Objectives: In this study, we evaulated the distribution and alteration of hepatitis C virus (HCV) genotypes throughout years which has a clinical importance in the treatment and follow-up. Materials and Methods: Test results obtained from blood samples sent to the molecular microbiology laboratory at Aydın State Hospital for HCV genotype determination were analyzed retrospectively. A total of 182 samples collected between 2014 and 2018 were enrolled in the study. The determination of genotype and viral load of the samples were performed by real time polymerase chain reaction. Results: 53.8% (98/182) of the samples were collected from male patients and 46.2% (84/182) from female patients. The mean age of the patients was 58.5±15.5 years. 69.2% of the samples were genotype 1b, 18.1%-genotype 1a, 2.2%-genotype 1 (those different from subtype 1a and 1b), 1.7%-genotype 2, 7.2%genotype 3, and 1.7% of the samples were genotype 4. Conclusion: In the present study, genotype 1 was the most common genotype (89.5%). Additionally, we have observed a decrease in the frequency of genotype 1b and a slightly increase in the frequency of other genotypes. Determination of HCV genotypes is important for treatment and prognosis of HCV infections.

Dermatofi tozların tanısında kullanılan klasik yöntemlerden direkt mikroskopik incelemenin tür ayrımını yapamaması, kültürün ise uzun süre gerektirmesi ve duyarlılığının düşük olması gibi dezavantajları bulunmaktadır. Bu çalışmada, klinik örneklerden dermatofi tlerin saptanmasında ve kültürden izole edilen suşların tanımlanmasında polimeraz zincir reaksiyonu (PCR) yönteminin performansının araştırılması amaçlanmıştır. Çalışmaya, dermatofi toz ön tanısı konulan 110 hastaya (69 kadın, 41 erkek; yaş aralığı: 4-82 yıl) ait 63'ü deri ve 60'ı tırnak kazıntısı olmak üzere toplam 123 örnek alınmıştır. Örnekler, rutin direkt mikroskopi ve mantar kül-türünün yanı sıra iki turlu (nested) PCR (nPCR) yöntemine dayalı iki ayrı protokol ile incelenmiştir. Birinci protokolde dermatofi tlerin kitin sentaz genini (CHS-1) hedefl eyen pan-dermatofi t nPCR; ikinci protokolde ise Trichophyton rubrum ve T. mentagrophytes'e özgül ITS-1 genlerini hedefl eyen primerlerin kullanıldığı nPCR uygulanmıştır. Kültürde üreyen suşlara da aynı şekilde PCR uygulanmıştır. Pan-dermatofi t nPCR'da pozitif, T.rubrum/T.mentagrophytes'e özgül nPCR ile negatif sonuç veren örnekler ayrıca dizi analizine alınmıştır. Çalışmamızda, örneklerin 62'sinde (%50) direkt mikroskopik incelemede hif ve/veya spor yapıları görülmüş, 30'unun (%24) kültüründe dermatofi t üremiştir. T.rubrum/T.mentagrophytes'e özgül nPCR ile kültürde üreyen izolatların 28'i T.rubrum, ikisi T.mentagrophytes olarak tanımlanmıştır. Direkt uygulamada klinik örneklerin 67'si (%55) pan-dermatofi t nPCR ile, 65'i (%53) ise T. rubrum/T. mentagrophytes'e özgül nPCR ile pozitif bulunmuştur. Direkt mikroskopide mantar elemanı görülmeyen örneklerin kültüründe de üreme olmamış, bu örneklerin dokuzu pan-dermatofi t nPCR, sekizi T. rubrum/T. mentagrophytes'e özgül nPCR ile pozitif sonuç vermiştir. This paper gives an update on the local distributions of HCV genotypes in Aydin province of Turkey, provides a comparison with the previous records, and discusses the potential causal reasons shaping the evolving genotype profiles. Patient files from 2011 to 2016 were retrospectively analyzed, and newly detected cases were documented. Out of 286 patients, male and female ratios were determined to remain nearly the same (~50%). Genotype 1 was still the most common (90.2%), followed by genotype 3 (5.9%), genotype 2 (2.1%), and genotype 4 (1.4%) in frequency. There were international patients (4.50%). One patient had genotyped 2+3 together. Genotypes 4 and 2+3 were detected for the first time, and the patients with genotype 4 were interestingly all male and also domestic individuals. However, these patients traveled or lived abroad in the past due to occupational reasons, thereby likely acquired the infection while abroad. HCV surveillance system is currently inadequate and some infected patients may go undetected in the province. Remapping the regional distribution of HCV genotypes from time-to-time is required for identifying the local dynamics and causes leading to it. This process enhances the clinical preparation and readiness for the better management of the disease.
