Fungi have the ability to degrade vegetal cell wall carbohydrates, and their presence in the digestive tract of ruminants can minimize the effects of lignified forage on ruminal fermentation. Here, we evaluated enzyme production by Aspergillus spp. isolates from the digestive tracts of cattle grazed in tropical pastures during the dry season. Filamentous fungi were isolated from rumen and feces by culture in cellulose-based medium. Ninety fungal strains were isolated and identified by rDNA sequence analysis, microculture, or both. Aspergillus terreus was the most frequently isolated species, followed by Aspergillus fumigatus. The isolates were characterized with respect to their cellulolytic, xylanolytic, and lignolytic activity through qualitative evaluation in culture medium containing a specific corresponding carbon source. Carboxymethyl cellulase (CMCase) activity was quantified by the reducing sugar method. In the avicel and xilan degradation test, the enzyme activity (EA) at 48 h was significantly higher other periods (P < 0.05). Intra- and inter-specific differences in EA were verified, and high levels of phenoloxidases, which are crucial for lignin degradation, were observed in 28.9% of the isolates. Aspergillus terreus showed significantly higher EA for avicelase (3.96 ±1.77) and xylanase (3.13 ±.091) than the other Aspergillus species at 48 h of incubation. Isolates AT13 and AF69 showed the highest CMCase specific activity (54.84 and 33.03 U mg-1 protein, respectively). Selected Aspergillus spp. isolates produced remarkable levels of enzymes involved in vegetal cell wall degradation, suggesting their potential as antimicrobial additives or probiotics in ruminant diets.
RESUMOAvaliou-se o desempenho de tilápias-do-nilo alimentadas com farelo da casca de pequi (Caryocar brasiliense). Foram utilizados 200 alevinos, com idade de 37 dias e peso corporal médio de 0,63±0,25g, distribuídos em delineamento inteiramente ao acaso, com quatro tratamentos zero, 20, 40 e 60% de substituição de ração comercial por farelo da casca de pequi e cinco repetições representadas por caixas de cloreto de polivinila com capacidade para 130L, contendo 10 peixes cada, totalizando 20 unidades experimentais. As características de desempenho avaliadas foram consumo de ração, peso corporal, ganho de peso, conversão alimentar, comprimento total e viabilidade criatória. A conversão alimentar 1,96 ; 2,21 ; 2,63 ; 3,12 piorou linearmente com a inclusão do farelo de casca da pequi, enquanto as demais variáveis de desempenho não foram influenciadas pelos tratamentos. Conclui-se que a inclusão do farelo da casca de pequi na ração piora a conversão alimentar, sem alterar as demais variáveis de desempenho. BackgroundBrucella ovis infection is one of the leading causes of sub fertility and infertility in ovine, been characterized mainly by epididymitis, orchitis and testicular atrophy in rams. This study aimed to determine the frequency of B. ovis positivity in rams and goats flocks in the State of Minas Gerais, Brazil, by agarose gel immunodiffusion (AGID), ELISA, Rose Bengal, PCR and bacteriological isolation as diagnostic tools.FindingsSerum and urine samples were collected from properties with sheep or goat flocks, or from properties with mixed flock. Out of 50 sheep flocks, 6 % (3/50) were seropositive by AGID while 4 % (2/50) were positive by urine PCR for B. ovis. Out of five goat farms, 20 % (1/5) were seropositive for B. ovis by AGID. Mixed flock farms had 11.1 % (2/18) of positivity by AGID. By ELISA, 19.5 % (8/41) of sheep properties and 61.1 % (11/18) of the properties with mixed flocks were positive for B. ovis. No samples were positive in the test of Rose Bengal, ruling out exposure to smooth LPS Brucella species (particularly Brucella melitensis) and indicating that the positive in the ELISA was associated with Brucella spp. LPS rough (presumably B. ovis). No urine sample from sheep or goat was positive by bacteriological isolation.ConclusionsOur results demonstrate serologic or molecular evidence of B. ovis infection in several rams and billy goats from meso-regions of the State of Minas Gerais, Brazil. Also, this study report the indirect ELISA as an important tool for the diagnosis of B. ovis infection, as indirect ELISA in this study demonstrated to be the most sensitive diagnostic method adopted.
RESUMO Objetivou-se, com a presente pesquisa, avaliar a viabilidade de Aspergillus spp. com potencial probiótico durante o armazenamento e diante da pressão da microbiota autóctone, bem como a tolerância aos principais ácidos graxos da fermentação ruminal. Verificou-se também a inocuidade micotoxicológica desses isolados fúngicos cultivados em meio de cultivo sólido. Foram avaliados 20 isolados de Aspergillus spp., provenientes do trato gastrointestinal de bovinos criados em Urochloa decumbens lignificada. Esses fungos foram identificados por análise de sequências do DNAr e foram selecionados por apresentarem expressivo potencial celulolítico. O método vapor de amônia foi utilizado para detecção de cepas produtoras de micotoxinas. Os isolados foram avaliados quanto à viabilidade de crescimento em fluido ruminal por até 96 horas e estocagem em condições ambientais. Observou-se que os fungos avaliados não produziram aflatoxinas e que 95% dos isolados apresentaram resistência aos ácidos ruminais. Dois isolados, selecionados a partir das análises anteriores, apresentaram viabilidade sob a pressão da microbiota autóctone e de metabólitos do ecossistema ruminal e permanecem viáveis por, no mínimo, dois anos. Conclui-se que os isolados do gênero Aspergillus selecionados nesta pesquisa apresentam características fisiológicas para serem utilizados com aditivos microbianos ou probióticos para o ambiente ruminal.
A pecuária é considerada umas das atividades agrícolas mais importantes do mundo, com cerca de 2,3 bilhões de beneficiários em todo o planeta. Os danos ambientais associados à agropecuária, como a degradação do solo, a contaminação por resíduos de agrotóxicos, a poluição da água e a redução da biodiversidade passam a prevalecer à medida que cresce a demanda por alimentos. De acordo com a extensão territorial, a pecuária é considera uma das atividades mais impactantes ao meio ambiente. Esse fato se deve ao grande índice de animais no planeta e a falta de manejo racional das pastagens, criando assim um esgotamento no solo. A emissão de gases do efeito estufa (GEE) é um fator que vem sendo bem discutido atualmente, pois diferente dos países desenvolvidos que grande parte da emissão provém do setor energético, no Brasil, a maior parte desta emissão provém da pecuária, em especial da criação de gado de corte. Em função de relatos apontados observa-se crescente busca por tecnologias e alternativas que permitam o desenvolvimento da pecuária de forma sustentável, com altos índices de produção e, redução dos danos causados ao meio ambiente. Observa-se a partir dos relatos da literatura científica que a produção sustentável tem sido uma realidade, as pesquisas apontam que os métodos mostrados nessa revisão são eficazes para alcançar a produção sustentável na bovinocultura. Cada dia mais os produtores estão buscando novas técnicas para obter maior produção de carne e leite sem danos ao meio ambiente e que não ofereçam um custo elevado ao setor. A adoção desses métodos é uma ótima maneira de alcançar essa produção. Objetivou-se com a seguinte revisão abordar os mecanismos disponíveis para a produção sustentável de bovinos de corte, bem como os resultados encontrados na literatura científica sobre a utilização destes.
This study aimed to assess the histomorphometry of the intestinal portions (duodenum and jejunum) in broilers supplemented with probiotic, and histological parameters of their immune system components. Rhizomucor spp. originated from samples of the intestinal content in Cobb 500 chickens was evaluated; these fungi were submitted to performance assay and selected based on in vitro probiotic characteristics. Complete randomized design was used (3 treatments × 7 replicates; 18 birds per experimental unit). One bird at 21 and 42 days of age was sacrificed in each unit and the organs were sampled. In all samples, fixation was conducted followed in order by dehydration and diaphanization; analyses were conducted by light microscopy and histomorphometry, and search was performed for spots indicating the lesion's presence. At 21 days, the duodenum villus height was higher in the group receiving Rhizomucor spp. probiotic, and the jejunum villus height was significantly increased in both the control and Rhizomucor spp. probiotic groups. At 42 days, difference of the duodenum crypt depth was observed in the group receiving commercial probiotic. With regard to the tissue changes of the intestines, spleen, and bursa, no histological alterations were observed at 21 days in all birds (100%) receiving Rhizomucor spp. probiotic, while alteration of the bursa was observed at 42 days in two animals. Inclusion of probiotic Rhizomucor spp. in the diet of broilers promoted an increase in the height of the villi after 21 days, and any influence of the probiotic was resolved after 42 days of age.
The objective of this review was to survey the main microbial species producing enzymes involved in the degradation of compounds such as cellulose, hemicellulose and lignin. Most of the microorganisms isolated from soil, waste and compost material are capable of producing a broad spectrum of degrading enzymes from the plant cell wall. This enzymatic activity is essential for the fermentation of carbohydrates in the ruminal environment, since ruminants are not able to degrade these polymers, thus establishing a symbiosis relationship between these animals and the autochthonous microbiota of the gastrointestinal tract. However, microbial enzymes have not only been receiving attention in animal nutrition, more and more these have been explored by the industry as biotechnological possibilities, such as the production of ethanol. It is observed a microbial diversity in the rumen, and variations in the production of enzymes. Further studies are needed to elucidate the mechanisms of enzyme modulation in the rumen of production animals, in order to obtain an increase in zootechnical indexes and a higher digestibility of the fiber
