Durch Einwerfen von Eisstückchen wurde die Azetylverbindung zur Abscheidung gebracht. Sie wurde Std. im Wasser stehcngelassen und dann mit einem Glasstab gut durchgeknetet. Dabei zediel die zunächst öligo Mame in ein bröckeliges Pulver. Das üherstehende Wasser wurde abgegossen und der Rückstand awcimal rnit Wasser gewaschen. Dann wurde unter Zusatz von wenig Kohle aus DBproz. Äthanol mehrere Male umkristallisiert. H exaece t y lg allo ca t ec hi n : Der au9 dem Papierbrei extrahierte Rückstand betrug etwa 0,l g. Er wurde rnit 1 ml Pyridin und 1 ml Essigsaureanhydrid versetzt und 2 Std. stehengelassen. Beim Einwcrfcn von Eisstückchen schied sich ein amorpher, flockiger Niederschlag ab, der zweimal mit 5 ml Wasser gewaschen wurde. Dann wurde au6 9Bproz. Athanol umkristallisiert. Reaktion nach Kahlenberg: Die in Chloroform gclöste Substanz wurde mit einigen Kristallen Antimonpentachlorid versetzt und kurz erwärmt. Dabei trat eine blauviolette Firbung auf. Eiden') erwähnte, aber nicht isolierte 4-Hydroxymethylenderivat des 1,2-Diphenyl-3,5-dioxo-pyrazolidins veranlaBt uns zu einer kurzen Notiz iiber eigene Untersuchungen2).Als methylenaktive Verbindung reagiert 1,2-Diphenyl-3,5-dioxo-pyrazolidin I mit den Vilsmeier-Reagenzien Dimethylformamid (DhJFA) und Phosphoroxychlorid zu 4-Dimethylaminomethylen-1,2-diphenyl-3,5-dioxo-p~azolidin 11, das F. Eiden weniger glatt durch direktes Erhitzen von I mit DMFA in Acetanhydrid darstellte.Alkalische Hydrolyse der Verbindung I1 fiihrt zum Natriumsalz des in der Enolform vorliegenden 4-Formyl-l,2-diphenyl-3,5-dioxo-pyrazolidins. Der Schmp. des gegen Alkali sehr beständigen, kristallinen Salzes liegt bei 290". Vorsichtige Einwirkung von Säure fällt die freie Verbindung I11 vom Schmp. 135', die wahrscheinlich als Chelat der Hydroxymethylenform vorliegt. 4-Formyl-I ist eine unbeständige und vor allem säureempfindliche Substanz. Acetoveratron gibt mit dem Vilsmeier-Komplex neben 1 unter schonenden Bedingungen den Dialdehyd 2, der rnit Morpholin 4 ergibt. Mit konz. Alkali wird 4 in das Na-Salz von 5 uberfiihrt. 2 gibt mit Hydroxylamin 8. 1 reagiert mit Pyridin nicht, bei Gegenwart von Siiuren wird das Pyridiniumsalz 9 erhalten. 1 gibt bei vielen Umsetzungen geringe Mengen von 10, das in DMF mit Salzsiiure Hauptpmdukt wird. Vilsmeier Reaction with Acetoveratrone and Properties of the Reaction ProductsFrom acetoveratrone with the Vilsmeier complex results besides 1 under mild conditions the dialdehyde 2 which reacts with morpholine to form 4. By use of concentrated alkali 4 is transferred into the sodium salt of 5. 2 yields 8 by reaction with hydroxylamine. 1 does not react with pyridine, in the presence of acids the pyridinium salt 9 is obtained. In many reactions 1 produces in small amounts 10 which becomes the main product in DME' and hydrochloric acid.Acetoveratron lafit sich nach Vilsmeier mit Dhnethylformamid/Phosphoroxychlorid zu ~-Chlor-3,4-dimethoxyzimtaldehyd 1 umsetzenl). &en besonders reinen Aldehyd erhalt man beim Arbeiten in Tetrahydrofuran-Losug. Dabei kann das ausgefallene Imoniumsalz-Zwischenprodulrt abgesaugt und fiir sich zum Aldehyd hydrolysiert werden. Die Ausbeuten betragen bei dieser Arbeitsweise maximal 50 yo.Bei Umsetzungen in DMF-Losung lrann man unter schonenden Bedingungen den sehr unbestandigen und zu spontaner Zersetzung neigenden Dialdehyd 2, ar-Formyl-~-chlor-3,4-dimethoxyzimtaldehyd, in Ausbeuten bis zu 38 yo isolieren. Die Abtrennung von 2 gelingt, w eil das als Zwischenprodukt entstehende Imoniumsalz bereits in einem pH-Bereich hydrolysiert, in dem das Imoniumsalz von 1 noch stabil ist. xhnliche Polyformylierungen wurden auch von Armold2) beobachtet. 2 kann aber nicht durch weitere Formylierung von 1 hervorgehen, da 1 rnit Vilsmeier-Reagens nicht weiterreagiert.
Eingegangen a m 19. April 1968 I -Morpholino-cyclohexen-( I) reagiert mit Zimtaldehyd zu 2-Morpholino-4-phenyl-bicyclo-[3.3. I]nonanon-(9) (l), wobei das nicht isolierte Diels-Alder-Zwischenprodukt 14 entsteht. Dabei werden erstmals drei der vier moglichen Stereoisomeren von 1 (a-d) erhalten, deren Stereochemie durch Abbau und Umlagerung zum aquatorialen 2-Phenyl-bicyclo[3.3.1 Inonan (5b) geklart wird. Umsetzung von Morpholino-cyclohexen mit ZimtaldehydZimtaldehyd und Morpholino-cyclohexen reagieren in Benzol oder Toluol unter Addition mit 70--80% d.Th. zu den drei isomeren 2-Morpholino-4-phenyl-bicyclo-[3.3.l]nonanonen-(9) l acl), die im Verhaltnis 10: 1 : 0.5 entstehen. Das vermutete vierte Isomere (1 d) konnte nicht sicher nachgewiesen werden.Die Kondensation von Zimtaldehyd mit Morpholino-cyclohexen unter Bildung von nur einem Reaktionsprodukt, das offenbar verunreinigtes 1 a darstellt, ist beschriebenz).Bei der Aufarbeitung mit verdiinnter Salzsaure/Isopropylalkohol bleibt das Hydrochlorid von 1 b groBtenteils gelost, wahrend die Hydrochloride von l a und c ausfallen.Die Trennung von l a und l c beruht auf der verschiedenek Loslichkeit der Hydrochloride in Wasser oder waRr. Athanol: Das Hydrochlorid von l c ist darin schwerer Ioslich; die Base 1 c lost sich in Isopropylalkohol leichter. Die Trennungen wurden diinnschichtchromatographisch verfolgt.Die Ausbeuten und das Verhaltnis von a : b : c schwanken bei verschiedenen Ansatzen; Siurekatalyse scheint die Gesamt-Ausbeute zu begiinstigen.
Acetoveratron (II) setzt sich mit Vilsmeier‐Reagens je nach Reaktionsführung zu den Aldehyden (I) oder (III) um.
