Four populations of Astyanax aff. fasciatus of the upper rio Tibagi (municipal district of Ponta Grossa, Paraná State, Brazil), had their karyotypes and morphometry analyzed. The cytogenetic data show the occurrence of distinct karyotypes (cytotypes), here named cytotype A, with 2n=48 chromosomes (6m+18sm+14st+10a), cytotype B, with 2n=50 chromosomes (8m+18sm+14st+10a) and cytotype C, with 2n=50 chromosomes (8m+18sm+14st+10a). The distribution pattern of the constitutive heterochromatin was very similar between cytotypes A and B, but diverged in relation to cytotype C. Distinct cytotypes may occur in sympatry in the upper rio Tibagi region, with the exception of the Furna 2 sample, which presents cytotype A exclusively. In addition, a specimen with 2n=49 chromosomes (7m+18sm+14st+10a) was also found and, by the characteristics presented, may be a consequence of a rare hybridization event between cytotypes A and B. The morphometric analyses of canonical variates indicate a consistent isolation of the Furna 2 sample, while the other samples seem to be superimposed, indicating a possible gene flow or even a recent isolation event. This model points to a probable complex of cryptic species in the studied region.Quatro populações de Astyanax aff. fasciatus do alto rio Tibagi (município de Ponta Grossa, Paraná, Brasil) foram citogeneticamente e morfometricamente analisadas. Os dados citogenéticos mostram a ocorrência de distintos cariótipos (citótipos), aqui nomeados citótipo A, com 2n=48 (6m+18sm+14st+10a), citótipo B, com 2n=50 (8m+18sm+14st+10a) e citótipo C, com 2n=50 cromossomos (8m+18sm+14st+10a). O padrão de distribuição da heterocromatina constitutiva foi muito similar entre os citótipos A e B, mas mostrou-se divergente em relação ao citótipo C. Citótipos distintos podem ocorrer em simpatria na região do alto rio Tibagi, com exceção da amostra da Furna 2, a qual apresenta somente o citótipo A exclusivamente. Além disso, um exemplar com 2n=49 cromoossomos (7m+18sm+14st+10a) foi também encontrado e, pelas características apresentadas, pode ser uma conseqüência de um raro evento de hibridização entre os citótipos A e B. As análises morfométricas de variáveis canônicas indicam um isolamento consistente da amostra da Furna 2 enquanto as demais amostras analisadas se apresentam sobrepostas indicando um possível fluxo gênico ou evento de isolamento recente. Este modelo aponta para um complexo de espécies crípticas na região estudada.

Cytogenetic studies show that there is great karyotypic diversity in order Testudines (2n = 26–68), and that this may be mainly attributed to the presence/absence of microchromosomes. Members of the Podocnemididae family have the smallest diploid numbers of this order (2n = 26–28), which may be a derived condition of the group. Diverse studies suggest that repetitive-DNA-rich sites generally act as hotspots for double-strand breaks and chromosomal reorganization. In this context, we used fluorescent in situ hybridization (FISH) to map telomeric sequences (TTAGGG)n, 45S rDNA, and the genes encoding histones H1 and H3 in two species of genus Podocnemis. We also observed conservation of the 45S rDNA and H1 histone sequences (probable case of conserved synteny), but multiple conserved and non-conserved clusters of H3 genes, which colocalized with the interstitial telomeric sequences in the Podocnemis genome. Our results suggest that fusions have occurred between macro and microchromosomes or between microchromosomes, leading to the observed reduction in diploid number in the family Podocnemididae.

Cytogenetic data are presented for Astyanax altiparanae populations from three Brazilian hydrographic systems. The chromosomal data obtained in A. altiparanae support the hypothesis of diploid number conservation. However, small differences in the karyotype formula and number of nucleolar organizer regions were observed in these populations. The apparent karyotypical similarity among the studied populations strongly suggests a close relationship among them with some chromosomal divergences due to gene flow restriction.

ABSTRACT. We examined chromosomes of three species of the genus Hypostomus, in order to contribute to the understanding of the karyotype evolution of this group. Specimens of H. ancistroides and H. nigromaculatus displayed differences in karyotype formulas, distribution and location of heterochromatin and nucleolus organizer regions when compared to other populations of the same species. We made the first cytogenetic characterization of H. tapijara, an endemic species in the Ribeira de Iguape River. These specimens had 2n = 66 chromosomes, while H. ancistroides showed 2n = 68 and H. nigromaculatus 2n = 76 chromosomes. Physical mapping of 18S and 5S rDNA sites of the three species showed simple, multiple and syntenic clusters. Synteny of ribosomal sites was found in H. ancistroides and H. tapijara, and an interspersed pattern between these sites in all chromosomes bearing the synteny was observed. We conclude that the genus Hypostomus has a high chromosome complexity that is accompanied by great morphological variation. It is evident that this group comprises an interesting model for understanding the chromosome evolution of Neotropical ichthyofauna.

Great efforts have been made to preserve manatees. Recently, a hybrid zone was described between Trichechus inunguis (TIN) and the Trichechus manatus manatus (TMM) in the Amazon estuary. Cytogenetic data on these sirenians are limited, despite being fundamental to understanding the hybridization/introgression dynamics and genomic organization in Trichechus. We analyzed the karyotype of TMM, TIN, and two hybrid specimens (“Poque” and “Vitor”) by classical and molecular cytogenetics. G-band analysis revealed that TMM (2n = 48) and TIN (2n = 56) diverge by at least six Robertsonian translocations and a pericentric inversion. Hybrids had 2n = 50, however, with Autosomal Fundamental Number (FNA) = 88 in “Poque” and FNA = 74 in “Vitor”, and chromosomal distinct pairs in heterozygous; additionally, “Vitor” exhibited heteromorphisms and chromosomes whose pairs could not be determined. The U2 snDNA and Histone H3 multi genes are distributed in small clusters along TIN and TMM chromosomes and have transposable Keno and Helitron elements (TEs) in their sequences. The different karyotypes observed among manatee hybrids may indicate that they represent different generations formed by crossing between fertile hybrids and TIN. On the other hand, it is also possible that all hybrids recorded represent F1 and the observed karyotype differences must result from mechanisms of elimination.
