Characterization of Bifidobacteria from infant feces with different mode of birth at Purwokerto, Indonesia

Abstract: Bifidobacteria belongs to the so-called beneficial intestinal flora. Before attempting to raise their intestinal levels to improve the health status of the host, it is importance to know about physiological variations in the Bifidobacterial colonization of the human intestine. Birth process influenced the diversity of Bifidobacteria in infant feces. This research was intended to isolate and characterize Bifidobacterium spp. as well to evaluate their presence in the feces of infants who were born by mode of nor… Show more

“…The milky white colonies have a flat edge/smooth (entire), some are uneven such as wool, round, shiny, and convex elevation with varied sizes. These isolates have morphological characters referring to Bifidobacterial genus, that is, colony with milky white color or slightly creamy, rounded form with the diameter of 0.1-0.5 mm as well as Gram-positive (Figure 1) [9], [10], [11], [12]. The morphology of E. coli colonies of the isolated bacteria exhibited pink color, small rod shaped, and Gram-negative bacilli (Figure 2) [13].…”

“…Bacterial incubation was carried out for 48 hours at 37 o C. Isolates that expressed a clear zone around the growing colony were assumed as Lactic Acid Bacteria. The bacterial characterization was performed following Hendrati et al (2017). The selected isolates were stocked on MRSA slant medium.…”

“…One loop of LAB isolate was inoculated in MRS Broth (MRSB) medium and incubated for 18 hours at 37 o C. The bacterial culture was centrifuged for 10 minutes at 10,000 rpm to obtain a bacterial cell-free supernatant (CFS). Its inhibition activity was evaluated using Kirby's Bauer method by following Hendrati et al (2017). The pathogenic bacteria were inoculated on Nutrient Broth (NB) medium (1% v/v), then was incubated for 8 hours at 37 o C. The pathogenic bacteria were spread on NA medium.…”
