O mel é um produto naturalmente produzido por abelhas e, devido a sua composição, possui sabor característico e alto valor nutritivo. Seu uso é relatado desde a antiguidade e, a cada dia ganha novos mercados consumidores. A exigência por produtos de qualidade disponíveis ao consumo humano reflete a necessidade de uma maior fiscalização durante o processo de produção. O objetivo desta pesquisa foi avaliar a qualidade físico-química e microbiológica de 22 amostras de méis com registro de inspeção e de comércio informal, de diferentes municípios da região Sul do Brasil. Foram analisados os parâmetros: acidez total, pH, hidroximetilfurfural, sólidos insolúveis, atividade diastásica, reações de Fiehe, Lugol, Lund e a presença de bolores e leveduras. Quanto às características físico-químicas, 54,54% das amostras apresentaram alguma inconformidade com os parâmetros estabelecidos pela legislação. Apesar da legislação não fazer exigências quanto à realização de análises microbiológicas, 45,45% das amostras apresentaram a presença de bolor e levedura.PALAVRAS-CHAVE: segurança alimentar, inspeção de produtos de origem animal, mercado informal.
This study aimed to evaluate the efficiency of stretching in the reduction of pathogens when compared to milk pasteurization, the official method to ensure safe cheese production. Whole buffalo milk was contaminated with Mycobacterium fortuitum, Listeria monocytogenes, Salmonella typhimurium, and Staphylococcus aureus. Part of the milk was used in mozzarella production and the other part was submitted to holder pasteurization. Pathogens were quantified before and after thermal processing (mozzarella stretching and milk pasteurization). Parameters for milk pasteurization were established a long time ago, considering the thermal resistance of Mycobacterium bovis, and the systematic adoption of this process has drastically reduced the incidence of human tuberculosis caused by this pathogen. However, more recently, molecular methods have allowed the identification of genetic variations in this bacterium that may lead to greater thermal resistance. The aim of this study was to investigate whether genetic variation leads to variation in the death pattern of this bacterium during the milk pasteurization process. Samples of UHT (ultrahigh temperature)-treated whole milk were artificially contaminated with four different Mycobacterium bovis spoligotypes and were subjected to pasteurization by low-temperature long-time (LTLT) and high-temperature short-time (HTST) treatments. The M. bovis spoligotypes were quantified (Colony Forming Unit per milliliter of milk) before and during the thermal process. The decay of the pathogen was quantified by calculating the difference between the measurements at the beginning and at the end of the thermal treatment. The data demonstrated that the LTLT and HTST pasteurization processes considerably reduced the M. bovis load in the milk; however, the bacterium was not eliminated. There was no difference in the thermal resistance of the spoligotypes tested or in the efficiency of pasteurization processes (LTLT versus HTST). However, heating phase was more effective in reducing the M. bovis load than the target temperature maintenance phase. Key words: Batch pasteurization. Bovine tuberculosis. Flow pasteurization. ResumoOs parâmetros para a pasteurização do leite foram estabelecidos há muito tempo, considerando a resistência térmica de Mycobacterium bovis e a adoção sistemática deste processo reduziu drasticamente a incidência de tuberculose humana causada por este patógeno. Entretanto, mais recentemente, métodos moleculares permitiram a identificação de variações genéticas nesta bactéria que podem levar a uma maior resistência térmica. O objetivo deste estudo foi investigar se a variação genética apresenta modificação no padrão de morte da bactéria durante o processo de pasteurização do leite. Amostras de leite tratadas por UHT (ultra-alta temperatura) integral foram contaminadas com quatro diferentes espoligotipos de Mycobacterium bovis e foram submetidos ao tratamento por temperatura baixa e tempo longo (LTLT) e temperatura alta e tempo curto (HTST). Os espoligotipos M. bovis foram quantificados (Unidades Formadoras de colônias por mililitro de leite) antes e durante o processo térmico. O objetivo desta pesquisa foi identificar os hábitos de consumo, preocupação quanto à segurança alimentar e percepção do risco alimentar nos hábitos de consumo da carne bovina em consumidores do município de Jataí, - GO. Foram entrevistadas 100 pessoas, sendo a maioria de mulheres (68%), com escolaridade entre fundamental completo e ensino médio completo (76%), e renda familiar de 1 a 2 salários mínimos (56%). A carne bovina é consumida por 98% dos entrevistados sendo que 73% possuem o hábito de consumir carne bovina nas refeições mais de quatro vezes durante a semana. A aparência foi o atributo mais relevante para a compra da carne bovina, assim como a higiene do funcionário e do local. Os entrevistados também relataram uma preocupação extrema com relação à carne bovina quanto ao controle higiênico do armazenamento e comercialização, presença de bactérias que fazem mal à saúde, resíduos de drogas veterinárias e doenças transmitidas pelo animal. O estudo da percepção dos consumidores em relação aos atributos e os riscos associados ao consumo de carne bovina torna-se uma opção estratégica no fomento de programas voltados à promoção da saúde pública e também ao desenvolvimento de outros estudos sobre o assunto no Brasil.
Decline in Mycobacterium bovis AbstractBrazilian legislation allows the manufacture of raw milk cheese with a maturation exceeding 60 days at room temperature above 5°C, but there is a lack of solid scientific evidence on the efficacy of this maturation process in inactivating important pathogens that may be present in milk, such as Mycobacterium bovis and Brucella abortus. Thus, the objectives of this study were to produce parmesantype cheese experimentally contaminated with M. bovis and B. abortus and evaluate the survival of these pathogens along 2-month maturation. Parmesan-type cheese was manufactured in the laboratory using whole pasteurized milk with or without inoculation with M. bovis (SB1033) or B. abortus (1119-3) and matured at 18°C for up to 63 days. M. bovis was inoculated in Stonebrink-Leslie medium supplemented with antibiotics and incubated at 37°C for 45 days, and B. abortus was incubated in Farrel medium at 36°C for 3 days. The average D 18°C value, weighted by variance, was 37.5 ± 5.3 days for M. bovis and 5.9 ± 0.7 days for B. abortus. The average physicochemical parameters in the cheese at the end of the study were as follows: pH = 4.89, water activity = 0.976, and moisture percentage = 43.1%. The pH might have contributed to the reduction in the population of B. abortus but seems not to have significantly influenced the population of M. bovis. We conclude that the duration of the maturation process influences the size of the surviving populations of M. bovis and B. abortus, and that the shortening of the maturation duration might not ensure a decline in pathogen levels to safe levels. Thus, complementary studies considering the effect of several other technological aspects on the survival of these pathogens are required, including the effect of the lactic acid bacterial population, salt content, and temperature of maturation. Assim, os objetivos desse estudo foram produzir o queijo tipo parmesão experimentalmente contaminado com M. bovis e B. abortus e avaliar a sobrevivência desses patógenos ao longo de 2 meses de maturação. ResumoEste estudo objetivou mostrar o efeito da gordura do leite caprino e bovino sobre a resistência térmica do Mycobacterium fortuitum. Amostras de leite foram divididas em duas porções, integral e desnatada, cada porção foi inoculada com M. fortuitum e então distribuída em tubos para a quantificação do agente durante a pasteurização lenta, em banho-maria. As amostras foram diluídas e semeadas em Lowenstein-Jensen (37 °C/5 dias), os resultados da contagem foram expressos em Log 10 UFC/mL. O tratamento térmico reduziu 4,4 Log 10 UFC/mL no leite caprino integral (2,8% de gordura), 4,9 Log 10 UFC/mL no leite caprino desnatado (0,3%), 3,9 Log 10 UFC/mL no leite bovino integral (5,9%) e 5,4 Log 10 UFC/mL no leite bovino desnatado (0,2%), sem diferença significante, possivelmente pelo baixo número de amostras. Os valores D 65 °C foram, respectivamente, 10,51 minutos, 8,61 minutos, 18,02 minutos e 7,82 minutos e o valor de R 2 baixo das equações da reta indicam que outros fatores, além dos que foram estudados, influenciam a morte térmica do agente. Os resultados sugerem uma tendência de influência da gordura do leite e também da espécie animal sobre a taxa de decaimento do M. fortuitum, sendo que a pasteurização foi menos eficaz sobre o M. fortuitum no leite bovino integral. AbstractThis study aimed to show the effect of goat and bovine milk fat on thermal resistance of Mycobacterium fortuitum. Milk samples were divided into two portions, whole and skimmed, each part was inoculated with M. fortuitum and then distributed in tubes for quantification of the agent during pasteurization, in a water bath. As samples were diluted and plated on Lowenstein-Jensen (37 °C/5 days), the count results were expressed as log 10 CFU/mL. The heat treatment reduced 4.4 log 10 CFU/mL for goat whole milk (2.8% fat), 4.9 log 10 CFU/mL for skim goat milk (0.3%), 3.9 log 10 CFU/ml for whole bovine milk (5.9%), and 5.4 log 10 CFU/mL for skim bovine milk (0.2%), without significant difference, possibly because of the low number of samples. Values of D 65 °C were, respectively, 10.51 minutes, 8.61 minutes, 18.02 minutes, and 7.82 minutes and the low R 2 value of the straight line equations indicated that other factors, in addition to the ones studied, influenced the heat death of the agent. The results suggest a trend of influence by fat milk, and by the animal species on the decay rate of M. fortuitum, and that pasteurization was less effective over M. fortuitum in whole bovine milk.
