RESUMO:Considerando-se a qualidade dos fi toterápicos, é importante salientar que a preocupação com esta questão inclui rigoroso acompanhamento das diferentes etapas do desenvolvimento e produção destes produtos, desde a coleta do vegetal até a disponibilidade do produto fi nal. Neste trabalho, foram realizados o controle da qualidade, o potencial antioxidante como também ensaios biológicos in vitro do fruto da goiabeira (Psidium guajava L.), para o desenvolvimento de uma formulação fi tocosmética. Os resultados obtidos mostraram que o fruto apresenta taninos e fl avonóides, bem como atividades antioxidante e antimicrobiana. A análise microbiológica não apresentou crescimento de patógenos na formulação desenvolvida entre os outros testes realizados. Destaca-se a importância do estabelecimento do controle da qualidade para as plantas, a fi m de que sejam utilizadas para o desenvolvimento de uma formulação fi tocosmética segura, efi caz e com qualidade.Unitermos: Psidium guajava, Myrtaceae, goiaba, atividade antioxidante, fi tocosmético, atividade antimicrobiana.ABSTRACT: "Phytochemical study of guava (Psidium guajava L.) with potential antioxidant activity aiming at developing a phytocosmetic formulation". In this work the quality control, the antioxidant potential were performed as also biological assays in vitro of guava fruit (P. guajava L.), for the development of a phytocosmetic formulation. The results had shown that the fruit presents tannins and fl avonoids, as well as antioxidant and antimicrobial activities. The microbiological analysis did not present growth of pathogens. The importance of the establishment of the quality control for the plants is relevant, so that they are used for the development of a phytocosmetic formulation considered safe, effi cient and with quality.

Para o controle físico-químico e microbiológico utilizaram-se metodologias farmacopéicas e não farmacopeicas. Os resultados obtidos mostraram que o fruto apresenta um teor de taninos de 4,2% e a análise fi toquímica preliminar dos frutos de S. cumini evidenciou taninos, fl avonóides, antocianidinas, iridóides, alcalóides e heterosídeos fenólicos simples. A análise microbiológica não apresentou crescimento de patógenos entre os outros testes realizados. In this work the quality control of the Syzygium cumini (L.) Skeels fruit, popularly known as jambolão, was carried out. Pharmacopoeic and no pharmacopoeic methodologies were employed to physico-chemical and microbiological quality control. The obtained results showed that the fruits presents a content of tannins of 4.2%, the preliminary phytochemical analysis of the S. cumini fruits evidenced tannins, fl avonoids, antocianidin, iridoids, alkaloids and heterosides simple phenols. Endopleura uchi is a typical Amazonian tree and its bark is popularly employed in the preparation of teas against myomas, arthritis, influenza, diarrhea and cancer. In this study, the antioxidant activity, cytotoxicity and antimicrobial activity of five different extracts of the bark, selected by their total tannin content, were assessed. The potential antioxidant activity of the extracts was determined by 2.2-diphenyl-1-picrylhydrazyl radical scavenging assay and the values found were very similar among the extracts and to the standards antioxidants used in the tests. Cytotoxicity analysis in mammalian cells indicated that all the tested extracts exhibited IC 50 values higher than the highest concentration used, showing that they do not present a risk when consumed under these conditions. Extract tested against five bacterial strains and one yeast strain did not show satisfactory growth inhibitory activity, and even the extracts that showed some antimicrobial activity were not OPEN ACCESS Int. J. Mol. Sci. 2011, 12 2758 effective at any dilution to determine the minimum inhibitory concentration. The results may serve as a reference for subsequent works, since such reference values described in the literature for the bark of E. uchi.

The aim of this study was to standardize the extractive solution of Syzygium cumini. The extractive solution was selected through a full factorial design, considering the extraction method (maceration-5-days, turbo extraction, percolation), extractor solvent (ethanol 50, 70, 96%) and plant material particle size 0.302 mm into a plant:solvent ratio (1:10), using the dried residue and antimicrobial activity determination as evaluation criteria. The tests were performed in triplicate. It appeared that the percolation is the best procedure extractive, and ethanol mixture: water (50:50) the best solvent. ANOVA analysis showed the importance of these parameters.Keywords: Syzygium cumini; extraction method; factorial design. INTRODUÇÃOO desenvolvimento de fitoterápicos inclui várias etapas e processos interdisciplinares e multidisciplinares. As áreas de conhecimento envolvidas vão desde a antropologia botânica, botânica, agronomia, ecologia, química, fitoquímica, farmacologia, toxicologia, biotecnologia, química orgânica, tecnologia farmacêutica até ensaios clínicos. 1Dados da literatura revelam que é muito mais provável encontrar atividade biológica em plantas orientadas pelo seu uso na medicina popular (etnofarmacologia) do que em plantas escolhidas ao acaso.Para o conhecimento das substâncias ativas de plantas, um dos aspectos que deve ser observado consiste na preparação de extratos vegetais, visando o isolamento de seus constituintes químicos ativos. Um dos métodos que pode ser considerado adequado para a análise químico-farmacológica é a preparação de extratos hidroetanólicos 50% por maceração. Este extrato é análogo às tinturas realizadas na cultura popular, onde se misturam as partes ativas das plantas com bebidas alcoólicas.
The antimicrobial efficacy of three preservatives used in cosmetic formulations was evaluated. Phenova® and imidazolidinyl urea inhibited the growth of Bacillus subtilis when added to leaf extract of Achillea millefolium L., whereas 0.2% Nipagin®/ Nipasol® in propylene glycol did not.
A precise, accurate and low cost spectrophotometric method was developed and validated for routine determination of total polyphenols, as pyrogallic acid equivalents, from the percolated and lyophilized extract of Syzygium cumini (L.) Skeels fruits. Validation was assessed experimentally and data were rigorously treated by statistical analysis. Analytical parameters were: linearity, interval (range), precision and recovery/accuracy, limit of detection (LOD, µg mL -1) and limit of quantification (LOQ, µg mL -1 ). The visible spectrophotometric method presented linearity (r 2 = 0.9979 ± 0.0010) over the concentration range 0.25-7.5 µg mL -1 of standard pyrogallic acid, precision < 2.918171%, recovery/accuracy ranging from 96.228693 to 107.17701%, LOD = 0.21 µg mL -1 and LOQ = 0.64 µg mL -1 .Uniterms: Total polyphenols. O extrato dos frutos de Syzygium cumini (L.) Skeels foi preparado empregando-se o método de percolação com posterior liofilização. A validação foi executada experimentalmente e os dados foram submetidos à análise estatística. Os parâmetros analíticos considerados foram: linearidade, intervalos de precisão e de recuperação, limite de detecção (LD, μg.mL -1 ) e o limite de quantificação (LQ, μg.mL -1 ). O método espectrofotométrico apresentou linearidade (r 2 = 0,9979 + 0,0010) ao longo do intervalo de concentração de 0,25-7,5 μg mL -1 de ácido pirogálico, padrão de precisão menor do que 2,918171%; recuperação/ precisão entre 96,228693 a 107,17701%, e LD = 0,21 μg-mL -1 e LOQ = 0,64 μg mL -1 .
A three-dimensional (3D) lung aggregate model based on sodium alginate scaffolds was developed to study the interactions between Paracoccidioides brasiliensis (Pb) and lung epithelial cells. The suitability of the 3D aggregate as an infection model was examined using cell viability (cytotoxicity), metabolic activity, and proliferation assays. Several studies exemplify the similarity between 3D cell cultures and living organisms, which can generate complementary data due to the greater complexity observed in these designed models, compared to 2D cell cultures. A 3D cell culture system of human A549 lung cell line plus sodium alginate was used to create the scaffolds that were infected with Pb18. Our results showed low cytotoxicity, evidence of increased cell density (indicative of cell proliferation), and the maintenance of cell viability for seven days. The confocal analysis revealed viable yeast within the 3D scaffold, as demonstrated in the solid BHI Agar medium cultivation. Moreover, when ECM proteins were added to the alginate scaffolds, the number of retrieved fungi was significantly higher. Our results highlight that this 3D model may be promising for in vitro studies of host–pathogen interactions.
