Most animals adapt readily to increased supplies of carbohydrate in the intestinal lumen by increasing enzymes for degradation and increasing glucose transporter activity. However, the extent of upregulation of Na+-dependent glucose cotransporter 1 (SGLT1) activity and content in response to increased delivery of carbohydrate to the small intestinal lumen of ruminants is unclear. Therefore, an experiment was conducted to determine the effect of glucose and starch hydrolysate on the activity and abundance of SGLT1 in the small intestine of steers. In a randomized complete block design, 40 crossbred beef steers (243+/-2 kg BW) were fed 0.163 Mcal of ME/(kg BW0.75(d; W), 0.215 Mcal of ME/(kg BW0.75 x d; 2M), or 0.163 Mcal ME/(kg BW0.75 x d) and infused for 35 d into the rumen (R) or abomasum (A) with 12.6 g/(kg BW0.75 x d) of starch hydrolysate (S) or into the abomasum with 14.4 g/(kg BW0.75 x d) of glucose (G). Steers were slaughtered, and brush-border membrane vesicles were prepared from the small intestinal samples obtained from five equidistant sites along the intestine. Maltase activity in vesicles and homogenates differed with intestinal sampling site (quadratic, P < 0.001). Steers on the AG treatment yielded a greater intestinal maltase activity (38 nmol glucose x mg protein(-1) x min(-1)) compared with the AS, RS, W, or 2M treatments (34, 26, 23, and 23 nmol glucose x mg protein(-1) x min(-1) respectively [SEM = 3; P = 0.02]). Sodium-dependent glucose uptake averaged 18.4+/-3.94 pmol glucose/(mg protein x s) and was not affected by treatment, but uptake decreased distally along the intestine (P < 0.001). There was no effect of treatment on SGLT1 protein abundance, but SGLT1 protein abundance increased linearly from the duodenum to the ileum (P = 0.05). The inverse relationship between glucose uptake and SGLT1 abundance suggests that the regulation of brush border Na+-dependent glucose transport capacity is complex, involving factors other than the presence of luminal carbohydrate.
O objetivo neste trabalho foi avaliar o efeito da inclusão de fontes de óleo na dieta de cabras em lactação sobre a produção, a composição e o perfil dos ácidos graxos da gordura do leite. Foram utilizadas quatro cabras da raça Saanen (54±1,02 kg PV) aos 80 dias em lactação, canuladas no rúmen. Os tratamentos consistiram de dietas suplementadas com diferentes fontes de óleo (canola, arroz e soja) e de uma dieta controle. O delineamento experimental utilizado foi o quadrado latino 4 x 4 e os dados foram analisados utilizando-se o procedimento MIXED do SAS (2001). As diferenças entre as médias dos tratamentos foram determinadas por contrastes ortogonais, a 10% de significância. A produção e composição do leite, à exceção do percentual de sólidos totais, não foram influenciadas pela suplementação lipídica na dieta. A utilização de óleos vegetais aumentou as concentrações de ácido linoléico conjugado (CLA), 18:0, 18:1n-9, 18:2, na gordura do leite. Constatou-se diminuição das concentrações de ácidos graxos saturados (AGS) e de cadeias curta (AGCC) e média (AGCM). Houve aumento das concentrações de ácidos graxos mono (AGMS) e poliinsaturados (AGPI), de ácidos graxos de cadeia longa, de ômega-3 e 6 e da relação AGPI/AGS. A inclusão do óleo de soja na dieta, em comparação à suplementação com óleo de canola, resultou em maior concentração de AGPI e n-6, em maior relação de AGPI/AGS e n-6/n-3 e menor de 22:6n-4 no leite. O enriquecimento de dietas para cabras com óleos vegetais melhorou a qualidade do leite elevando a concentração de ácidos graxos desejáveis.
Twenty-four Holstein steer calves (88 ± 3kg) with abomasal catheters were randomly assigned within blocks to one of four treatments. The treatments consisted of four abomasal infusions: water (control), 4 g/kg BW.d-1 of partially hydrolyzed starch (SH), 0.6 g/kg BW.d-1 of casein, and a mix of SH + casein. The small intestine was collected and five equidistan, 1m sites were identified (duodenum, jejunum 1, jejunum 2 jejunum 3 and ileum). Maltase specific activity in mucosal homogenate and brush border membrane vesicles, SGLT1 protein abundance, and sodium-dependent glucose uptake in brush border membrane vesicles did not differ between the calves receiving different abomasal infusion treatments. However, maltase specific activity in homogenates and brush border membrane vesicles increased four-fold from the duodenum to the first jejunal site before declining steadily towards the ileum (p=0.0145 p=0.0020, respectively). The SGLT1 abundance differed by intestinal sampling site (p=0.0162). These data indicated that cattle might not have the ability to alter the capacity for glucose uptake when challenged with different substrates and that the regulation of SGLT1 expression could differ between bovine and other species
RESUMO -O objetivo deste trabalho foi avaliar o efeito da substituição do milho pela farinha de mandioca de varredura, em dietas de cabras Saanen em lactação, sobre o desempenho, a composição do leite (sólidos totais e proteína bruta), a digestibilidade da matéria seca (MS), matéria orgânica (MO), proteína bruta (PB), extrato etéreo (EE), fibra insolúvel em detergente neutro (FDN), e amido e a concentração dos nutrientes digestíveis totais (NDT) das dietas. Foram utilizadas quatro cabras há 100 dias em lactação. O delineamento utilizado foi o quadrado latino 4 x 4, em que os tratamentos consistiram em níveis de 0, 33, 67 e 100% de substituição do milho pela farinha de varredura. Os tratamentos não influenciaram a ingestão, excreção fecal, digestão total e a digestibilidade total da MS, MO, PB, FDN e carboidratos não-fibrosos (CNF). Houve diminuição linear na excreção fecal e aumento na digestibilidade do amido, com a inclusão da farinha de mandioca de varredura. O NDT não diferiu entre as dietas estudadas, com média de 71,59%. Houve correlação positiva entre a digestibilidade da matéria orgânica (DIGMO) e o NDT (r = 0,9472), permitindo estimativas do NDT a partir da equação NDT (%) = 0,8897DIGMO (%) + 10,9940 (R 2 =0,8972). A substituição da farinha da mandioca de varredura não alterou a produção, bem como a composição do leite. Recomenda-se a utilização da farinha de mandioca de varredura em dietas de cabras em lactação, em total substituição ao milho, sem prejuízos na digestibilidade dos nutrientes e na produção.Palavras-chave: cabras leiteiras, mandioca, NDT, produção de leite, subproduto Corn Replacement by Cassava by-Product Meal in the Lactating Goat Diets: Effects on Milk Production and Composition and Nutrients DigestibilityABSTRACT -The objectives of this work were to evaluate effects of replacing corn by cassava by-product meal, in diets of Saanen lactating goats, on milk production and composition (total solids and crude protein), nutrient apparent digestibility and total digestible nutrients (TDN) of diets. Four multiparous goats fitted with ruminal cannula, with 100 days of lactation, were used. The design was a 4 x 4 Latin square and treatments as following: 0, 33, 67 and 100% replacement of corn by cassava by-product meal. Treatments did not affect intake, fecal flow and dry matter (DM), organic matter (OM), crude protein (CP), neutral fiber detergent (NDF) and non-fiber carbohydrate (NFC) total digestion (AD) and total digestibility (DIG). There was a linear fecal flow decrease and a starch digestibility increase with cassava by-product meal inclusion. TDN did not differ among diets with an average of 71.59%. There was a positive correlation (r = 0.9472) between OMDIG and TDN, making possible TDN estimation from the equation: TDN (%) = 0.8897OMDIG (%) + 10.9940 (R 2 = 0.8972). The silage quality of Brachiaria brizantha cultivars ensiled with different levels of millet meal was evaluated. The experimental design was a completely randomized with three replications in a factorial 3 x 4, with three cultivars of Brachiaria brizantha: marandu, xaraés, and piatã and four levels of millet meal 0, 5, 10, and 15%. The addition of millet meal improved the quality of B. brizantha silage. The inclusion of the additive at 15% provided the best nutritional values. The piatã silage had the lowest contents of neutral detergent fiber, acid detergent fiber, and lignin and the highest content of total digestible nutrients, being a better quality silage as compared to those of xaraés and marandu grasses. RESUMO -O experimento foi realizado no município de Luiziana, noroeste do Estado do Paraná, no período de junho a outubro de 2000. Foram utilizados 24 novilhos inteiros, com nove meses de idade e peso inicial médio de 223 kg, pertencentes a dois grupos genéticos distintos, sendo 15 ½ Red Angus ½ Nelore e nove ½ Marchigiana ½ Nelore. Avaliou-se o desempenho dos animais submetidos a três formas de suplementação e a qualidade e produtividade da forragem. A área experimental foi constituída por quatro piquetes, sendo três de Coastcross (Cynodon dactylon (L.) Pers), com área total de 7,35 ha e um de Aveia (Avena strigosa Schreb), consorciada com Azevém (Lolium multiflorum Lam), totalizando 2 ha. Os novilhos permaneceram em pastagens de Coastcross e foram submetidos a três tratamentos: pastejo em Aveia + Azevém (AA) por quatro horas diárias; suplemento (S) a 1,2% do peso vivo; suplemento + lasalocida (SL) a 1,2% do peso vivo. Estimativas da biomassa e da qualidade das forrageiras foram realizadas. Decorridos 112 dias experimentais, S e SL não apresentaram diferença para ganho médio diário (GMD = 1,063 kg e 1,026 kg, respectivamente), mas foram superiores a AA (0,856 kg). Concluiu-se que a adição de lasalocida ao suplemento não produziu diferença no desempenho dos animais. Estes resultados comprovam que animais mantidos em pastagem de Coastcross, com quatro horas de pastejo em pastagem anual de inverno ou com suplementação (1,2% do peso vivo), independentemente da adição de ionóforo, apresentaram elevados ganhos na época crítica do ano.Palavras-chave: aveia, azevém, desempenho, lasalocida, novilhos, pastejo Cattle Supplementation on Coastcross Pasture (Cynodon dactylon (L.) Pers) and three types of supplementation: grazing Oat + Ryegrass pasture (OR) for four hours/day; 1.2% of body weight (BW) supplement (S); 1.2% of BW supplement + lasalocid (SL). The experimental area consisted of four padocks: three of Coastcross, with 7.35 ha of total area; and one of Oat (Avena strigosa Schreb) and Ryegrass (Lolium multiflorum Lam), with two ha. Pasture accumulation and quality were evaluated. After 112 days of experimental period, average daily gain did not show any difference between S and SL (1.063 kg and 1.026 kg, respectively), but both were superior than OR (0.856 kg). Lasalocid addition to supplement did not improve animal performance. Avaliaram-se os coeficientes de digestibilidade (CD) parcial e total dos nutrientes, o pH, a concentração de nitrogênio amoniacal (N-NH3) do líquido ruminal e a eficiência de síntese microbiana em bovinos alimentados com rações contendo 0; 8; 16 e 24% de resíduo de cervejaria fermentado (RCF). O resíduo de cervejaria foi conservado em silo de superfície (5.000 kg). Foram utilizados quatro bovinos da raça Holandesa com peso corporal de 480 ± 100 kg, portadores de cânula ruminal e duodenal, distribuídos em delineamento quadrado latino 4 × 4. O consumo médio de MS pelos animais foi de 1,7% do peso corporal (PC). Os coeficientes de digestibilidades total e ruminal dos nutrientes não diferiram entre os níveis de RCF utilizados nas rações. Os níveis de RCF nas rações tiveram efeito linear crescente sobre o coeficiente de digestibilidade intestinal de matéria seca e carboidratos totais, calculado em porcentagem do que chegou ao compartimento. Não houve efeito dos níveis de RCF nas rações sobre o pH e a concentração de N-NH3 do líquido ruminal. Os níveis de RCF nas rações não influenciaram a eficiência de síntese microbiana em g N-Mic/kg de matéria orgânica aparentemente degradável no rúmen (MOADR) e da matéria orgânica verdadeiramente degradável no rúmen (MOVDR). A inclusão de até 24% do RCF na MS em rações para bovinos não altera os processos de fermentação ruminal e digestão dos nutrientes.
RESUMO -Os objetivos deste trabalho foram avaliar os efeitos de dois níveis de volumosos nas rações e dois períodos experimentais sobre a eficiência de síntese microbiana, a concentração ruminal de amônia, o pH ruminal, o fluxo duodenal e a digestibilidade de nitrogênio (N) em bovinos. Foram utilizados quatro novilhos, Holandês Preto e Branco, com dois anos de idade e 340 kg de peso vivo, implantados com cânulas ruminal e duodenal. O delineamento experimental foi um quadrado latino 4 x 4, em que os animais receberam quatro tratamentos, que consistiram de dois níveis de volumoso (70 e 30%) e dois períodos experimentais (14 e 21 dias). Houve efeito significativo do nível de volumoso para pH e concentração de amônia no fluido ruminal. Houve efeito do período experimental sobre a ingestão de N. A eficiência de sintese microbiana aparente variou de 29,3 a 32,0 g N microbiano/kg de MODR. O pH mínimo obtido foi de 6,5 para 70% de volumoso e 6,2 para 30% de volumoso. Four Holstein steers, two years old and 340 kg of body weight, ruminally and duodenally cannulated, were used. The experimental design was a 4 x 4 latin square and animals received four treatments as folowing: two levels of forage (70 and 30%) and two experimental periods (14 and 21 days). There were effects of the forage level on the ruminal ammonia concentrations and pH. There was effect of experimental period on the intake of N. The apparent microbial efficiency synthesis ranged from 29.3 to 32.0 g microbial N/ kg OMDR. The minimum pH for high forage diet (70%) was 6.5 and for low forage diet (30%) was 6.2. The maximum ruminal ammonia concentration of 16.9 mg/100 mL was obtained for the level of 70% of forage and 18.9 mg/100 mL of ruminal fluid to the level of 30% of forage.
