Background: Alpha-amylases are enzymes capable of degrading polysaccharides, such as starch and glycogen. Found in various organisms, such as fungi and bacteria, these enzymes have great biotechnological potential due to their insertion in several industrial sectors raging from food to biofuels. Objective: The aim of this study was to analyze patents deposited in intellectual property databases on alpha-amylases in the fields of food, beverages, detergents, animal feeds, biofuels, pharmaceuticals and textiles, with the search period being the 5 years. Method: This study targeted the use of alpha-amylases in various industrial sectors, so searches were carried out on the intellectual property database Espacenet website (European Patent Office - EPO) which contains more than 90 million patents deposited in its database. Results: During the search for patents filed in the last 5 years, 186 were found related to the use of alpha-amylases. These were disturbed as follows: 84 (biofuel), 41 (drinks), 16 (pharmaceuticals), 15 (detergents), 11 (food), 10 (animal feed), 9 (textiles). From the total number of patents found, we selected 6 from each area, except pharmaceutical products, to discuss and provide information on the application of this enzyme. Conclusion: This study demonstrated that the sectors of beverages and animal feed have preferences for thermostable alpha-amylases while sectors such as food, biofuels and textiles only regarded the importance of enzymatic efficiency. The detergent sector presented the greatest use of alpha-amylases which had distinct biochemical characteristics as solvent resistance and thermostability. The pharmaceutical sector was the one that presented less patents related to the application of alpha amylases. In addition, this work showed that China is the country with the highest patent registration for the use of alpha-amylases in the analyzed period.
Tecoma stans (L.), popularly known as ipê-mirim, is used in traditional medicine for the treatment of diabetes and digestive problems. The components of the hexane (HE) and methanol (ME) extracts obtained from the pericarp of T. stans were identified by gas chromatography-mass spectrometry (GC-MS) in their methyl ester forms (FAME).The antioxidant and cytotoxic activities of extracts, fatty acids, and methyl esters were evaluated. Methyl linolenate, methyl linoleate, and methyl palmitate were the major compounds in the HE, while methyl hexacosanoate was the main component in the ME. The samples exhibited significant antioxidant potential by DPPH assay. In the A. salina larvae bioassay, FAME (HE) and FAME (ME) were considered toxic. This study showed, for the first time to our knowledge, the chemical composition of the hexane and methanol extracts from T. stans pericarp as well as the antioxidant and cytotoxic activities of the extracts, fatty acids, and methyl esters.
Handroanthus impetiginosus, popularly known as "ipê-roxo", is used in folk medicine to treat skin inflammations, infections, stomach diseases, and cancer. Fatty acid methyl esters (FAMEs) obtained from the esterification reaction of fatty acids (FA) found in the hexane extract (HE) of seeds of H. impetiginosus were identified by gas chromatography-mass spectrometry (GC-MS). The antioxidant and cytotoxic activities of the HE and FAMEs were evaluated. Methyl palmitate, methyl linoleate, methyl oleate, and methyl stearate were the major FAMEs obtained from the HE. The samples, especially the HE, exhibited a significant antioxidant potential analyzed by ferric reducing ability power (FRAP) assay. In the A. salina larvae bioassay, the HE showed no cytotoxic effects, but the FAMEs exhibited a high toxicity. This study reported, for the first time, the antioxidant and cytotoxic activities of the HE and FAMEs obtained from H. impetiginosus seeds.
Introdução: Handroanthus impetiginosus é conhecida popularmente como ipê roxo, sendo amplamente distribuída em florestas da América Central e Sul. Possui grande importância econômica, devido ao uso de sua madeira em construções, importância ecológica, na recuperação de ecossistemas florestais, e também importância medicinal, por seus compostos bioativos. Diversas plantas são capazes de produzir substâncias que exercem papel fundamental na manutenção da saúde humana e muitos medicamentos disponíveis atualmente são originados a partir de substâncias extraídas de em extratos vegetais. Objetivos: Caracterizar ácidos graxos e ésteres etílicos obtidos a partir do extrato hexânico de sementes de Handroanthus impetiginosus por meio de cromatografia gasosa acoplada à espectrometria de massas. Metodologia: O extrato hexânico foi preparado a partir de sementes de Handroanthus impetiginosus, em aparelho de Sohxlet. O extrato hexânico foi analisado por cromatografia gasosa acoplada à espectrometria de massas (CG/EM). A identificação dos picos foi baseada nos espectros de massas, comparados ao banco de dados NIST 17 e os índices de retenção foram calculados a partir de dados de alcanos de C9 a C35, comparados com dados de referência na literatura. Resultados: O extrato hexânico apresentou um rendimento de 20,47%. A análise por CG/EM permitiu identificar uma mistura de ácidos graxos saturados e insaturados e ésteres etílicos, com predominência ácido oleico e oleato de etila, respectivamente. Conclusão: Os resultados demonstraram o potencial para a obtenção de ácidos graxos a partir de sementes de Handroanthus impetiginosus. Os óleos ou lipídeos apresentam importante valor nutricional para os seres humanos e os ácidos graxos constituem grande parte de sua composição.
Introdução: Handroanthus impetiginosus ou ipê-roxo é uma espécie conhecida por sua beleza exuberante e também pelo seu uso medicinal. Esta espécie é utilizada a séculos pelos povos indígenas como remédio para inflamações, câncer, sífilis, malária, febres, ulceras, tripanossomíase, infecções fúngicas e bacterianas. As atividades biológicas relatadas para H. impetiginosus são atribuídas aos metabólitos secundários encontrados principalmente nas cascas, como naftoquinonas, cumarinas, flavonoides, taninos e esteroides. Bioensaios são muito utilizados para investigar o potencial tóxico de compostos de origem natural. Os testes de toxicidade são fundamentais para avaliar e prevenir os possíveis efeitos tóxicos de substâncias naturais advindos das plantas. Objetivos: Avaliar as atividades antioxidante e larvicida frente Artemia salina e Culex quinquefasciatus do extrato hexânico de sementes de Handroanthus impetiginosus. Metodologia: O extrato hexânico foi preparado a partir de sementes de Handroanthus impetiginosus, em aparelho de Sohxlet. A atividade antioxidante foi determinada pelo método de radicais 2,2-difenil-1-picrilhidrazil (DPPH) e as amostras foram testadas nas concentrações de 1, 10, 100, 250 e 500 µg/mL. O BHT (2,6-di-tert-butil-4- metilfenol) foi utilizado como composto de referência. Para avaliação de letalidade frente à Artemia salina e Culex quinquefasciatus foram avaliadas amostras dos extratos nas concentrações de 125, 250, 500 e 1000 µg/mL, sendo consideradas ativas quando o valor da DL50 foi menor que 1000 µg/mL. Resultados: As amostras do extrato hexãnico apresentaram atividade antioxidante inferior a 50%, em comparação com o padrão BHT, o qual apresentou inibição superior a 90% nas concentrações de 250 e 500 µg/mL e de 86% na concentração de 100 µg/mL (P < 0,0001). Nas concentrações de 1 a 10 µg/mL houve uma ligeira redução na atividade antioxidante das amostras, entretanto, estas atividades foram superiores àquela observada para o padrão BHT. Não se observou efeito tóxico do extrato hexânico em Artemia salina. Todas as amostras testadas provocaram uma elevada mortalidade de larvas C. quinquefasciatus, independente da concentração empregada (P < 0,0001), mostrando uma seletividade para esse organismo. Conclusão: Os dados encontrados até o momento encorajam a realização de novos testes para a confirmação dos resultados e também para a detecção de outras atividades biológicas. Tecoma stans (L.) Juss. Ex Kunth is a species belonging to the Bignoniaceae family, popularly known as yellowing, yellow bell, and garden little ipe. Studies with T. stans seeds are scarce and it have potential in the search for natural compounds with biological activities. This study aimed to evaluate the toxicity and antioxidant activity of the ether extract (EE) and fractions from T. stans seeds. The EE was obtained in Soxhlet apparatus with petroleum ether and fractions were obtained by hydrolysis and esterification reactions. Phytochemical screening evaluated the presence of steroids, triterpenoids, alkaloids, coumarins and anthraquinones in EE. The toxicity was evaluated by the A. salina lethality test and antioxidant activity by DPPH method. It was observed the presence of steroids, triterpenoids and alkaloids in EE, which had no toxicity to A. salina (LC50 > 1000 µg/mL). The fractions of seeds exhibited toxicity on A. salina. All samples showed antioxidant activity with EC50 between 0.9 and 1.1 µg/mL. These results indicated potential applications for T. stans seeds.
