Background: It is known that glycerol in an oil-in-water emulsion has a protective effect against irritating substances. Objective: To answer the question: is the protection effect of glycerol based on a regenerative process? Methods: Upon irritation by either tape stripping or acetone treatment, we applied glycerol to the skin surface under an occlusive dressing to create transepidermal water movement. As a control we used water under the occlusive dressing on the contralateral forearm. After 5 h we compared the barrier function using biological tests. Results: A significant improvement of the protective barrier function was observed in the glycerol-treated areas, as shown by the alkali resistance and by the irritant effect of dimethyl sulfoxide (DMSO) as well as sodium lauryl sulfate. Surprisingly, at the same time penetration of hexyl nicotinate improved on the glycerol-treated areas. A direct physicochemical protection effect on the surface of the skin was ruled out in additional studies using NaOH and DMSO. Conclusions: Under the given conditions glycerol leads to a more rapid reconstitution of the protective skin barrier and initiates a regenerative skin protection. In contrast to that, it is acting as a penetration enhancer.
This paper aimed to describe milk production in Southern Rio Grande do Sul state, Brazil, and to identify factors that affect milk quality at this region. The average age of regional dairy farmers is 49, and 67.9% has not concluded elementary school. Dairy farming is carried out on properties with an average of 26.06 hectares and 8.4 lactating cows. Most of them (32.83%) yield 50 to 100 L/day. Among the properties, 13.21% yield up to 30 L/day, and only 1.89% produces over 500 milk liters a day. Average yield was 6.8 L/day. Regarding to milking procedure, 39.3% farmers milk manually the animals, only 14.2% performed pre-milking teat disinfection, and 53.9% uses a single cloth to dry all animal teats. For infrastructure, 52.8% milks animals in wooden cowsheds. We observed that the average somatic cell count (SCC) was within legal parameters, presenting negative correlation with milk production (r = -0.23) and lactose content (r = -0.39). However, the total bacterial count (TBC) was above legal parameters. Education level seems to interfere in management and milk quality, because the less educated groups are, the less adequate are infrastructure, management, and product quality. Data show that there is a lower quality milk production where poor management techniques are adopted and owner education level affects milk quality. Key words: Milk quality, small farmer, milking, education ResumoEste trabalho teve como objetivo descrever a produção de leite no Sul do Rio Grande do Sul, Brasil, e identificar os fatores que afetam a qualidade do leite na região. A idade média dos produtores de leite da região é de 49 anos, e 67,9% não concluíram o ensino fundamental. A atividade leiteira é realizada em propriedades com média de 26,06 hectares e 8,4 vacas em lactação. A maioria dos produtores (32,83%) produz entre 50 a 100 litros de leite por dia. Entre as propriedades, 13,21% produz até 30 L/dia, e apenas 1.89% produz mais de 500 litros de leite por dia. A produtividade média foi de 6,8 L/vaca/dia. No que diz respeito ao procedimento de ordenha, 39,3% dos produtores de leite ordenha seus animais manualmente, apenas 14,2% realizam desinfecção dos tetos antes da ordenha, e 53,9% usa um único pano para secar todos os tetos dos animais. Com relação à infra-estrutura, 52,8% dos produtores ordenham seus animais em estábulos de madeira. Observou-se que a contagem média de células somáticas (CCS) ficou dentro dos parâmetros legais, apresentando correlação negativa com a produção de leite (r = -0,23) e teor de lactose (r = -0,39). No entanto, a contagem bacteriana total (CBT) ficou acima dos parâmetros legais. O nível de escolaridade parece interferir na adoção de técnicas adequadas de ordenha e, consequentemente, na qualidade do leite, porque os produtores menos instruídos trabalham em uma infra-estrutura mais precária e obtém um produto de menor qualidade. Atividade antiviral e virucida de extratos hidroalcoólicos de própolis marrom, verde e de abelhas Jataí (Tetragonisca angustula) frente ao herpersvírus bovino tipo 1 (BoHV-1) e ao vírus da diarreia viral bovina (BVDV) 1 Cristina Mendes Peter Among the biological properties of propolis, the antimicrobial activity has received prominent attention. In this paper, we describe the antiviral and virucidal effect of three hydroalcoholic extracts of propolis (brown, green and jataí bees (Tetragonisca angustula), against bovine herpesvirus type-1 (BoHV-1) and bovine viral diarrhea Virus (BVDV). All hydroalcoholic extracts were obtained from ethanol extraction. The chemical composition of propolis extracts was determined by high-performance liquid chromatography coupled to mass spectrometer (UFLC-PDA-ESI-TOF/MS) to identify and quantify compounds such as caffeic acid and p-coumaric acid, chlorogenic acid, ferulic, and flavonoids such as rutin. Cell toxicity and antiviral activity of propolis extracts in monolayers of MDBK cells (Madin-Darby Bovine Kidney) were assessed by microscopic observation and quantified by the MTT assay (3-(4.5 dimethylthiazol-2yl) -2-5-diphenyl-2H-tetrazolato bromine). Propolis extract from Jataí bees proved to be less cytotoxic (1.57mg / ml) when compared to green extracts (0.78mg / ml) and brown (0.39mg/mL). Regarding antiviral activity, propolis has shown greater efficacy in both cellular treatments (post and pre-exposure) against BoHV-1 when compared to other extracts, ie, there was increased cell viability compared to cell and virus controls. Extracts from Jataí showed activity against both viruses (BoHV-1 and BVDV) infection in the pre-test, whereas brown propolis demonstrated action only against the BoHV-1 in the pre-infection method. To determine the virucidal activity, it were used different dilutions of virus, as well as different temperatures and incubation times. RESUMO: A busca por alternativa aos fármacos sintéticos têm revelado descobertas no campo da farmacologia e, nesse sentido, melitina e apamina, dois constituintes do veneno de abelhas, foram descritas com várias ações farmacológicas. Este estudo objetivou avaliar in vitro as capacidades antiviral e virucida destes componentes. Para tanto, células MDBK (Madin Darby Bovine Kidney), após verificação das respectivas doses tóxicas por ensaio MTT ((3-(4,5 dimetiltiazol-2yl)-2-5-difenil-2H tetrazolato de bromo), foram cultivadas em microplacas e tratadas com diferentes concentrações de apamina, melitina e sua associação. Esse tratamento ocorreu antes e após a infecção com 0,1 MOI (multiplicidade de infecção) de cepas citopatogênicas de herpesvírus bovino tipo 1 (BoHV-1) cepa Los Angeles e vírus da diarreia viral bovina (BVDV) cepa NADL. Após incubação por 72 horas, 37oC, as células foram submetidas ao ensaio MTT para estimativa da viabilidade celular. Em experimento paralelo, placas que foram submetidas ao mesmo procedimento sofreram ciclo de congelamento e descongelamento das células, para rompimento das mesmas e mensuração dos títulos virais. O ensaio virucida foi realizado incubando-se suspensões de BoHV-1 e BVDV com as soluções de apamina, melitina e associação por 24 horas a 37oC e 22oC. O título viral foi avaliado às 0 horas, 1, 2, 4, 8 e 24 horas de incubação. A concentração citotóxica para 50% das células (CC50) de melitina foi 2,32 μg/ml e apamina não demonstrou toxicidade à maior concentração testada (100μg/ml). Houve efeito antiviral da melitina sobre BoHV-1, especialmente na concentração de 2μg/ml, onde observou-se 85,96% de viabilidade celular quando o tratamento foi realizado antes da infecção e 86,78% de viabilidade quando o tratamento foi realizado após a infecção. Houve ainda redução de 90% das partículas virais de BoHV-1. Em menores concentrações (1 e 1,5μg/ml) de melitina não houve atividade antiviral, pois a viabilidade celular foi baixa, demonstrando efeito citopático do vírus. Na associação das duas substâncias houve queda no título de BVDV e observou-se maior viabilidade celular quando comparados à ação isolada dos composto sobre este vírus. Isso se confirma na atividade virucida, uma vez que houve decréscimo de 90% das partículas virais de BVDV com a associação dos dois compostos do veneno de abelhas. Atuando individualmente, melitina apresentou efeito antiviral e virucida frente ao BoHV-1, zerando seu título em apenas 2 horas a 37oC. Conclui-se que melitina tem ação antiviral e virucida frente ao BoHV-1 e sua associação com apamina potencializou seus efeitos frente ao BVDV.
É por meio da educação que se formam cidadãos conscientes de seu papel na sociedade. A educação ambiental deve estar presente em todas as séries escolares, em diferentes níveis de profundidade. Este processo educativo é fundamental, mostrando aos estudantes correlações entre sociedade, cultura e ecologia, contribuindo assim, para o fortalecimento da sua responsabilidade na conservação do meio ambiente. O objetivo foi discutir com escolares da zona rural sobre a nossa responsabilidade perante a conservação do meio ambiente e principalmente a conservação da água. As atividades foram conduzidas com crianças de escolas da zona rural da região sul. O trabalho foi desenvolvido através de visitas as escolas e visitas a locais relacionados às diferentes temáticas abordadas. Foram visitados fontes preservadas, represas, o Arroio Pelotas, a Lagoa dos Patos, e a praia do Casino, discutindo a conservação dos recursos hídricos e contaminação por resíduos. Tudo foi registrado através de fotografias e redações elaboradas pelas crianças. Em cada escola foram expostas as fotografias e redações em um grande painel, realizado palestras e discutido junto com os familiares. Com isso pode-se oportunizar uma visão diferenciada da realidade ambiental da região e uma conscientização através de mudança de atitude nas escolas, nas suas propriedades e comunidades rurais.
Salami is a ready-to-eat (RTE) product frequently purchased at street fairs in Porto Alegre. Salmonella enterica, Listeria monocytogenes, and coagulase-positive Staphylococcus (CPS) are important causes of foodborne disease and can be transmitted through the consumption of RTE foods. The aim of this study was to evaluate the presence of these pathogens in salami sold at street fairs. Ninety salami samples from three commercial brands available at street fairs were analyzed by routine bacteriological methods for Salmonella spp. and Listeria spp., as well as enumeration of CPS. In addition, two samples from each commercial brand were analyzed for water activity (aw). Samples of brand A showed aw values (0.938 and 0.942) above those set by the legislation, while brand B (0.849 and 0.860) and brand C (0.826 and 0.854) were compliant. Microbiological analyses showed that 67.7% were negative to all investigated bacteria. Salmonella Typhimurium was isolated from 4.4% (4/90) of salami samples, all from commercial brand A. Listeria monocytogenes was detected in 3.3% (3/90) of samples, from commercial brands B and C. Moreover, 7.7% (7/90) of samples contained CPS populations non-compliant with legislation. Although the great majority of salami sold at street fairs of Porto Alegre was compliant with standards, S. enterica, L. monocytogenes, and CPS ≥ 5 × 103 cfu.g-1 could be found in this RTE product. Therefore, control measures in the processing industry and consumer’s education about foodborne illness prevention should be maintained.
