This study was carried out with an objective to investigate the antibacterial and antifungal potentials of leaves of Cassia fistula Linn. The aim of the study is to assess the antimicrobial activity and to determine the zone of inhibition of extracts on some bacterial and fungal strains. In the present study, the microbial activity of hydroalcohol extracts of leaves of Cassia fistula Linn. (an ethnomedicinal plant) was evaluated for potential antimicrobial activity against medically important bacterial and fungal strains. The antimicrobial activity was determined in the extracts using agar disc diffusion method. The antibacterial and antifungal activities of extracts (5, 25, 50, 100, 250 μg/ml) of Cassia fistula were tested against two Gram-positive—Staphylococcus aureus, Streptococcus pyogenes; two Gram-negative—Escherichia coli, Pseudomonas aeruginosa human pathogenic bacteria; and three fungal strains—Aspergillus niger, Aspergillus clavatus, Candida albicans. Zone of inhibition of extracts were compared with that of different standards like ampicillin, ciprofloxacin, norfloxacin, and chloramphenicol for antibacterial activity and nystatin and griseofulvin for antifungal activity. The results showed that the remarkable inhibition of the bacterial growth was shown against the tested organisms. The phytochemical analyses of the plants were carried out. The microbial activity of the Cassia fistula was due to the presence of various secondary metabolites. Hence, these plants can be used to discover bioactive natural products that may serve as leads in the development of new pharmaceuticals research activities.
The present study is aimed to investigate antioxidant activity of the extracts of Cassia fistula Linn. (Leguminosae) fruit pulp. Cassia fistula Linn., a Indian Laburnum, is widely cultivated in various countries and different continents including Asia, Mauritius, South Africa, Mexico, China, West Indies, East Africa and Brazil as an ornamental tree for its beautiful bunches of yellow flowers and also used in traditional medicine for several indications. The primary phytochemical study and in vitro antioxidant study was performed on hydro alcoholic extract of fruit pulp. Phytochemical screening of the plant has shown the presence of phenolic compounds, fatty acids, flavonoids, tannins and glycosides. Phenolic content was measured using Folin-Ciocalteu reagent and was calculated as gallic acid equivalents. Antiradical activity of hydro alcoholic extract was measured by DPPH (2,2-diphenyl-1- picrylhydrazyl) assay and was compared to ascorbic acid. Ferric reducing power of the extract was also evaluated by Oyaizu method. In the present study, three methods were used for evaluation of antioxidant activity. First two methods were for direct measurement of radical scavenging activity and third method to evaluate the reducing power. Results indicate that hydro alcoholic fruit pulp extracts have marked amount of total phenols which could be responsible for the antioxidant activity. These in vitro assays indicate that this plant extract is a significant source of natural antioxidant, Cassia fistula fruit pulp extract shows lower activity in DPPH and total phenol content as compared with standard which might be helpful in preventing the progress of various oxidative stresses.
Triphala is categorized as a rejuvenator and antioxidant-rich Ayurvedic herbal formulation and has traditionally been used in various gastric problems including intestinal inflammation. The aim of the present study was to examine the comparative enteroprotective effect of Triphala formulations against methotrexate-induced intestinal damage in rats. Triphala formulations were prepared by mixing equal (1:1:1) and unequal (1:2:4) proportions of Terminalia chebula Retz., Terminalia belerica (Gaertn.) Roxb. and Emblica officinalis Gaertn. Intestinal damage was induced by administering methotrexate (MTX) in a dose of 12 mg/kg, orally for 4 days to albino rats. The intestinal damage response was assessed by gross and microscopical injury, measuring the intestinal permeability to phenol red and tissue biochemical parameters. Triphala equal and unequal formulations at the dose of 540 mg/kg significantly restored the depleted protein level in brush border membrane of intestine, phospholipid and glutathione content and decreased the myeloperoxidase and xanthine oxidase level in intestinal mucosa of methotrexate-treated rats. In addition, Triphala unequal formulation showed significant decrease in permeation clearance of phenol red with significant attenuation in the histopathological changes, level of disaccharidase in brush border membrane vesicles and lipid peroxidation content of intestinal mucosa. Based on the data generated, it is suggested that Triphala unequal formulation provides significantly more protection than Triphala equal formulation against methotrexate-induced damage in rat intestine.
New Antihistaminic Agents: Synthesis and Evaluation of H1-Antihistaminic actions of 3-[(N,N-Dialkylamino)alkyl)-1,2,3,4-tetrahydro-(1H)-thioquinazolin-4(3H)-ones and Their oxo Analogues
Amalakayas Rasayana (AR) is a polyherbal formulation mentioned in Ayurveda to treat aging and age-associated diseases. Being an antiaging drug, AR may have antioxidants and free radical scavenging activity to minimize free radical-induced damage which is a key cause of aging. The methanolic extract of AR was evaluated in vitro for total phenolic and tannin content, free radical scavenging activity, superoxide radical scavenging activity, and reducing power. The total phenolic content was measured using Folin-ciocalteu reagent against gallic acid [relative standard deviation (R2) = 0.998]. Total tannin was estimated using the Stephen method and was found to be 2.82% w/w. Free radical scavenging activity was measured by 2,2-diphenyl-1-picryl hydrazyl assay and R2 was 1. Superoxide radical scavenging activity was done by ethylene diamine tetra acetate and Nitro Blue Tetrazolium Chloride assays against ascorbic acid and R2 was 0.976 (EC50= 77.5 μg/ml). Ferrous reducing power was evaluated by Oyaizu method where R2 was 0.986. All studies showed that AR possesses antioxidant activity. The results of this study suggest that the antioxidant and free radical scavenging activity of AR may explain its rasayana effect and justify its use as a medicine for age associated diseases.
Herbal medicines have a long therapeutic history and are still serving many of the health needs of a large population of the world. However, the quality control and quality assurance still remains a challenge because of the high variability of chemical components involved. Herbal drugs, singularly and in combinations, contain numerous compounds in complex matrices in which no single active constituent is responsible for the overall efficacy. This creates a challenge in establishing quality control standards and standardization of finished herbal drugs. Many preparations have been mentioned in Ayurvedic text books for the treatment of Urdhwaga Amlapitta (non-ulcer dyspepsia). Dashanga Kwatha is one such known formulation. In this study, Dashanga Kwatha was converted into tablet form to increase the shelf life, make it easy to dispense, for dose fixation, etc. The Dashanga Kwatha Ghana tablet was subjected to organoleptic analysis, phytochemical analysis, and qualitative analysis to detect the presence of various functional groups, and to high performance thin layer chromatography (HPTLC) examination by optimizing the solvent systems. The investigation revealed the presence of tannins, mucilage, ascorbic acid, alkaloids, saponins, glycosides, flavonoids and carbohydrates mainly.
