BackgroundDoxorubicin, a widely used anti‐tumour drug, is known to cause muscle loss in cancer patients.MethodsFollowing an acute dose of doxorubicin injection (2.5 mg/kg per body weight), we examined macrophage distribution in rat soleus muscle challenged by eccentric exercise (downhill running). Long‐term doxorubicin treatment (one injection every 3 days) on muscle mass and survival were also determined.ResultsUnder non‐exercised condition, increased tumour necrosis factor (TNF)‐alpha mRNA and decreased IL‐10 mRNA were observed in soleus muscle of doxorubicin‐treated rats, compared with saline‐treated control rats. However, increases in inflammation score (leukocyte infiltration), nitrotyrosine level, and M1 macrophage (CD68+) invasion in exercised soleus muscle were absent in doxorubicin‐treated rats, whereas increased M2 macrophage (CD163+) localization in exercised muscle was less affected by doxorubicin. Despites coenzyme Q (Q10) supplementation significantly elevated TNF‐alpha mRNA, nitrotyrosine, and anti‐oxidant gamma‐glutamylcysteine synthetase (GCS) levels in non‐exercised soleus muscle, these pro‐inflammatory responses were also abolished in doxorubicin‐treated rats. Results from long‐term doxorubicin treatment show a significant muscle loss followed by an accelerated death, which cannot be reversed by Q10 supplementation.Conclusions(i) Doxorubicin impairs inflammation mechanism by depleting M1 macrophage in exercised skeletal muscle; (ii) Muscle loss and accelerated death during prolonged doxorubicin treatment cannot be reversed by Q10 supplementation.
Rationale:Calcium-sensing receptor (CaSR) mutations can cause life-threatening neonatal severe hyperparathyroidism (NSHPT). The medical management of NSHPT is often challenging and complex. Here, we present a case of NSHPT caused by a novel homozygous CaSR mutation.Patient concerns:A Chinese female infant presented with poor feeding, constipation, severe hypotonia, and periodic bradycardia. Biochemistry tests revealed markedly elevated serum levels of Ca2+ and parathyroid hormone (PTH).Diagnoses:Genetic sequencing revealed a previously undescribed CaSR mutation in exon 3 (c.242T>A; p.I81K). A diagnosis of NSHPT secondary to homozygously inherited familial hypocalciuric hypercalcemia syndrome was established.Interventions:Cinacalcet was administered after the common treatments (low-calcium intake, hydration, and furosemide), calcitonin, and pamidronate therapy all failed.Outcomes:Serum Ca2+ decreased and stabilized with cinacalcet therapy. During a 10-month follow-up, total calcium was maintained within the high-normal range and PTH was normalized.Lessons:A trial of cinacalcet therapy might be undertaken in cases of NSHPT while definitive results of the genetic analysis are awaited.

Xiyingmen Building Materials and Home Furnishings Plaza was founded in 1997 and headquartered in Shanghai. It specializes in the development, operation and management of urban complex and building materials and furniture plaza projects. Because of its early start and perfect supporting facilities, it also achieved good financial performance in the early stage. However, with the continuous emergence of large commercial complexes, the pattern of new and old business districts has changed significantly, and the competition between commercial complexes has become increasingly fierce. The financial performance of Xiyingmen Building Materials and Home Furnishing Plaza has declined year by year. Therefore, it is urgent to carry out special research, propose feasible measures, summarize effective experience, which is conducive to promoting the sustainable development of the industry, bringing better benefits to the operation of the industry, and can provide theoretical reference for the formulation of marketing strategies for many similar commercial complexes. From the perspective of the demand and service of target merchants and consumers, this paper first analyzes the macro environment analysis, regional business development analysis, competitor comparison analysis, project positioning analysis, SWOT analysis and other aspects of Xiyingmen Building Materials and Home Furnishing Plaza, which provides a solid foundation and reference value for the project to formulate marketing strategies.
There is so much influence to the fatigue life of the whole structural component from cross-section transition zone radian, and more than 100 times difference is made by different structural details for fatigue life [1]. According to structure and features of variable cross-section strut-and-tie,combing with its’ geometric properties and applied stress ways, at the basis of nominal stress method, the fatigue life of variable cross-section strut-and-tie is analyzed in the paper. Then, transitional zone is improved and fatigue life is analyzed. At last, the improved is compared with previous structure and analysis of Influence of fatigue life from variable cross-section strut-and-tie transition zone radian in theoretical. The conclusions of this paper guide other similar researches.
