Activation of the estrogen receptor-α (ER-α) signaling pathway is a significant factor in the initiation of carcinogenesis in various types of tumors due to the genomic and non-genomic effects of estradiol in cancer cells. However, data on the expression of ER-α and aromatase on stromal and immune cells in the tumor microenvironment (TME) point to an additional mechanism by which estrogens increase tumor malignancy. There is growing evidence that TME can affect tumor immunity by increasing the immune response or reducing immunoreactivity.The important role of estrogen and the estrogen receptor signaling pathway in the response of the tumor microenvironment in cancer of various localizations, not only classical hormone-dependent cancers, has been proven. However, the clinical effectiveness of blocking the effect of estrogen on tumor growth has been primarily shown in cancer of the female reproductive system. At the same time, data on the significant role of TME in the development of endocrinotherapy resistance in breast cancer treatment are of great interest.Despite the possibilities of standard therapy, a more in-depth study on the role of various TME components in cancer evolution, creation of a micrometastatic niche, as well as in the response to therapy may result in development of new strategies for cancer treatment. It is also necessary to study the possibilities of overcoming the immunosuppressive effect of the estrogen receptor signaling pathway on TME in order to increase the survival rates in patients with hormone-dependent cancers, particularly, breast cancer.
The epithelial‑to‑mesenchymal transition (EMT) is a cellular biological process, that occurs in a wide range of cells and tissues and is triggered by complex regulatory networks involving transcriptional control with SNAIL, ZEB1, ZEB2, Twist, SLUG, E‑cadherin, vimentin, tumor microenvironment and genetic characteristics. EMT is represented by the multi‑stage development and transformation of cells of the epithelial phenotype into cells that acquire mesenchymal features of various severity. These quasi‑mesenchymal cells are characterized by stemness, tumor heterogeneity, increasing invasiveness, drug resistance and a tendency to distant metastasis, which leads to the proliferation of neoplastic cells, tumor dissemination and initiation of metastasis, which induces the therapy resistance and the oncological recurrence.This review is based on the latest scientific publications about the EMT phenomenon, indexed in PubMed. The aim of the study was to evaluate the biochemical and molecular pathogenetic mechanisms of EMT and the effect of EMT markers on the progression of neoplastic processes and the effectiveness of the treatment. Nowadays the proper EMT scheme that combines all the molecular transformations of sells with quasi‑mesenchymal phenotype doesn’t exist. But analyzing the features of this cellular program, we can find the proper therapy, that could be able to suppress the plasticity of cancer cells, prevent EMT induction by blocking contextual signals, and induce mesenchymal‑epithelial transition. All these aspects will lead to the reduction of the risk of tumor dissemination and the increase of the effectiveness of cancer treatment.
Oropharyngeal squamous cell carcinoma has been traditionally associated with tobacco and alcohol consumption. Nevertheless, latter 30 years have shown squamous cell carcinoma (OPSCC) incidence stagnation and increasing, despite the decrease in smoking prevalence. The incidence was recognized among white men of middle age, often non-smokers or former smokers in the investigations with smoking cessation data. It differs from traditional patient with OPSCC, older men, heavy smoker or alcohol drinker. The incidence of OPSCC is increased due human papilloma virus (HPV) infection. The infection transition is associated with sex and oro-genital contact may lead to HPV-infection of oropharynx and oral cavity. There are multiple types of HPV, but the majority of OPSCC is associated with HPV 16 type. Epidemiology of HPV-associated OPSCC, HPV-infection of oral cavity and/or oropharynx and HPV detection are discussed in the review.
Acute respiratory disease COVID-19 caused by the SARS-CoV-2 coronavirus demonstrate weak clinical manifestation in most patients. However, pneumonia and acute respiratory distress syndrome in some cases may cause serious problems due to the lack of effective etiotropic and pathogenetic therapy. Presumably, SARS-CoV-2 leads to the delayed type I interferon activation and loss of control over virus replication in the early stages of infection, which is why the adaptive CD8+T-cell response must be controlled to avoid the development of pulmonary pathology. These data should be taken into account when developing strategies for COVID-19 therapy. Mesenchymal stem cells therapy serves as possible treatment opportunity for severe forms of the disease due to their homing, pronounced anti-inflammatory and antifibrotic properties. It was found that in viral infections, including COVID-19, mesenchymal stem cells can synthesize antiviral defense mediators under the influence of interferon causing resistance to viruses. Thus, mesenchymal stem cells are able to provide comprehensive anti-inflammatory protection, which leads to clinical improvement in patients with COVID-19.
The aim – to evaluate the functional state of ocular adnexa and of the eyeball after cryodestruction of benign eyelid and conjunctiva tumors using modern cryosurgical equipment.
Materials and methods. Into the study, 87 patients (87 eyes) were included with eyelid and conjunctiva benign tumors after cryosurgical treatment of atheroma, cyst, papilloma, nevus, granuloma, xanthelasma using an autonomous cryoapplicator of porous permeable titanium nickelide.
Results and conclusions. The study showed a high efficacy of cryogenic treatment method using an autonomous cryoapplicator of porous permeable titanium nickelide. Depending on dimensions of the lesion, exposure duration, and the number of repeated applications at the session, the recovery occurred within 1–2 months without functional defect and healthy tissue injury.
