ResumoO leite é um alimento natural e rico em nutrientes, porém a forma como muitas vezes é obtido interfere na sua qualidade microbiológica e nutricional. E a qualidade dos produtos é uma exigência cada vez maior do mercado consumidor. Assim, objetivou-se identificar micro-organismos presentes no leite de tanques resfriadores, na região sul do Rio Grande do Sul, e estabelecer uma correlação entre o manejo dos animais e sua presença no tanque. Foram coletadas amostras de leite em propriedades de seis municípios e realizadas análises microbiológicas para identificação microbiana. As médias das contagens foram: Staphylococcus sp. 5,32x10 6 UFC/mL, S. aureus 1,33x10 5 UFC/mL, enterobactérias 1,82x10 7 UFC/ mL. Houve presença de Escherichia coli (27,8% das propriedades), Streptococcus agalactiae (6,2%), S. dysgalactiae (37,2%), S. uberis (16,8%), Candida sp. (15,7%), Aspergillus sp. (5,8%), Trichosporum sp. (3,6%) e Cryptococcus sp. (1,5%). Identificou-se uma Odds Ratio de 3,2 para S. agalactiae com relação à ordenha manual e de 3,1 quando um único pano era usado para secagem dos tetos. Para S. dysgalactiae houve 55,8% de probabilidade da ocorrência em caso de não secagem dos tetos. Para S. bovis, uma Odds Ratio de 2,8 em propriedades que ordenham seus animais em estábulos de madeira. Houve aumento significativo (p=0,003) na contagem de células somáticas (CCS) de propriedades que realizam ordenha manual em relação àquelas mecanizadas. A ordenha manual aumentou as contagens de S. aureus (p=0,04). A realização de pré-dipping contribuiu para a redução da contagem de Staphylococcus sp. e houve diferença significativa em relação ao grupo que não realizava esta prática (p=0,04). Conclui-se que as técnicas de manejo mais precárias influenciam negativamente na qualidade microbiológica do leite, aumentando os riscos de ocorrência de agentes infecciosos e elevando as contagens de micro-organismos. The study aimed to evaluate the anti-Sporothrix sp. activity of the essential oil of Origanum majorana Linn. (marjoram), its chemical analysis, and its cytotoxic activity. A total of 18 fungal isolates of Sporothrix brasiliensis (n: 17) from humans, dogs and cats, and a standard strain of Sporothrix schenckii (n: 1) were tested using the broth microdilution technique (Clinical and Laboratory Standard Institute – CLSI M27-A3) and the results were expressed in minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC). The MIC50 and MIC90 of itraconazole against S. brasiliensis were 2 μg/mL and 8 μg/mL, respectively, and the MFC50 and MFC90 were 2 μg/mL and >16 μg/mL, respectively, with three S. brasiliensis isolates resistant to antifungal. S. schenckii was sensitive at MIC of 1 μg/mL and MFC of 8 μg/mL. For the oil of O. majorana L., all isolates were susceptible to MIC of ≤2.25–9 mg/mL and MFC of ≤2.25–18 mg/mL. The MIC50 and MIC90 were ≤2.25 mg/mL and 4.5 mg/mL, respectively, and the MFC50/90 values were twice more than the MIC. Twenty-two compounds were identified by gas chromatography with a flame ionization detector (CG-FID) and 1,8-cineole and 4-terpineol were the majority. Through the colorimetric (MTT) assay, the toxicity was observed in 70–80% of VERO cells between 0.078 and 5 mg/mL. For the first time, the study demonstrated the satisfactory in vitro anti-Sporothrix sp. activity of marjoram oil and further studies are needed to ensure its safe and effective use.
This paper aimed to describe milk production in Southern Rio Grande do Sul state, Brazil, and to identify factors that affect milk quality at this region. The average age of regional dairy farmers is 49, and 67.9% has not concluded elementary school. Dairy farming is carried out on properties with an average of 26.06 hectares and 8.4 lactating cows. Most of them (32.83%) yield 50 to 100 L/day. Among the properties, 13.21% yield up to 30 L/day, and only 1.89% produces over 500 milk liters a day. Average yield was 6.8 L/day. Regarding to milking procedure, 39.3% farmers milk manually the animals, only 14.2% performed pre-milking teat disinfection, and 53.9% uses a single cloth to dry all animal teats. For infrastructure, 52.8% milks animals in wooden cowsheds. We observed that the average somatic cell count (SCC) was within legal parameters, presenting negative correlation with milk production (r = -0.23) and lactose content (r = -0.39). However, the total bacterial count (TBC) was above legal parameters. Education level seems to interfere in management and milk quality, because the less educated groups are, the less adequate are infrastructure, management, and product quality. Data show that there is a lower quality milk production where poor management techniques are adopted and owner education level affects milk quality. A idade média dos produtores de leite da região é de 49 anos, e 67,9% não concluíram o ensino fundamental. A atividade leiteira é realizada em propriedades com média de 26,06 hectares e 8,4 vacas em lactação. A maioria dos produtores (32,83%) produz entre 50 a 100 litros de leite por dia. Entre as propriedades, 13,21% produz até 30 L/dia, e apenas 1.89% produz mais de 500 litros de leite por dia. A produtividade média foi de 6,8 L/vaca/dia. No que diz respeito ao procedimento de ordenha, 39,3% dos produtores de leite ordenha seus animais manualmente, apenas 14,2% realizam desinfecção dos tetos antes da ordenha, e 53,9% usa um único pano para secar todos os tetos dos animais. Com relação à infra-estrutura, 52,8% dos produtores ordenham seus animais em estábulos de madeira. Observou-se que a contagem média de células somáticas (CCS) ficou dentro dos parâmetros legais, apresentando correlação negativa com a produção de leite (r = -0,23) e teor de lactose (r = -0,39). No entanto, a contagem bacteriana total (CBT) ficou acima dos parâmetros legais. O nível de escolaridade parece interferir na adoção de técnicas adequadas de ordenha e, consequentemente, na qualidade do leite, porque os produtores menos instruídos trabalham em uma infra-estrutura mais precária e obtém um produto de menor qualidade. RESUMOA mastite bovina é uma doença importante na exploração leiteira, não apenas pelas perdas econômicas diretas que promove, mas também pelas perdas indiretas e o potencial risco à saúde pública. Dentre as principais causas de infecções intramamárias, destacam-se as bactérias do gênero Staphylococcus spp., sendo que Staphylococcus aureus é o agente etiológico predominante em mastite subclínica. O objetivo desse trabalho foi verificar a frequência de mastite subclínica em oito rebanhos localizados na região Sul do Rio Grande do Sul (Brasil) e a relação da enfermidade com a presença de S. aureus. Adicionalmente, pesquisou-se a presença de S. intermedius e S. hyicus nas amostras de leite obtidas. Para identificação da doença, utilizou-se o California Mastitis Test (CMT). A identificação da espécie de Staphylococcus spp. foi feita em meio de cultura ágar BairdParker, com posterior confirmação das colônias suspeitas em coloração de gram, prova de catalase, pesquisa de coagulase livre e pesquisa de termonuclease. A mastite subclínica foi constatada em 53,6% dos animais testados. A presença de Staphylococcus coagulase positiva foi identificada em 12,6% dos animais com mastite subclínica. Nesses mesmos animais, a bactéria identificada como S. aureus foi o agente etiológico presente em 17,6% dos casos. Adicionalmente, pode-se perceber que, dentre o grupo identificado como coagulase positiva, 85,7% corresponderam a S. aureus, enquanto 8,5% mostraram características bioquímicas compatíveis com S. intermedius e 5,8% foram consideradas S. hyicus. ABSTRACT FREQUENCY OF STAPHYLOCOCCUS AUREUS FROM BOVINE SUBCLINICAL MASTITIS CASES, IN SOUTHERN RIO GRANDE DO SUL, BRAZIL.Bovine mastitis is an important disease in dairy farming, not only by promoting direct economic losses, but also for indirect losses and the potential risk to public health. The main causes of intramammary infections include the bacteria of the genus Staphylococcus spp., Staphylococcus aureus being the predominant etiologic agent in subclinical mastitis. Atividade antiviral e virucida de extratos hidroalcoólicos de própolis marrom, verde e de abelhas Jataí (Tetragonisca angustula) frente ao herpersvírus bovino tipo 1 (BoHV-1) e ao vírus da diarreia viral bovina (BVDV) 1 Cristina Mendes Peter Among the biological properties of propolis, the antimicrobial activity has received prominent attention. In this paper, we describe the antiviral and virucidal effect of three hydroalcoholic extracts of propolis (brown, green and jataí bees (Tetragonisca angustula), against bovine herpesvirus type-1 (BoHV-1) and bovine viral diarrhea Virus (BVDV). All hydroalcoholic extracts were obtained from ethanol extraction. The chemical composition of propolis extracts was determined by high-performance liquid chromatography coupled to mass spectrometer (UFLC-PDA-ESI-TOF/MS) to identify and quantify compounds such as caffeic acid and p-coumaric acid, chlorogenic acid, ferulic, and flavonoids such as rutin. Cell toxicity and antiviral activity of propolis extracts in monolayers of MDBK cells (Madin-Darby Bovine Kidney) were assessed by microscopic observation and quantified by the MTT assay (3-(4.5 dimethylthiazol-2yl) -2-5-diphenyl-2H-tetrazolato bromine). Propolis extract from Jataí bees proved to be less cytotoxic (1.57mg / ml) when compared to green extracts (0.78mg / ml) and brown (0.39mg/mL). Regarding antiviral activity, propolis has shown greater efficacy in both cellular treatments (post and pre-exposure) against BoHV-1 when compared to other extracts, ie, there was increased cell viability compared to cell and virus controls. Extracts from Jataí showed activity against both viruses (BoHV-1 and BVDV) infection in the pre-test, whereas brown propolis demonstrated action only against the BoHV-1 in the pre-infection method. To determine the virucidal activity, it were used different dilutions of virus, as well as different temperatures and incubation times.
