonas axonopodis pv. phaseoli, em feijoeiro. In vitro foram utilizados extratos aquosos de basidiocarpo, micélio e filtrado de cultura de P. sanguineus nas concentrações de 1, 5, 10, 15 e 20%, além das testemunhas água, acibenzolar-S-metil (ASM - 125 mg i.a. L-1) e antibiótico (22,5 mg L-1 de oxitetraciclina + 225 mg L-1 de estreptomicina). In vivo foram realizadas avaliações de severidade e atividade de peroxidase, polifenoloxidase, β-1,3 glucanase e fenilalanina amônia-liase, com o uso de extrato aquoso de micélio e de basidiocarpo e filtrado de cultura de P. sanguineus a 5% e 10%. Verificou-se atividade antibacteriana apenas para o filtrado de cultura em concentrações acima de 15% e para o extrato de basidiocarpo nas concentrações de 1 a 20%. In vivo, os resultados indicaram o potencial de extratos de basidiocarpos de P. sanguineus para o controle de X. axonopodis pv. phaseoli em feijoeiro, com redução média de 56% na severidade, o que pode ter ocorrido tanto por atividade antimicrobiana direta quanto por indução de resistência, envolvendo principalmente a ativação das enzimas de defesa vegetal peroxidase e polifenoloxidase.

Induced resistance emerges as an alternative method for controlling plant diseases. This study aimed to evaluate the efficacy of biotic and abiotic resistance inducers for controlling white rust in rocket (Eruca sativa), as well as biochemical changes (peroxidase) and fitness costs. The experiments were developed with the abiotic inducers acibenzolar-S-methyl (ASM) (12.5, 25, and 50 mg ai L-1) and citrus biomass (CB) (0.1, 0.25, and 0.5%), as well as with the biotic ones Saccharomyces cerevisiae (25 mg mL-1), Bacillus thuringiensis (25 mg p.c. mL-1), Saccharomyces boulardii (25 mg mL-1), and phosphorylated mannan oligosaccharide (PMO) (0.25%), in preventive and curative interventions. Fungicide mancozeb (1.6 g ai L-1), Bordeaux mixture (1%), and water were the control treatments. Leaf samples were collected 3, 7, 11, 15, and 19 days after the treatments to determine peroxidases and assess the severity and production. Concerning abiotic inducers, all doses of ASM and CB 0.5% (preventive) and CB 0.25% (curative) reduced the severity of white rust, whereas, among biotic inducers, only PMO applied preventively controlled the disease. Peroxidase activity was higher for CB 0.25% and ASM 50 mg L-1. Bordeaux mixture induced higher peroxidase activity.

Ion exchange chromatography (IEC) and gel filtration chromatography (GFC) were applied to separate power eliciting fractions from five hundred female root knot nematodes (Meloidogyne javanica). The purification of elicitors, through IEC, resulted in sixty glicidic fractions and six glycoprotein ones. They were classified according to their nature, being twenty-six glicidic fractions and thirty-seven glycoprotein ones, with molecular masses ranging from 29.19 to 2,989.25 kDa. From the purified fractions, eight of them presented phaseolin inducing effect potential, whereas fifteen fractions presented gliceolin inducing effect. Chromatography proved to be efficient in purifying the eliciting compounds. Compounds having gliceolin and phaseolin suppressing characteristics were verified in the bioassays. For those fractions obtained through IEC and posteriorly submitted to GFC that did not induce phytoalexin, it is suggested that the molecules need to act jointly so there is eliciting effect and thus induce a defense response in the plant.
RESUMO Este trabalho objetivou avaliar a indução de fitoalexinas e a atividade de peroxidases em sorgo e soja tratados com extratos de basidiocarpos de Pycnoporus sanguineus. Os extratos diclorometânico, hexânico e etanólico com 100, 250, 500 e 750 mg/L foram testados em relação à indução da produção de fitoalexinas e atividade de peroxidases em cotilédones de soja e mesocótilos estiolados de sorgo. Acibenzolar-S-metil (200 mg/L do produto comercial) e água destilada + Tween 20 foram utilizados como tratamentos controles positivo e negativo, respectivamente. Parafitoalexinas em mesocótilos de sorgo, o extrato hexânico (750 mg/L) proporcionou a maior indução, porém sem diferir significativamente do ASM. Para fitoalexinas em cotilédones de soja, os extratos de P. sanguineus não induziram atividade significativamente diferente dos tratamentos controles positivo e negativo, havendo uma tendência de supressão da síntese de gliceolina pelo extrato diclorometânico. Em relação às peroxidases, os extratos diclorometânico para sorgo e soja e etanólico para soja inibiram a atividade enzimática. A indução verificada para o extrato hexânico em sorgo não diferiu do controle ASM. A atividade específica de peroxidase em soja foi inibida pelo extrato etanólico e induzida pelo hexânico, mas sem diferença do tratamento com ASM. Esses resultados indicam o pequeno potencial destes extratos para a indução de resistência em patossistemas envolvendo sorgo e soja.
