RESUMO As condições de estresse biótico e abiótico impostas às plantas induzem a superprodução de espécies reativas de oxigênio (ROS), podendo causar danos às estruturas celulares e mesmo acarretar a morte da planta. As respostas bioquímicas INTRODUÇÃOO oxigênio molecular (O 2 ) surgiu na atmosfera terrestre há bilhões de anos e sua presença permite que os organismos aeróbios o utilizem como aceptor de elétron terminal durante a respiração celular, que proporciona um rendimento de energia superior ao da fermentação. Embora necessário para o desempenho das funções celulares, o O 2 leva, inevitavelmente, à formação de espécies reativas de oxigênio (ROS, do inglês reactive oxygen species) em eventos metabólicos que ocorrem, principalmente, nas mitocôndrias, cloroplastos e peroxissomos (BHATTACHARJEE, 2010;KARUPPANAPANDIAN et al., 2011).As plantas desenvolveram mecanismos de defesa enzimáticos e não enzimáticos capazes de neutralizar a citotoxidade das ROS. O sistema celular de defesa antioxidante começa com uma cascata enzimática, mas envolve também componentes não enzimáticos, dentre os quais se destacam o ascorbato (AsA), a glutationa (GSH), o β-caroteno e o -tocoferol. Tais antioxidantes podem evitar a formação de radicais livres, sequestrá-los ou promover sua degradação, prevenindo a ocorrência de danos às células das plantas (SERKEDJIEVA, 2011). -In the northeastern region of Brazil, saline soils are constraints to banana production, becoming necessary to understand the mechanisms of salt tolerance. two bananas genotypes, tap Maeo, tolerant, and Berlin, sensitive, were subjected to treatment with 50 mol m -3 Nacl or without salt. this study evaluated the effects of salt on the following physiological aspects: leaf area, content and distribution of Na + , membrane integrity, proton atPase activity. Besides, a search for differentially expressed genes was performed using the Differential Display technique. tap Maeo genotype showed the smallest reduction in leaf area, smaller accumulation of Na + and malondialdehyde (MDa), and higher activity of proton atPase activity. two sequences differentially expressed in the tolerant genotype, (Musa 07, Musa 23), shared a high degree of identity with the amino acid sequences of the genes SOS1 and SOS2, respectively. the clone Musa 10 was highly similar to amino acid sequence of the ascorbate peroxidase gene, and Musa 26, encodes the enzyme betaine aldehyde dehydrogenase. These significant biological markers indicate that salinity tolerance in banana involves at least two simultaneous mechanisms: the activation of the SOS pathway, increasing the extrusion of Na + , and the activation of antioxidative system, increasing the synthesis of aPX and betaine aldehyde dehydrogenase enzyme. MECANISMOS DE TOLERÂNCIA À SALINIDADE EM BANANEIRA : ASPECTOS FISIOLÓGICOS, BIOQUÍMICOS E MOLECULARESRESUMO -Na região Nordeste do Brasil, solos salinos são restritivos à produção de banana, tornandose necessário compreender os mecanismos de tolerância de sal. Dois genótipos de banana, tap Maeo, tolerante, e Berlim, sensível, foram submetidos ao tratamento com 50 mol m-3 Nacl ou sem sal. este estudo avaliou os efeitos do sal sobre os seguintes aspectos fisiológicos: área foliar, conteúdo e distribuição de Na + , integridade da membrana, atividade da atPase. além disso, uma busca por genes diferencialmente expressos foi realizada usando a técnica Differential Display. O genótipo tap Maeo apresentou a menor redução na área foliar, menor acúmulo de Na + e malondialdeído (MDa) bem como maior atividade da H + atPase. Duas sequências diferencialmente expressas no genótipo tolerante (Musa 07, Musa 23) compartilham alto grau de identidade com as sequências de aminoácidos dos genes SOS1 e SOS2, respectivamente. O clone Musa 10 é muito semelhante à sequência de aminoácidos do gene da peroxidase do ascorbato, e o Musa 26 codifica a enzima aldeído betaína desidrogenase. -The study was conducted with shoot tip explants of neem (Azadirachta indica A. Juss) to identify a viable regenerative process. Shoot tips were obtained from neem embryos cultured alternatingly in DKW medium supplemented with BAP and medium without hormones. Initial shoot development was influenced by cotyledon presence. Basal callus, excised from in vitro stem base, also presented organogenic potential. In some cases, plant lines, obtained from each seed, presented different characteristics. The most common characteristic observed in vitro was callus formation at the stem base. However, the rarest characteristics were stem callus formation and leaf senescence. The regenerated shoot tips were further subculture and rooted on a medium supplemented with IBA so that complete plants could be obtained. The rooted plants were transplanted to a greenhouse and successfully acclimatized. No significant differences in in vivo development were observed between neem plants from callus and from shoot tip propagation.Keywords: Micropropagation; In vitro developmental characteristics; Callus organogenesis. In view of the ecological, social, and economic importance of Myracrodruon urundeuva Allemão, the objective of this study was to investigate the strategies of this species under drought during its initial phase of development. Two-monthold plants were cultivated under continuous irrigation or no irrigation for 20 d. After this period, the water-stressed plants were rehydrated for 20 d. Physiological, biochemical, and anatomical variables were evaluated on 20 th and 40 th day. Water deficit (25 and 85 % leaf relative content) caused senescence followed by leaf abscission. Growth in height was negatively affected by water deficit (37 % reduction). A decrease in the thickness of the mesophyll was accompanied by a decrease of the total chlorophyll content. Water deficit affected saccharide metabolism and altered cellular component dynamics. Enzyme activities were higher during the rehydration period than during the water stress. There was no increase in lipid peroxidation in plants subjected to water deficit. A reduction in the stomatal opening during water stress was a strategy of reducing water loss through transpiration.
RESUMO Avaliou-se a capacidade de adaptação à salinidade de mudas de cana-de-açúcar da variedade RB98710 após pré-condicionamento (priming) in vitro com cloreto de sódio (NaCl). O experimento foi conduzido no período de maio a outubro de 2013. Plantas foram micropropagadas em biorreatores de imersão temporária, com tratamentos de priming caracterizados pela adição de 25 mM de NaCl, durante 0, 12, 24 e 36 horas. Concluídos os períodos de priming, as plantas foram cultivadas em meio de enraizamento sem NaCl, aclimatizadas e submetidas a tratamento salino gradativo: S0 = sem NaCl; S1 = regas com 20→40→60 mM de NaCl acrescido à solução nutritiva, durante 30 dias. Procedeu-se à supressão do NaCl e avaliaram-se as plantas 15 dias depois. As variáveis foram: matéria seca da parte aérea (MSPA) e radicular (MSR), número de perfilhos, estimativa de clorofila (Chl), conteúdo hídrico relativo e dano de membrana. As plantas pré-condicionadas não reduziram a MSPA quando submetidas ao NaCl. O efeito do estresse salino sobre a MSR foi anulado nas plantas submetidas ao priming por 24 h. O priming favoreceu o perfilhamento das plantas dos tratamentos por 24 ou 36 h. A recuperação após o estresse, avaliado 15 dias após a supressão do NaCl, não diferiu entre plantas submetidas ou não ao priming. De acordo com os resultados, o priming utilizado na variedade RB98710, pode favorecer a tolerância à salinidade.

Bioemulsifier is bioactive molecules produced by different microorganisms with reducing power and surface and interfacial tension. Among the microorganisms producing this molecule is yeast, which can produce different bioemulsifiers in different substrates. Undoubtedly, this biomolecule has excellent potential for industrial applications, but high production costs are the biggest problem in production. Aiming at cost reduction the present study using crude residual glycerol for biosurfactant production by Yarrowia lipolytica. Then isolates were grown in residual glycerol compound medium, rotating 200 rpm at 28ºC for 48 hours. Bioemulsifier production was observed by analysis of dry biomass, pH, surface tension and emulsification index. The results indicated that the emulsion produced from biosurfactant using glycerol as a carbon source by Y. lipolytica has the potential for bioemulsifier production. All isolates obtained similar results for all analyzes, indicating that this species has a linear production among the isolates. Biomass reached 10.08 ± 0.62 g.L-1 , there was a sharp drop in pH reaching 4.6, surface tension averaged 41.7 mN.m-1 and emulsification index reached 56%. The isolates tested show potential for bioemulsifier production using glycerol as an unconventional carbon source.
