BACKGROUND: Oxidative stress is closely related to all aspects of cancer. Cosmos caudatus ethanolic extract (CCEE) has been proved to have antioxidant effect that inhibited cancer cell growth due to its bioactive compounds such as catechin, quercetin and chlorogenic acid. This study aimed to evaluate antioxidant and anticancer activity of CCEE and its compounds.METHODS: Total phenol was measured according to the Folin-Ciocalteu method. Catechin, quercetin and chlorogenic acid contained in CCEE were identified by high-performance liquid chromatography (HPLC). Antioxidant activity was evaluated by 2,2′-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS)-reducing activity, 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity, and ferric reducing antioxidant power (FRAP) activity test. The cytotoxic activity of CCEE was determined by MTS [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium] assay on HeLa cells.RESULTS: The result showed that total phenol of CCEE was 181.64±0.93 µg Cathecin/mg extract. ABTSreducing activity test showed that catechin had the highest activity (2.90±0.04 µg/mL), while CCEE had moderate activity compared to other compounds. FRAP activity test demonstrated that catechin had the highest activity (315.83 µM Fe(II)/µg) compared to other compounds. DPPH scavenging activity of CCEE was 22.82±0.05 µg/mL. Cytotoxicity test on HeLa cell showed that CCEE had lower activity (inhibitory concentration (IC)50= 89.90±1.30 µg/mL) compared to quercetin (IC50 = 13.30±0.64 µg/ mL).CONCLUSION: CCEE has the lowest antioxidant activity compared to quercetin, catechin, and chlorogenic acid and has the lowest anticancer activity compared to quercetin. However, CCEE and its compounds has potential as antioxidant and anticancer properties.KEYWORDS: antioxidant, anticancer, catechin, Cosmos caudatus, quercetin
Kepatuhan petugas terhadap SOP di laboratorium merupakan kesalahan pra analitik yang memberikan kontribusi paling besar pada kesalahan di laboratoriun, yaitu 77,1%. Salah satu faktor penyebab kesalahan yaitu sistem managemen rumah sakit seperti individu, organisasi, dan lingkungan kerja. Penelitian ini bertujuan untuk mengetahui faktor-faktor yang berhubungan dengan kepatuhan penerapan standar operasional prosedur pengambilan darah vena. Penelitian ini merupakan penelitian deskriptif analitik dengan pendekatan cross sectional. Sampel dalam penelitian ini adalah 38 orang flebotomis yang bekerja di Instalasi Laboratorium Patologi Klinik RSUP Dr. Hasan Sadikin Bandung. Hasil analisis secara statistik Chi Square dengan taraf kepercayaan 95% (p<0,05) menunjukkan adanya hubungan antara variabel beban kerja dengan kepatuhan penerapan standar operasional prosedur (p=0,008). Dari hasil penelitian ini diharapkan dapat memberikan masukan pada pihak RSUP Dr. Hasan Sadikin Bandung untuk dapat menambah petugas flebotomi pada saat waktu puncak pasien saat diambil darah meningkat.

Pisang mengandung antioksidan yang tinggi. Selain itu, kulit pisang juga memiliki kandungan kimia seperti alkaloid, flavonoid, saponin, fenol, juga terpenoid yang mampu menghambat pertumbuhan bakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri kulit pisang ambon, mengetahui variasi konsentrasi dari rentang 0%; 0,2%; 0,4%; 0,6%; 0,8%; dan 1% , juga mengetahui waktu kontak (waktu inkubasi) yang paling efektif. Waktu yang dipakai disesuaikan dengan kurva pertumbuhan bakteri yaitu 0 jam, 5 jam, 15 jam, 30 jam, dan 45 jam dengan 3 kali pengulangan sehingga terdapat 75 unit eksperimen termasuk kontrol positif dan negatif. Jenis penelitian yang digunakan adalah quasi eksperimen. Metode yang digunakan adalah makrodilusi dengan pembuatan seri pengenceran pada tabung reaksi yang berisi media cair Mueller Hinton Broth (MHB) sebagai media yang dapat menumbuhkan bakteri Staphylococcus aureus dan sejumlah tertentu suspensi Staphylococcus aureus. Seri tabung diinkubasi pada suhu 37°C selama waktu tertentu dan diukur absorbannya menggunakan spektrofotometer UV-Vis pada panjang gelombang 620 nm. Ekstrak kulit pisang ambon dapat menghambat pertumbuhan bakteri Staphylococcus aureus pada konsentrasi 0,4% dengan waktu kontak 30 jam.
Penyakit karena infeksi dapat ditularkan dari satu orang ke orang lain, dari hewan ke manusia dan disebabkan juga oleh mikrooganisme seperti, bakteri, jamur, virus dan parasit. Salah satunya adalah bakteri Staphylococcus aureus. Upaya pengendalian terhadap bakteri Staphylococcus aureus telah banyak dilakukan dengan adanya antibiotik. Namun penyalahgunaannya dapat menyebabkan resistensi dalam penggunaan jangka waktu lama. Untuk mengatasinya maka perlu mengembangkan antibakteri dari obat tradisional terutama bahan alamiah yang dapat digunakan sebagai alternatif pengganti obat antibakteri. Salah satunya adalah kulit pisang muli (Musa acuminata L.) sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui konsentrasi minimum ekstrak kulit pisang muli sebagai antibakteri dan waktu kontak yang efektif terhadap Staphylococcus aureus. Jenis penelitian ini adalah analitik laboratorik yang dilaksanakan pada bulan Maret 2019 hingga Mei 2019. Simplisia kulit pisang muli dilakukan dengan maserasi dengan etanol 96% kemudian dilakukan tahap freeze drying dibuat konsentrasi 0,2%, 0,4%, 0,6%, 0,8% dan 1%. Sampel uji dilakukan metode makrodilusi yaitu pegukuran absorban menggunakan spektrofotometer UV-Vis panjang gelombang 620 nm dengan lama waktu kontak 0 jam, 5 jam, 15 jam, 30 jam dan 40 jam. Data absorban dianalisis dalam bentuk tabel dan grafik kemudian dari rerata hasil absorban diperoleh konsentrasi minimum dan waktu kontak efek ekstrak kulit pisang muli. Hasil penelitian diperoleh bahwa ekstrak kulit pisang muli (Musa acuminata L.) terhadap pertumbuhan Staphylococcus aureus adalah 0,4% dengan waktu kontak optimum 30 jam.
