Pesq. Vet. Bras. 33(6) ), at the ratio of 4g/kg of pelleted feed. The completely randomized design with two treatments was used: one control group and one group fed on the mentioned diet. The evaluation of survival rate, the intestinal microbiota analysis by microbiological culture, histomorphometrical analysis of intestinal mucosa and chemical analysis of carcass was performed. The results showed that tilapias from the treated group had higher relative survival rate (P<0.05) than the control group, higher number of colony--forming units (P<0.05) regarding intestinal colonization by B. cereus and B. subtilis, and higher rates of intestinal mucosal integrity (P<0.05), evaluated by histomorphometry. As for the latter, the group being fed on feed with probiotic additive was observed to have higher and larger villi, besides having a higher number of goblet cells than the control group. Concerning the carcass quality, the results showed that there was positive interference (P<0.05) of the probiotic on the treated group in comparison to the control group as in regard to levels of protein and ether extract. These results allow the inference that the supplementation with probiotic, as tested in this experiment, led to the intestinal colonization by beneficial bacteria and resulted in higher relative survival rate, decreased the mucosal desquamation and helped in the increase of the number of goblet cells.
The present study aimed to describe and characterize the cellular components during the evolution of chronic granulomatous inflammation in the teleost fish pacus (P. mesopotamicus) induced by Bacillus Calmette-Guerin (BCG), using S-100, iNOS and cytokeratin antibodies. 50 fish (120±5.0 g) were anesthetized and 45 inoculated with 20 μL (40 mg/mL) (2.0 x 106 CFU/mg) and five inoculated with saline (0,65%) into muscle tissue in the laterodorsal region. To evaluate the inflammatory process, nine fish inoculated with BCG and one control were sampled in five periods: 3rd, 7th, 14th, 21st and 33rd days post-inoculation (DPI). Immunohistochemical examination showed that the marking with anti-S-100 protein and anti-iNOS antibodies was weak, with a diffuse pattern, between the third and seventh DPI. From the 14th to the 33rd day, the marking became stronger and marked the cytoplasm of the macrophages. Positivity for cytokeratin was initially observed in the 14th DPI, and the stronger immunostaining in the 33rd day, period in which the epithelioid cells were more evident and the granuloma was fully formed. Also after the 14th day, a certain degree of cellular organization was observed, due to the arrangement of the macrophages around the inoculated material, with little evidence of edema. The arrangement of the macrophages around the inoculum, the fibroblasts, the lymphocytes and, in most cases, the presence of melanomacrophages formed the granuloma and kept the inoculum isolated in the 33rd DPI. The present study suggested that the granulomatous experimental model using teleost fish P. mesopotamicus presented a similar response to those observed in mammals, confirming its importance for studies of chronic inflammatory reaction.
Summary
The study objective was to make histological, histochemical and morphometric evaluations on the splenic Melanomacrophage centers (MMCs) of tilapias, Oreochromis niloticus (Linnaeus, 1758), that were subjected to chronic inflammation stimuli by implantation (IMP) of a glass coverslip in the subcutaneous tissue and through inoculation of the bacillus Calmette‐Guerin (BCG). Randomly distributed in four groups were 150 tilapias: IMP (n = 45); IMP+BCG (n = 45); BCG (n = 45); and control (n = 15). Nine fish per treatment and three control fish were sampled on days 3, 7, 14, 21 and 33. The results demonstrated that increased numbers and areas of these structures were related to the type of stimulus, and that these were greater for the specific response. The principal pigment component identified was hemosiderin. Results suggest that the intensity of the MMC response in O. niloticus depended on the type of inflammatory stimulus used, and that it was greater in fish inoculated with BCG, which induced a granulomatous inflammation when compared to the foreign body inflammatory response induced by the glass coverslips.
Aeromonas bacteria can cause an infection characterized by septicemia and is one of the most common pathogens in tropical fish. This disease is responsible for high morbidity and mortality rates, causing considerable losses in aquaculture. Thus, the understanding of its pathophysiology is crucial to develop control strategies of this bacterial infection in farmed fish. This study aimed to characterize early pathological aspects of acute sepsis in pacu (Piaractus mesopotamicus) experimentally infected with Aeromonas hydrophila. A total of 160 juvenile pacus were inoculated intraperitoneally with A. hydrophila (1.78 x 109 CFU/mL) and at 0 (control), 1, 3, 6, and 9 hours post-inoculation (hpi), animals were anesthetized and samples were collected for microbiological, light microscopy and transmission electron microscopy (TEM) analyzes. The results showed the occurrence of hemodynamic alterations, such as hemorrhage and congestion, which were observed mainly after 6 and 9 hpi. It was possible to re-isolate Aeromonas at all sampling times except in control group. However, just after 9 hpi it was possible to find the bacteria in all fish and tissues. Light microscopy analyses revealed a degenerative process, necrosis and vascular damage mainly at 6 and 9 hpi. According to the ultrastructural examination, areas of cellular death were identified in all examined tissues, especially at 6 and 9 hpi. However, the most severe, related to necrosis, were observed after 6 and 9 hpi. The findings suggested that this bacterium spreads in the first hpi through the fish organs, mainly affecting spleen, liver and kidney, causing irreversible lesions at the molecular level.
This study was performed to investigate the effect of food supplementation with chromium carbochelate on cortisolemia, glycemia and unspecific acute inflammation of pacus (Piaractus mesopotamicus) challenged by inactivated Aeromonas hydrophila in the swim bladder. 120 pacus (80.5 ± 11.3 g) were randomly distributed into 24 tanks (310 L, n = 5). After the adaptation period, the pacus were fed diets supplemented with 0 (control), 12, 18 and 36 mg of chromium carbochelate/kg of dry diet for 90 days (ad libitum). Fish fed diets containing 18 and 36 mg of Cr•kg −1 resulted in a reduction of cortisolemia and glycemia, and in an increase in the inflammatory response, seen through the greater accumulation of lymphocytes and thrombocytes. These results together suggest that food supplementation with chromium carbochelate, at the concentrations of 18 and 36 mg•kg −1 of feeds, brought homeostatic benefits to P. mesopotamicus. These benefits were translated, at least partially, as reductions in the consequences from inflammatory stress, with diminished cortisolemia and glycemia, which favored accumulation of lymphocytes and thrombocytes at the affected focus, as an increase in the immune response.
