Burns are lesions in which the thermal energy of the causative agent transfers heat to the surface of the body, causing superficial or deep damage to the skin with protein denaturation in cells and biochemical maladjustments, which delay and disrupt the cicatricial process, increasing the chances of functional and aesthetic sequelae. This study evaluates the influence of adipose tissue-derived stem cells (ADSCs) on burn healing in terms of the size of the cicatricial space and quantified measures of collagen deposition, inflammatory infiltrate, blood vessels, and lymphatic vessels. Initially, intra-abdominal adipose tissue was resected from a single donor Wistar rat that was not part of any of the subsequent groups to obtain ADSCs by isolation and cell culture. Burns were made in the left lateral abdominal region of Wistar rats by contact with a square ceramic paper with a 484 mm2 area heated to 100°C for 30 seconds. Intradermal ADSC transplantation was performed in two stages. The first was on the same day of the burn, when 3.2×106 ADSCs were transplanted shortly after the burned region cooled, while the second stage occurred four days later with the same number of ADSCs. The progress was evaluated by immunohistochemical methods and H&E, Masson’s trichrome, Picrosirius red, and Lyve-1 immunofluorescence staining. Despite the quantitative similarity of blood vessels and the inflammatory infiltrate observed by H&E, there were statistically significant differences between the groups on the fourteenth day of evolution. The group that received ADSCs showed a reduction in the scar tissue area, increased collagen type III deposition, and a quantifiable reduction in lymphatic vessels, so we conclude that ADSCs influence the healing of total thickness burns in rats.
RESUMOObjetivo: Avaliar a ação cicatrizante do extrato hidroalcoólico da Aroeira em bexigas de ratos. Métodos: Foram utilizados 40 ratos machos da linhagem Wistar, divididos em dois grupos de 20, denominados grupo Aroeira (GA) e grupo controle (GC). Todos foram submetidos à incisão abdominal mediana com cistotomia de 1 cm, seguida de cistorrafia em plano único com pontos separados de poliglactina 910 5-0 (Vicryl ® ). Após este procedimento, nos animais GA injetou-se 100mg por quilo de peso de extrato hidroalcoólico da Aroeira na cavidade peritoneal e nos GC injetou-se 1 ml por quilo de peso de solução salina a 0,9% . Cada grupo foi dividido em dois sub-grupos de 10 animais de acordo com a data da morte: sub-grupo A3 e C3, sacrificados no 3º dia pós-operatório e sub-grupo A7 e C7, sacrificados no 7º dia. A parede, a cavidade abdominal e a sutura da bexiga foram avaliadas macroscopicamente. Amostras de tecido da bexiga foram retiradas e analisadas histologicamente, utilizando a coloração de Hematoxilina-Eosina (HE) e tricrômio de Masson. Resultados: Na análise macroscópica observou-se infecção na incisão cirúrgica em três ratos do GC e em um do GA, e aderências peritoneais em 29 ratos do GC controle e 17 no GA. A avaliação microscópica mostrou processo inflamatório agudo mais severo no 3° (p=0,045) e no 7° dia (p=0,002). Evidenciou-se ainda diferença estatística nos parâmetros utilizados para a avaliação histológica da cicatrização da bexiga nas variáveis colagenização (p = 0.001), reepitelização (p = 0,046) e neoformação (p = 0) nos subgrupos GC e GA no 3º dia e na variável neoformação vascular (p=0,001) no subgrupo do 7º dia. Conclusão: O uso de extrato hidroalcoólico de Aroeira mostrou efeito cicatrizante favorável nas cistotomias em ratos. Descritores: Schinus terebinthifolius. Cicatrização de Feridas. Bexiga. Ratos ABSTRACT Purpose: To evaluate the hydroalcoholic Aroeira extract in the healing process of surgical incisions in the bladder of rats in an experimental comparative study. Methods: Forty male Wistar rats were used. All of them underwent a median abdominal incision, with an 1cm cistotomy, followed by one plan suture with separated stitches of poliglactine 910 5-0. The animals were then divided into two groups of 20 rats each, and named the Aroeira group (GA) and the control group (GC). In the first one, 80% hydro-alcoholic plant extract was injected in the peritoneal cavity, in a one dose of 100mg/kg. The second group, a 0.9% saline solution was injected in place of Aroeira extract. Each group was divided into two subgroups (SGA and SGC) of 10 animals. According to their scheduled death, they were named subgroups SGA3 and SGC3 when killed in the 3th day after surgery and subgroups SGA7 and SGC7 in the 7th day. Objectives: The aim of this work was to analyze the dominance patterns of the circulation of the human heart, the number of branches from the right coronary artery to the left ventricle, the number of branches from the left coronary artery to the right ventricle and the frequency and location of intercoronary anastomoses. Methods: Casts were made by the injection of colored acrylic resin in 25 hearts and subsequent corrosion using hydrochloric acid at the experimental surgery laboratory of FURB. Specimens with lesions or scars were discarded.Results: The hearts, from both men (17 -68%) and women (8 -32%), had a mean age of 40.2 (15 to 70) years old. Right dominance occurred in 18 (72%) subjects, with 1, 2, 3 and 4 branches leading to the left ventricle in 2, 14, 2 and 2 molds, respectively. Left dominance occurred in 5 (20%) with 1 branch leading to the right ventricle in 4 molds and 2 in one. Balanced circulation was observed in two molds (8%). There were significant differences between right and left dominance (á > 5%) and between right dominance and balanced circulation (á > 5%), however the same was not true between left dominance and balanced circulation (á < 5%). Conclusion:The most common form of coronary circulation is right dominance with an average of 2.16 branches leading to the left ventricle: when dominance is left, the average is 1.2 branches. PURPOSE: To verify the effect of highly concentrated platelet-rich plasma (hPRP) in the pathways of bone repair using non-critical defects in the calvaria of rabbits. METHODS: The hPRP was produced from collected venous blood of 21 rabbits. Four non-critical defects of 8 mm in diameter were created on the calvaria of each animal. The defects were all treated differently: autogenous particled bone (APB, group 1), autogenous particled bone associated with hPRP (APB + hPRP, group 2), isolated hPRP (group 3), and blood clot (control, group 4). Animals were submitted to euthanasia on the 2nd, 4th and 6th week postoperatively. Histological and histomorphometric analysis were carried through. RESULTS: After two weeks, groups 1 and 2 were in more advanced stage of repair than 3 and 4. At this period, comparing the groups 1 and 2, no significant differences were found between both, which also happened between the groups 3 and 4. However, after four and six weeks, the group 1 showed a more advanced stage of repair among all the other studied groups, while group 2 was in more advanced signs of bone repair than groups 3 and 4. Comparing groups 3 and 4, after four and six weeks, the least advanced stage of bone repair was found to be within group 3. CONCLUSION: The use of a highly concentrated PRP was considered prejudicial to the repair of non-critical defects in the calvaria of rabbits, either in the association of autogenous particled bone, when compared to autogenous particled bone alone, or in its isolated form, when compared to blood clot (control).
PURPOSE: To investigate the results of the healing process on surgical wounds in the back of Wistar rats using nanocristaline and ionic silver dressing. METHODS: Sixty rats Wistar were submitted to surgical wounds with punch of 8mm in diameter. In 30 animals (groups PN - nanocristaline and AD - control) two surgical wounds were done diametrically opposite on the upper back side. On the right side was used nanocristaline (PN) silver dressing and on the left side, distilled water dressing (AD). On the other group of 30 rats, only one wound was made with the punch, on the right side, and was used ionic silver dressing. So, the groups were divided into three subgroups, according to the day of death (7th, 14th and 21st day). In each of these days the wounds diameter were measured to evaluate the wound contraction. Microscopic data were analyzed using the H&E staining to verify the inflammatory process and neovascularization. The Masson trichrome staining was used to verify the fibrosis. RESULTS: Macroscopically only the subgroup of 21st day showed statistical significance; between the groups AD and PI inflammatory process appeared in the 7th day subgroup in 90% of the cases. In neovascularization there was statistical significance between the groups PN and AD in the subgroup of 7th day. Fibrosis did not show statistical significance in the studied groups. CONCLUSIONS: In relation to wound contraction, PN and PI groups showed better results than the AD group. In regard to histological analysis, H&E staining showed that there was presence of inflammation in all groups, and at the end, the control group (AD) on 7th day, was superior to PN and PI groups. In relationship to fibrosis, no differences were obtained among groups.
PURPOSE:To evaluate the usefulness of a knee osteoarthritis model through functional, radiological and microscopic changes of the synovial membrane. METHODS:Forty eight rats were divided randomly into two groups. The first received 0.9% saline in the joint and corresponded to the control group. The second was submitted to experimental osteoarthritis of the right knee induced by monosodium iodoacetate and corresponded to the osteoarthritis group. All animals were subjected to comparative tests of forced ambulation and joint movements, inability to articulate and tactile allodynia on day 1 post-experiment by forced ambulation (Roto-rod test), joint assessment of disability (weight bearing test) and assessment of tactile allodynia (Von Frey test). After inflammatory induction they were divided into four subgroups corresponding to the scheduled death in 7, 14, 21 and 28 days when they were submitted to radiographic examination of the knee, arthrotomy and collection of the synovial membrane. RESULTS:The osteoarthritis group showed significant differences compared to control group on days 7 and 14 in Roto-rod, in weight bearing and Von Frey tests in all days, and in radiological evaluation. Microscopic examination of the synovial membrane showed abnormalities of inflammatory character at all stages. CONCLUSION:The osteoarthritis induced by intra-articular monosodium iodoacetate in rats knee is a good model to be used in related research, because it provides mensurable changes on joint movements, tactile allodynia, progressive radiological degeneration and microscopic inflammation of the synovial membrane, that represent markers for osteoarthritis evaluation
