INOCULATION OF BEAN SEEDS WITH Fusarium oxysporum f. sp. phaseoli THROUGH WATER RESTRICTION TECHNIQUEABSTRACT -The objective of the present work was to test methodology of inoculation of bean seeds with Fusarium oxysporum f. sp. phaseoli using water restriction technique. For that, three solutes, sucrose, potassium chloride and manitol, added to the medium potato-sucrose-agar, PSA at three water potentials (-0.8, -1.0, -1.2 MPa) and four duration periods of the exposition of seeds to the fungus (36, 72, 108 and 144h) were used. The effects of the fungus on the performance of the seeds and emerged plants were evaluated looking at germination and health ( blotter-test). The mycelial growth of the fungus in vitro, was not reduced by any osmotic treatment. Higher growth of the fungus was observed at osmotic potentials of -0.8 e -1.0 MPa. At longer durations of exposition of the seeds to the pathogen, 108 e 144h, mycelial growth of the fungus was higher and affected seed performance. The greatest mean incidence of Fusarium oxysporum f. sp. phaseoli (64%) was detected on seeds in wich KCl was used. At 144h of inoculation period on that solute, the incidence was of 70%.

RESUMO -A disponibilidade de sementes com diferentes níveis de infecção por fungos ou outros patógenos transmissíveis por esta via é extremamente importante para diversos estudos em patologia de sementes. A presente pesquisa teve como objetivo avaliar o uso da restrição hídrica na inoculação de sementes de algodoeiro com vistas à obtenção de sementes infectadas por Colletotrichum gossypii, C. gossypii var. cephalosporioides, Botryodiplodia theobromae e Fusarium oxysporum f. sp. vasinfectum. A premissa foi baseada no fato de que infecção de sementes pode ser assegurada e controlada pela manutenção das sementes em contato com a colônia fúngica em desenvolvimento em meio agarizado contendo manitol em potencial osmótico que impeça, temporariamente, a germinação por períodos de incubação mais prolongados. Neste trabalho manitol foi testado nos potenciais hídricos de 0, -0,4, -0,6, -0,8 e -1,0 MPa. As sementes foram incubadas em câmara de crescimento vegetal a 25ºC, com fotoperíodo de 12 horas, permanecendo nestas condições por tempos variáveis, de acordo com o potencial hídrico utilizado. As sementes foram retiradas do meio e secadas sobre papel de filtro, em condições de laboratório e submetidas aos testes de germinação, emergência em bandejas e sanidade. Pelos resultados, o substrato com potencial hídrico na faixa de -0,8 a -1,0 MPa, proporcionou condições mais apropriadas para impedir temporariamente a germinação das sementes por um período de 4 e 5 dias, respectivamente, promovendo um maior índice de infecção das sementes, bem como uma maior percentagem de plântulas infectadas.Termos para indexação: infecção de sementes, patologia de sementes, pré-condicionamento de sementes. The coating technique is a strategy that guarantees high seedling emergence in the field, gain in productivity and better aggregation of agricultural inputs to the seeds. Thus, this study had the objective of evaluating the effect of the incorporation of antagonistic microorganisms, mycorrhiza, amino acids, micronutrients and growth regulators through the seed coating technique on the quality of lettuce seeds. The seeds were coated with two types of polymer that were applied together with antagonistic microorganisms (Trichoderma viride, Trichoderma polysporhum, Trichoderma stromaticum, Beauveria bassiana, Metarhizium anisopliae), mycorrhiza, amino acids, micronutrients and growth regulators according to the methodology proposed by the manufacturer. This work was aimed at evaluating the possibility of using bromophenol blue as an indicator for detecting the presence of Sclerotinia sclerotiorum in the seeds of dry-beans (Phaseolus vulgaris) and soybean (Glycine max), through incubation of the seeds on an agar medium and "blotter" substrates. The seeds were artificially inoculated with four S. sclerotiorum isolates, plated on the agar medium, named Neon, and on modified Neon agar media all incubated at 14 and 20 ºC for seven days in the dark. Half of the seeds inoculated were surface desinfested prior to plating on the medium. The seeds showing change of colour in the medium, from blue to light yellow, as well as formation of typical mycelium and sclerotia in some cases, were considered to be infected or contaminated by S. sclerotiorum. The two incubation temperatures compared did not show significant (P<0.05) differences in detection level for most of the isolates tested on the different media. According to results obtained in this study, the Neon agar medium with incubation at 14 or 20 ºC has proved to be a reliable and quick method for the detection of S. sclerotiorum mycelium in naturally infected seeds of bean and soybean.
Foi instalado um experimento em Viçosa, MG, em 11 de abril, em área infestada por escleródios de Sclerotinia sclerotiorum, com o objetivo de testar a eficiência de quatro fungicidas aplicados via água de irrigação por aspersão no controle do mofo-branco do feijoeiro (Phaseolus vulgaris). Os seguintes fungicidas e doses do ingrediente ativo por hectare foram testados: benomyl (1 kg), iprodione (0,75 kg), procimidone (0,5 kg) e fluazinam (0,5 l). As aplicações foram simuladas com regadores (35.000 l/ha). A primeira aplicação foi feita aos 39 dias após a emergência (DAE); a segunda, 13 dias depois. Nesses mesmos dias, o fluazinam também foi aplicado com pulverizador (667 l/ha) ou com regador (35.000 l/ha) entre as fileiras de feijão e rente ao solo. Este último tratamento teve por objetivo verificar se o fluazinam aplicado apenas no solo tem efeito no controle da doença. Ademais, foi utilizada uma testemunha que não recebeu fungicida. Após a colheita, avaliou-se a incidência do fungo nas sementes. Os fungicidas fluazinam (aplicado com pulverizador ou via água de irrigação sobre as plantas), benomyl e procimidone foram os mais eficientes no controle do mofo-branco e, dentre eles, apenas o procimidone não proporcionou rendimento maior que o da testemunha. O fluazinam aplicado apenas no solo reduziu a incidência da doença e a quantidade de escleródios produzidos. Os rendimentos variaram de 1.406 (testemunha) a 2.054 kg/ha (fluazinam, pulverização). Não houve influência dos tratamentos na incidência do fungo nas sementes, a qual variou de 0,25% (procimidone) a 1,08% (fluazinam aplicado no solo).
Mycotoxins are produced by the secondary metabolism of many fungi and can be found in almost 25% of the world's agricultural commodities. These compounds are toxic to humans, animals, and plants and therefore, efforts should be made to avoid mycotoxin contamination in food and feed. Besides, up to 25% of all harvested fruits and vegetables are lost due to storage molds and/ or mycotoxin contamination and many methods have been applied to mitigate these issues, but most of them rely on the use of fungicides. Although chemicals are often the first defensive line against mycotoxigenic fungi, the indiscriminate use of fungicides are awakening the public perception due to their noxious effects on the environment and human/animal health. Thus, there is an increasing public pressure for a safer and eco-friendly alternative to control these organisms. In this background, biological control using microbial antagonists such as bacteria, fungi and yeasts have been shown to be a feasible substitute to reduce the use of chemical compounds. Despite of the positive findings using the biocontrol agents only a few products have been registered and are commercially available to control mycotoxin-producing fungi. This review brings about the up-to-date biological control strategies to prevent or reduce harvested commodity damages caused by storage fungi and the contamination of food and feed by mycotoxins.Index terms: Biocontrol, microbial antagonists, postharvest decay, food safety. RESUMOAs micotoxinas são produzidas pelo metabolismo secundário de várias espécies de fungos e podem ser encontradas em quase 25% das commodities agrícolas. Esses compostos são tóxicos a humanos, animais e plantas e, portanto, esforços para evitar a contaminação de micotoxinas em alimentos e rações devem ser feitos. Além disso, até 25% das frutas e legumes em pós-colheita são perdidos em decorrência do ataque de fungos de armazenamento e/ou contaminações por micotoxinas. Vários métodos têm sido aplicados para mitigar os problemas de micotoxinas, mas a maioria deles se baseia no uso de fungicidas. Embora os produtos químicos sejam, muitas vezes, a primeira estratégia de defesa contra fungos micotoxigênicos, o uso indiscriminado de fungicidas vem despertando a percepção pública, em razão de seus efeitos nocivos sobre o meio ambiente e à saúde humana/animal. Assim, existe uma crescente pressão pública em busca de alternativas mais segura e não nocivas ao meio ambiente para controlar estes organismos. Nesse contexto, o controle biológico utilizando antagonistas microbianos, tais como bactérias, fungos e leveduras têm mostrado ser um substituto viável para reduzir a utilização de produtos químicos. Para impedir ou reduzir a germinação de sementes de espécies dicotiledôneas, o uso de 2,4-D (sal de sódio) tem sido a alternativa disponível havendo, no entanto, questionamentos sobre este composto por inúmeras razões, inclusive o aspecto toxicológico. Neste trabalho, o objetivo foi estudar a viabilidade do uso da restrição hídrica como recurso para reduzir a germinação das sementes em substituição ao 2,4-D. Foram utilizados os restritores hídricos manitol e Cloreto de sódio, nos potenciais -0,6, -0,8, -1,0 e -1,2 MPa, sendo avaliados seus efeitos sobre a germinação das sementes, comprimento de plântulas e crescimento micelial in vitro dos principais fungos associados a sementes de algodoeiro e seu desenvolvimento sobre as sementes no teste de incubação em papel de filtro. A restrição hídrica proporcionada pelos solutos e potenciais osmóticos testados foi capaz de reduzir a germinação e o comprimento das plântulas em níveis considerados satisfatórios para a análise sanitária de sementes de algodoeiro, além de não interferir na recuperação dos principais fungos sobre as sementes no teste de papel de filtro, revelando-se desta forma como uma alternativa eficaz em substituição ao uso do 2,4-D. O crescimento micelial dos fungos em meio agarizado foi reduzido nos potenciais osmóticos a partir de -0,6 MPa. ABSTRACT Potential of water restriction use in cotton seed health testingThe fast germination of cotton seeds that occurs in blotter and agar seed health tests may become a difficulty in the identification of fungi, thus affecting the efficiency of these tests. The available alternative to impede or to reduce seed germination of dicotyledoneous species is the incorporation of 2.4-D (sodium salt formulation) solution to the substrate.
