Sulfate is present in foods, beverages, and drinking water. Its reduction and concentration in the gut depend on the intestinal microbiome activity, especially sulfate-reducing bacteria (SRB), which can be involved in inflammatory bowel disease (IBD). Assimilatory sulfate reduction (ASR) is present in all living organisms. In this process, sulfate is reduced to hydrogen sulfide and then included in cysteine and methionine biosynthesis. In contrast to assimilatory sulfate reduction, the dissimilatory process is typical for SRB. A terminal product of this metabolism pathway is hydrogen sulfide, which can be involved in gut inflammation and also causes problems in industries (due to corrosion effects). The aim of the review was to compare assimilatory and dissimilatory sulfate reduction (DSR). These processes occur in some species of intestinal bacteria (e.g., Escherichia and Desulfovibrio genera). The main attention was focused on the description of genes and their location in selected strains. Their coding expression of the enzymes is associated with anabolic processes in various intestinal bacteria. These analyzed recent advances can be important factors for proposing possibilities of metabolic pathway extension from hydrogen sulfide to cysteine in intestinal SRB. The switch from the DSR metabolic pathway to the ASR metabolic pathway is important since toxic sulfide is not produced as a final product.
The small–large intestine axis in hydrogen sulfide accumulation and testing of sulfate and lactate in the gut–gut axis of the intestinal environment has not been well described. Sulfate reducing bacteria (SRB) of the Desulfovibrio genus reduce sulfate to hydrogen sulfide and can be involved in ulcerative colitis development. The background of the research was to find correlations between hydrogen sulfide production under the effect of an electron acceptor (sulfate) and donor (lactate) at different concentrations and Desulfovibrio piger Vib-7 growth, as well as their dissimilatory sulfate reduction in the intestinal small–large intestinal environment. Methods: Microbiological, biochemical, and biophysical methods, and statistical processing of the results (principal component and cross-correlation analyses) were used. Results: D. piger Vib-7 showed increased intensity of bacterial growth and hydrogen sulfide production under the following concentrations of sulfate and lactate: 17.4 mM and 35.6 mM, respectively. The study showed in what kind of intestinal environment D. piger Vib-7 grows at the highest level and produces the highest amount of hydrogen sulfide. Conclusions: The optimum intestinal environment of D. piger Vib-7 can serve as a good indicator of the occurrence of inflammatory bowel diseases; meaning that these findings can be broadly used in medicine practice dealing with the monitoring and diagnosis of intestinal ailments.
Виразковий коліт (ВК) -це одна з найпоширеніших основних форм ідіопатич-ної запальної хвороби кишечника [8,80,90,100]. ВК вражає товсту (переважно пряму) кишку, буває гострим або хронічним, може спричинити тяжкі ускладнення і бути невиліковним. Сучасні методи терапії зводяться до застосування протиза-пальних препаратів, антибіотиків і стероїдів [20,52,54]. У разі їх неефективності здійснюють часткове або повне видалення кишки хірургічним способом [8].У кишечнику хворих на ВК людей виявлено збільшену кількість СВБ, порівняно зі здоровими [31,38,78,79]. G. R. Gibson та співавтори встановили, що 92% усіх СВБ, виділених від хворих на ВК людей, належать до роду Desulfovibrio, зокрема Desulfovibrio desulfuricans. Вони стійкіші до умов середовища, порівняно зі штамами, виділеними з навколишнього середовища [38]. Це спричинило зацікавленість цими бактеріями під час дослідження ВК. Встановлено, що ці мікроорганізми є причиною кривавого проносу, втрати апетиту і ваги у тварин [20,34]. Під час гістологічних до-сліджень тканин кишечника хворих на ВК виявлено епітеліальну гіперплазію, абсце-си, порушення функції келихоподібних клітин і запальні інфільтрати [8].M. C. Pitcher зі співавторами встановили, що кількість життєздатних клітин СВБ є більшою у фекаліях хворих з активною стадією хвороби, порівняно з при-хованою [78]. Фізіолого-біохімічні характеристики цих бактерій, виділених від хворих із різними стадіями колітів, були різні. Приблизно 30% СВБ, виділених із фекалій хворих колітом людей, характеризувалися збільшеною швидкістю росту й активнішою сульфатредукцією, порівняно з СВБ, що виділені від здорових лю-дей [40]. T. H. J. Florin та співавтори виявили, що концентрація гідроген сульфіду у фекаліях хворих на коліти була значно вищою, ніж у здорових людей [30]. Най-більш інтенсивне відновлення сульфату встановлено для СВБ, виділених від хворих із гострою стадією хвороби [8].Інгібування росту кишкових СВБ спричиняє зниження концентрації гідроген сульфіду у просвіті кишечника людини [8,20,92]. Крім того, H 2 S із просвіту кишеч-ника ефективно нейтралізують за допомогою S-метилювання за участю тіолме-тилтрансферази [76]. Активність цього ферменту у мембранах еритроцитів людей із ВК є більшою, порівняно зі здоровими [78]. Ймовірно, збільшення концентрації ендогенного гідроген сульфіду зумовлює гомеостатичний захист.Незважаючи на численні дослідження неспецифічного виразкового коліту, остаточна роль СВБ у розвитку цього захворювання малодосліджена [8,20,79,92]. Тому необхідно детально вивчати кишкові СВБ, їх метаболізм і закономірності утворення ними гідроген сульфіду в кишечнику, з'ясовувати всі можливі механізми його токсичної дії на епітеліальні клітини. Це дасть змогу розробляти нові підходи до лікування ВК та багатьох інших захворювань кишечника.Метою роботи було проаналізувати результати сучасних досліджень і узагаль-нити нові наукові дані про роль сульфатвідновлювальних бактерій у розвитку за-хворювань, а також розглянути можливі механізми дії гідроген сульфіду на клітини кишечника людини.
The study of the intestinal sulfate-reducing bacteria, the process of dissimilatory sulfate reduction and accumulation of hydrogen sulfide, as well as their role in the inflammatory bowel diseases, including ulcerative colitis, in animals and human have increasingly attracted the attention of scientists. New opportunities for studying inflammatory bowel disease and the assessment of the effectiveness of its treatment is an urgent problem of modern biology and medicine. In this review, brief characteristics of these bacteria and their mechanism of dissimilatory sulfate reduction were described based on modern literature data and own research. The characteristics of substrates for intestinal sulfate-reducing bacteria and the thermodynamic properties of their electron donors were also described. Special attention was paid to the mechanism and stages of sulfate dissimilation including role of enzymes involved in this process. Based on our results, general scheme of dissimilatory sulfate reduction sho wing the activity of each enzyme of the process was demonstrated. The described physiological and biochemical parameters are important for a more detailed understanding of sulfate dissimilation in the human and animal bowel, as well as studying the mechanisms of action of the antimicrobial prophylactics and the therapy against specific components involved in the pathogenesis of the disease. It is also essential for understanding the mechanisms of bowel diseases and for evaluating the effectiveness of its therapy.
