Pesq. Vet. Bras. 33(6) ), at the ratio of 4g/kg of pelleted feed. The completely randomized design with two treatments was used: one control group and one group fed on the mentioned diet. The evaluation of survival rate, the intestinal microbiota analysis by microbiological culture, histomorphometrical analysis of intestinal mucosa and chemical analysis of carcass was performed. The results showed that tilapias from the treated group had higher relative survival rate (P<0.05) than the control group, higher number of colony--forming units (P<0.05) regarding intestinal colonization by B. cereus and B. subtilis, and higher rates of intestinal mucosal integrity (P<0.05), evaluated by histomorphometry. As for the latter, the group being fed on feed with probiotic additive was observed to have higher and larger villi, besides having a higher number of goblet cells than the control group. Concerning the carcass quality, the results showed that there was positive interference (P<0.05) of the probiotic on the treated group in comparison to the control group as in regard to levels of protein and ether extract. These results allow the inference that the supplementation with probiotic, as tested in this experiment, led to the intestinal colonization by beneficial bacteria and resulted in higher relative survival rate, decreased the mucosal desquamation and helped in the increase of the number of goblet cells.
-The objective of this work was to evaluate the effect of food supplementation with vitamin C on macrophage and multinucleated giant cell (MGC) activities of pacus at two stocking densities. The experiment was carried out in a 2x2x3 split-plot factorial arrangement with: 0 and 500 mg kg -1 vitamin C; 5 and 20 kg m -3 stocking densities; and evaluation times at 3, 6, and 12 days after the subcutaneous implantation of glass coverslips (DPI). The number of macrophages and MGC, as well as cortisol and glucose plasma levels were determined. The number of macrophages and MGC with two to five nuclei was significantly greater in fish supplemented with vitamin C at 5 kg m -3 stocking density at 3 DPI in comparison to nonsupplemented ones. The macrophage and MGC counts were lower in fish with high-plasma cortisol concentration. Supplementation with 500 mg vitamin C benefits macrophage activity on foreign-body inflammation, and high-cortisol concentration has suppressive effects on this response.Index terms: Piaractus mesopotamicus, ascorbic acid, glucocorticoid, inflammation, Langhans cells, multinucleated giant cells.
Vitamina C e cortisol endógeno em resposta inflamatória tipo corpo estranho em pacusResumo -O objetivo deste trabalho foi avaliar o efeito da suplementação com vitamina C no aumento da atividade de macrófagos e células gigantes multinucleadas (CGM), em pacus mantidos em duas densidades de estocagem. O experimento foi realizado em parcelas subdivididas, em arranjo fatorial 2x2x3 com: 0 e 500 mg kg -1 de vitamina C; densidade de estocagem de 5 e 20 kg m -3 ; e tempos de avaliação de 3, 6 e 12 dias após o implante subcutâneo (DPI) de lamínulas de vidro. Foram determinados o número de macrófagos e de CGM, bem como os níveis de cortisol e glicose no plasma. O número de macrófagos e de CGM com 2 a 5 núcleos foi significativamente maior nos peixes suplementados com vitamina C, à densidade 5 kg m -3 , aos 3 DPI, em relação aos não suplementados. Constatou-se menor quantidade de macrófagos e CGM em peixes com baixa concentração plasmática de cortisol. Abstract.-The present study assessed the kinetics of cell accumulation at the site of inflammation induced by thioglycolate, Escherichia coli lipopolysaccharide (LPS) and heat-inactivated Aeromonas hydrophila, in the pacu, Piaractus mesopotamicus (Characidae), swim bladder. The Aquaculture Center of Unesp, Jaboticabal, SP, Brazil, received fishes for diagnosis from fish farmers reporting fish crowding at pond edges and in water inlets. Fifty-three out of 194 cases showed round to oval, immobile whitish structures, measuring up to 162 mm diameter, identified as the dinoflagellate Piscinoodinium pillulare. In 34 diagnosed cases the parasites were present in the gills, in 2 on body surface, and 9 in both. Thirty-one out of 53 were tambacu hybrids hosts; 7, Piaractus mesopotamicus; 6, Colossoma macropomum; 5, Leporinus macrocephalus; 3, Oreochromis niloticus; and 1, Prochilodus lineatus. Fish showed increased mucous production on body surface and gills, while ecchymosis in the caudal peduncle and operculum was present. The gills also showed paleness, congestion, and petechiae. Histopathology presented a great number of trophonts situated between secondary lamellae, fixed to or detached from the epithelium. Primary lamellae presented interstitial hemorrhages, severe hyperplasia of the epithelium, goblet cells, and mononuclear infiltrates. The present work is the first report of P. pillulare in Brazil and emphasizes the importance of this dinoflagellate which caused significant economic losses from 1995 through 1997.
Piauçus (Leporinus macrocephalus), were raised in 300 m² ponds (density of 10 fish/m²) presenting asphyxia signals and daily mortality of 27 fishes. Specimens with 8-cm total body length, were collected for necropsy. Mucus of body surface and pieces of organs were collected and examined microscopically, in wet mounts, stained or in histological sections. The smears examination showed the presence of several spores in the secondary lamellae of the gill filaments, identified as Henneguya leporinicola n.sp (Myxozoa: Myxobolidae). Histopatological study showed epithelial hyperplasia and fulfilling of the spaces between the secondary lamellae, congestion and teleangiectasia sinusoidal. It was also observed hyperplasia of the goblet cells and several cysts of parasite with 70.3mum diameter. Such cysts were situated among the secondary lamellae, covered or not by the hyperplasic epithelium. With this diagnostic, three applications of formalin solution 10 ml/m³ were carried out. Fifteen days after that, fish were examined again to ascertain whether the treatment was efficient on disease caused by the protozoa. The tissue alterations present in the gills after the treatment were just a moderate sinusoidal congestion and a slight epithelial hyperplasia on the base of the secondary lamellae.
Summary
The study objective was to make histological, histochemical and morphometric evaluations on the splenic Melanomacrophage centers (MMCs) of tilapias, Oreochromis niloticus (Linnaeus, 1758), that were subjected to chronic inflammation stimuli by implantation (IMP) of a glass coverslip in the subcutaneous tissue and through inoculation of the bacillus Calmette‐Guerin (BCG). Randomly distributed in four groups were 150 tilapias: IMP (n = 45); IMP+BCG (n = 45); BCG (n = 45); and control (n = 15). Nine fish per treatment and three control fish were sampled on days 3, 7, 14, 21 and 33. The results demonstrated that increased numbers and areas of these structures were related to the type of stimulus, and that these were greater for the specific response. The principal pigment component identified was hemosiderin. Results suggest that the intensity of the MMC response in O. niloticus depended on the type of inflammatory stimulus used, and that it was greater in fish inoculated with BCG, which induced a granulomatous inflammation when compared to the foreign body inflammatory response induced by the glass coverslips.
