Penyakit karena infeksi dapat ditularkan dari satu orang ke orang lain, dari hewan ke manusia dan disebabkan juga oleh mikrooganisme seperti, bakteri, jamur, virus dan parasit. Salah satunya adalah bakteri Staphylococcus aureus. Upaya pengendalian terhadap bakteri Staphylococcus aureus telah banyak dilakukan dengan adanya antibiotik. Namun penyalahgunaannya dapat menyebabkan resistensi dalam penggunaan jangka waktu lama. Untuk mengatasinya maka perlu mengembangkan antibakteri dari obat tradisional terutama bahan alamiah yang dapat digunakan sebagai alternatif pengganti obat antibakteri. Salah satunya adalah kulit pisang muli (Musa acuminata L.) sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui konsentrasi minimum ekstrak kulit pisang muli sebagai antibakteri dan waktu kontak yang efektif terhadap Staphylococcus aureus. Jenis penelitian ini adalah analitik laboratorik yang dilaksanakan pada bulan Maret 2019 hingga Mei 2019. Simplisia kulit pisang muli dilakukan dengan maserasi dengan etanol 96% kemudian dilakukan tahap freeze drying dibuat konsentrasi 0,2%, 0,4%, 0,6%, 0,8% dan 1%. Sampel uji dilakukan metode makrodilusi yaitu pegukuran absorban menggunakan spektrofotometer UV-Vis panjang gelombang 620 nm dengan lama waktu kontak 0 jam, 5 jam, 15 jam, 30 jam dan 40 jam. Data absorban dianalisis dalam bentuk tabel dan grafik kemudian dari rerata hasil absorban diperoleh konsentrasi minimum dan waktu kontak efek ekstrak kulit pisang muli. Hasil penelitian diperoleh bahwa ekstrak kulit pisang muli (Musa acuminata L.) terhadap pertumbuhan Staphylococcus aureus adalah 0,4% dengan waktu kontak optimum 30 jam.
