RESUMONeste estudo foram avaliados diferentes métodos para a superação da dormência em sementes de sucupira-preta. O experimento foi realizado inteiramente ao acaso em esquema fatorial 3 x 7, correspondendo a três lotes de sementes e sete tratamentos, com 4 repetições de 25 sementes. Foram utilizados ácido sulfúrico por 4, 8 e 12 minutos, água a 80°C por 5 e 10 minutos, escarificação mecânica, mantendo-se uma testemunha. Foi observado que a espécie possui dormência tegumentar e que todos os tratamentos utilizados permitiram a entrada de água nas sementes. Contudo, alguns métodos estudados, como a água a 80°C por 10 minutos, resultou em um elevado número de sementes mortas. O método mais eficiente para a superação da dormência de sementes de sucupira-preta é a imersão em ácido sulfúrico, por 4, 8 ou 12 minutos, beneficiando a porcentagem e a velocidade de germinação.Termos para indexação: Germinação, escarificação, impermeabilidade do tegumento. ABSTRACTThis study evaluated diferent methods to overcome dormancy of black sucupira seeds. The experiment was installed in a fully randomized design composed by a factorial scheme 3x7, correspoding to three lots of seeds and seven treatments, with 4 replicates of 25 seeds. Sulfuric acid for 4, 8 and 12 minutes, water at 80°C for 5 and 10 minutes, sandpaper treatments were compared to the control. It was observed that this species has tegument dormancy and all treatments allowed seed imbibition. However in some of the methods used, such as like water at 80°C for 10 minutes, occurred a high number of dead seeds. The most suitable method for dormancy overcoming of black sucupira seeds was the imersion in sulfuric acid for 4, 8 and 12 minutes, that showed higher percentage and speed of germination.
INOCULATION OF BEAN SEEDS WITH Fusarium oxysporum f. sp. phaseoli THROUGH WATER RESTRICTION TECHNIQUEABSTRACT -The objective of the present work was to test methodology of inoculation of bean seeds with Fusarium oxysporum f. sp. phaseoli using water restriction technique. For that, three solutes, sucrose, potassium chloride and manitol, added to the medium potato-sucrose-agar, PSA at three water potentials (-0.8, -1.0, -1.2 MPa) and four duration periods of the exposition of seeds to the fungus (36, 72, 108 and 144h) were used. The effects of the fungus on the performance of the seeds and emerged plants were evaluated looking at germination and health ( blotter-test). The mycelial growth of the fungus in vitro, was not reduced by any osmotic treatment. Higher growth of the fungus was observed at osmotic potentials of -0.8 e -1.0 MPa. At longer durations of exposition of the seeds to the pathogen, 108 e 144h, mycelial growth of the fungus was higher and affected seed performance. The greatest mean incidence of Fusarium oxysporum f. sp. phaseoli (64%) was detected on seeds in wich KCl was used. At 144h of inoculation period on that solute, the incidence was of 70%.
Para estudar a eficiência do teste de raios X na avaliação de defeitos internos em sementes de ipê-amarelo (Tabebuia serratifolia) e ipê-roxo (T. impetiginosa), bem como verificar a conseqüência destes defeitos na germinação, as sementes foram submetidas a diferentes intensidades e tempos de exposição à radiação. Definida a intensidade de 55 kV por 25 segundos como sendo a que permitiu melhor visualização das estruturas internas, as sementes das duas espécies foram divididas em três categorias de acordo com a sua análise radiográfica em: Sem Defeitos, Com Defeitos e Vazias. As Sementes Com Defeitos foram divididas em três subcategorias: Com Pequenos Danos (menos de 50% do embrião danificado), Com Danos Severos (mais de 50% do embrião danificado) e Deformadas. As sementes foram, então, submetidas ao teste de germinação em substrato sobre areia, a 30ºC, sob luz constante. O teste de raios X é eficiente na avaliação de defeitos em sementes de ipê-amarelo e ipê-roxo. Defeitos internos detectados nas radiografias afetam a germinação dessas sementes, reduzindo a qualidade do lote.
Resumo -O objetivo deste trabalho foi avaliar os padrões eletroforéticos das proteínas resistentes ao calor e a atividade da enzima endo-β-mananase durante a germinação de sementes de alface, em alta temperatura. Sementes de oito cultivares de alface foram submetidas aos testes de germinação, primeira contagem e emergência em duas temperaturas, 20 e 35 o C. Foram calculados o índice de velocidade de germinação (IVG) e o índice de velocidade de emergência (IVE). Avaliou-se, também, a expressão das proteínas resistentes ao calor e da enzima endo-β-mananase, para todos os tratamentos. Utilizou-se o delineamento experimental inteiramente casualizado, em arranjo fatorial 2x8, com duas temperaturas e oito cultivares. A maior germinação é observada a 35 o C na cultivar Everglades, considerada termotolerante. Os padrões de proteínas resistentes ao calor em sementes de alface apresentam bandas específicas na cultivar Everglades, a 35ºC. A atividade da enzima endo-β-mananase é maior na cultivar Everglades, nessa temperatura. Essa cultivar tem potencial para utilização em programas de melhoramento de alface com vistas à tolerância a altas temperaturas durante a germinação.Termos para indexação: Lactuca sativa, endo-β-mananase, proteínas tolerantes ao calor, termoinibição. Physiological and biochemical aspects of lettuce seed germination at different temperaturesAbstract -The objective of this work was to evaluate the electrophoretic patterns of heat-tolerant proteins and the activity of the endo-β-mannanase enzyme during lettuce seed germination, at high temperature. Seeds of eight lettuce cultivars were subjected to germination, first count, and emergence tests at two temperatures, 20 and 35°C. The index of germination speed (IVG) and the emergence rate index (IVE) were calculated. The expression of heat-tolerant proteins and of the endo-β-mannanase enzyme was also evaluated for all treatments. The experimental design was completely randomized, in a 2x8 factorial arrangement, with two temperatures and eight cultivars. The highest germination is observed at 35°C in the Everglades cultivar, considered thermotolerant. The patterns of heat-tolerant proteins in lettuce seeds present specific bands in the Everglades cultivar, at 35ºC. The activity of the endo-β-mannanase enzyme is higher in the Everglades cultivar, at this temperature. This cultivar has potential for use in lettuce breeding programs aiming for tolerance to high temperatures during germination.
RESUMOO tratamento de sementes de soja com fungicidas vem sendo utilizado como importante ferramenta no controle de patógenos. Nesse trabalho, objetivou-se estudar o desempenho de sementes de soja tratadas com fungicidas e peliculizadas, antes e após o armazenamento. Foram utilizados cinco lotes de sementes, cultivar Monsoy 6101, submetidos aos tratamentos: thiabendazole+thiram sem polímero, carbendazin+thiram sem polímero, sem fungicida (testemunha) sem polímero, além desses tratamentos realizados via peliculização (com polímero). A qualidade fisiológica e sanitária das sementes foi avaliada inicialmente e após seis meses de armazenamento em condições ambientais, pelas seguintes determinações: teor de água, teste de germinação, emergência em bandeja, teste de frio e de sanidade. Conclui-se que a peliculização, em associação com fungicidas, não afeta a qualidade fisiológica das sementes de soja e o tratamento de lotes de sementes de soja com os fungicidas thiabendazole+thiram e carbendazin+thiram melhora seu desempenho e qualidade sanitária.Termos para indexação:Glycine max, thiabendazole, thiram, carbendazin, polímeros. ABSTRACTThe treatment of soybean seeds by fungicides has been used as an important tool in pathogen control. The objective of this work was to verify the performance of soybean seeds treated with fungicide and film coating, prior and after storage. Five seed lots, cultivar Monsoy 6101, were treated with thiabendazole+thiram without polymer, carbendazin+thiram without polymer, without fungicide (control), without polymer, and treatments by film coating (with polymer). The physiological and sanitary quality of the seeds was evaluated initially and after six months storage, under environmental conditions, according to the following characteristics: water content, test of germination and emergence on tray, blotter test and cold test. The film coating associated with fungicides does not affect the physiological quality of soybean seeds and the soybean seed treatment with the fungicides thiabendazole+thiram and carbendazin+thiram improves performance and sanitary quality.

RESUMOConduziu-se este trabalho com o objetivo de estudar o efeito do fungo Trichoderma harzianum como promotor de crescimento de sementes de milho tratadas com diferentes fungicidas. O experimento foi realizado em vasos em casa-devegetação, adotando o delineamento experimental inteiramente casualizado em esquema fatorial (3x2), utilizando os tratamentos com fungicidas: Captan®, Maxim® e testemunha e inoculação (com e sem inóculo) em quatro repetições. As características agronômicas avaliadas foram: altura de planta, peso da matéria seca da parte aérea e de raízes e também a técnica de isolamento do fungo nas raízes aos 45 dias após semeadura. Observou-se diferença significativa somente para o fator inóculo na característica peso de matéria seca de raízes. Pelos resultados obtidos, verificou-se que o fungo Trichoderma harzianum estimulou maior acúmulo de matéria seca nas raízes das plantas de milho e também estava presente nas raízes das plantas oriundas de sementes inoculadas, independentemente do tratamento fungicida das sementes. ABSTRACTThe objective of this work was to study the effect of the fungus Trichoderma harzianum as a growth promoter on the corn seeds treated with different fungicides. This experiment was carried out in pots in a greenhouse by adopting the experimental completely randomized design in (3 X 2) factorial scheme, by utilizing the treatments with fungicides: Captan®, Maximim® and control and inoculation (with and without an inoculum) with four replicates. The agronomic characteristics evaluated were: plant height, shoot and root dry matter weight and also the inoculum isolation technique on the rots after sowing. Significant differences were found only for the factor inoculum in the characteristic root dry matter weight. The results obtained revealed that the fungus Trichoderma harzianum stimulated greatest dry matter accumulation in the roots of corn plants and also it was present in the roots of plants from inoculated seeds regardless of the treatment of the seeds with fungicides.
