Plants that use resources to defend themselves in the absence of pests or pathogens will pay off with energy costs that can reflect in the productivity. Thus, the objective of this work was to evaluate the fitness costs of the resistance induced in bean plants, before the presence of the pathogen, by using biotic (Bacillus cereus) and abiotic (acibenzolar-S-methyl, ASM) inducers, applied 2, 3 or 4 times during the cycle of the culture. RESUMOPlantas que utilizam recursos para defesa na ausência de pragas ou patógenos, arcarão com custos energéticos que podem refletir na sua produtividade. Assim, teve-se por objetivo avaliar os custos adaptativos da indução de resistência, antes da chegada do patógeno, em feijoeiro induzido por um indutor biótico (Bacillus cereus) e um abiótico (acibenzolar-S-metil, ASM), em 2, 3 ou 4 aplicações distribuídas ao longo do ciclo da cultura. Avaliou-se o efeito protetor contra a bactéria Xan thomonas axono podi s pv. ph ase oli , além d a at ivid ade de peroxidase, quitinase, β-1,3-glucanase, síntese de lignina, fenóis e crescimento com base na matéria seca. Observou-se que os indutores Kuhn, O.J. & Pascholati, S.F.. The ASM increased the activity of peroxidase, chitinase and β-1,3-glucanase, while B. cereus only increased peroxidase activity. Unlike B. cereus, the ASM increased lignin synthesis and decreased ph enolic c ompound cont ent and pla nt growt h . Th erefore, t he resistance induced by ASM represents high fitness costs for bean plants, while the resistance induced by B. cereus represents low cost and its potential use can be explored.
Este trabalho teve por objetivo avaliar, em condições de casa de vegetação e de campo, os danos causados pelo PRSV-W e ZYMV em abobrinha-de-moita (Cucurbita pepo cv. Caserta). As plantas em casa de vegetação foram inoculadas com os vírus individualmente e em mistura aos 12 e 22 dias após emergência (DAE) e aos 5, 15 e 25 DAE no campo. Em casa de vegetação, as infecções com PRSV-W + ZYMV, PRSV-W e ZYMV, na primeira época de inoculação, ocasionaram reduções de área foliar de 39,6%, 36,8% e 12,1%, respectivamente. As massas fresca e seca também foram significativamente afetadas na primeira época de inoculação. No campo, as plantas com infecções individuais ou mistas dos potyvírus produziram frutos não comerciais em quantidades que variaram de 14 a 861 g/planta, dependendo da idade que foram inoculadas. As plantas tratadas com tampão fosfato aos 5, 15 e 25 DAE produziram em média 573 g, 937 g e 1172 g de frutos comerciais e 282 g, 221 g e 192 g de frutos não comerciais, respectivamente. A redução na massa fresca das plantas foi diretamente relacionada com a época de inoculação, com médias de 60,7% para aquelas inoculadas aos 5 DAE e de 22,7% para aquelas inoculadas aos 15 DAE. Na terceira época de inoculação não houve diferença significativa de massa fresca entre os tratamentos.
This work aimed to control the root-knot nematode Meloidogyne incognita in tomato Solanum lycopersicum L plants with high-diluted solutions of Thuya occidentalis, and to study its effects on growth and plant defense responses. The in vivo experiment was carried out over two years (2013 and 2014) at a climatized greenhouse, whilst the in vitro experiment was carried out in the laboratory. Eight treatments were used [6, 12, 24, 50, 100, 200 and 400CH (Hahnemannian centesimal) of T. occidentalis, with water as control treatment]. For the in vivo assay, in 2013 plants were inoculated with about 4850 eggs and second-stage juveniles (J2), while in 2014 they were inoculated with 5050 eggs and J2. The treatments were applied once a week, as 0.1% aqueous solutions onto the plant shoots, for 50 and 40 days respectively. For the in vitro experiment, the nematodes were directly exposed to the same 0.1% treatments. The treatments did not show nematostatic or nematicide effects in the in vitro assay, and had no influence on the hatching of the eggs. For the in vivo assay in 2013, T. occidentalis 100CH decreased the number of J2 present in the roots, whilst the dynamization of 200CH stimulated root development and increased the weight of the fruits of the first cluster. In 2014, 100CH decreased numbers of J2 in the soil. Some dynamization increased the plant's defense enzyme activity, such as peroxidase (24, 50, 200 and 400CH), polyphenoloxidase (200CH), and phenylalanine ammonia-lyase (24 and 50CH). In this study, T. occidentalis 100CH showed potential for the control of M. incognita, whilst 24 and 200CH influenced the growth of plants.
RESUMOCom o objetivo de desenvolver métodos alternativos para o controle da mancha angular do feijoeiro, causada por Pseudocercospora griseola, filtrados de cultura de Pycnoporus sanguineus foram testados contra P. griseola. Foi investigado ainda o potencial desses filtrados em ativar enzimas de defesa do feijoeiro, como peroxidase, polifenoloxidase e β-1,3 glucanase, bem como alterar teores de proteína total e clorofilas. Para os experimentos in vitro e em casa de vegetação foram utilizados extratos de filtrado de cultura de P. sanguineus em concentrações variando de 1 a 20% e os controles água, acibenzolar-S-metil (ASM: 75 mg i.a. L -1 ) e fungicida (azoxystrobin: 40 mg i.a. L -1 ). No experimento in vitro, o extrato não apresentou efeito inibitório sobre o crescimento micelial, na esporulação e na germinação de conídios de P. griseola. A área abaixo da curva de progresso da mancha angular foi reduzida em 82 e 49% respectivamente, nas plantas cultivadas em casa de vegetação e no campo em relação ao controle água. A atividade das enzimas peroxidase e polifenoloxidase e os teores de proteínas e clorofilas foram maiores nas plantas tratadas com o extrato. Estes resultados indicam a eficiência do filtrado de cultura de P. sanguineus para o controle alternativo da mancha angular do feijoeiro. ABSTRACT Induction of resistance in beans against Pseudocercospora griseola by culture filtrates of Pycnoporus sanguineusWith the aim of developing an alternative method for the control of angular leaf spot of common bean, caused by Pseudocercospora griseola, aqueous extracts of Pycnoporus sanguineus culture filtrates were evaluated in vitro for antimicrobial activity against P. griseola, and possible induction of resistance in bean against P. griseola. The role of the defense enzymes peroxidase, polyphenol oxidase, and β-1,3-glucanase, and the content of proteins and chlorophyll, were also investigated. For the in vitro and greenhouse experiments the culture filtrate of P. sanguineus at the concentrations of 1 to 20% was used, and water, acibenzolar-S-methyl (ASM: 75 mg i.a. L -1 ) and fungicide (azoxystrobin: 40 mg i.a. L -1 ) served as control treatments. In the in vitro study, culture filtrate did not inhibit mycelial growth, sporulation and conidia germination of P. griseola. The total area under the angular leaf spot progress curve was reduced by 82 and 49% respectively, for greenhouse and field experiments in relation to the water control. The activities of peroxidase and polyphenol oxidase, and the content of proteins and chlorophylls, increased in plants treated with the extract. The results from this study indicate the potential of P. sanguineus culture filtrate as an alternative control for angular leaf spot on beans.
O objetivo deste trabalho foi avaliar porta-enxertos de videira na fase de formação em campo, bem como o desenvolvimento inicial do enxerto da variedade 'BRS Violeta'. Mudas oriundas de estacas enraizadas de 17 porta-enxertos de videira ('SO4', 'Teléki 8B', 'Harmony', 'Golia', 'Riparia de Traviú', 'Paulsen', '420 A', '99R', '5C', 'RR101-14', 'Kober 5BB', 'Rupestris Du Lot', 'IAC 313 Tropical', 'IAC 766 Campinas', 'IAC 572 Jales', 'VR 043-43' e 'IAC 571-6 Jundiaí') foram levadas ao campo no fim da primavera, em condições subtropicais. Após quatro meses, avaliou-se a severidade da antracnose e ferrugem da videira nas folhas das brotações dos porta-enxertos e após cinco meses o vigor dos mesmos. No inverno seguinte, foi realizada a enxertia por garfagem da variedade 'BRS Violeta' e após 60 dias, avaliaram-se a porcentagem de brotação, o diâmetro e o comprimento médio do enxerto. Nos porta-enxertos 'IAC 572 Jales' e 'IAC 313 Tropical' apresentaram menor infecção foliar a antracnose e ferrugem. O porta-enxerto 'IAC 572 Jales' proporcionou maior desempenho em campo, previamente à operação de enxertia, enquanto os porta-enxertos 'SO4', 'Harmony', 'Paulsen' e 'IAC 766 Campinas' promoveram maior vigor ao desenvolvimento do enxerto 'BRS Violeta'.
