<p class="MsoNormal" style="margin: 0cm 21.6pt .0001pt 18pt; text-align: justify; text-indent: 27pt;"><span lang="en-us" xml:lang="en-us">The objectives of this study are to </span><span lang="en-us" xml:lang="en-us">determine the substitution level of defatted rice bran flour substituted to wheat flour, to know the impact of defatted rice bran flour substitution to physical,  chemical and organoleptic characteristics of simulated chips, and to compare defatted rice bran flour with whole rice bran flour as substituted raw material in making of simulated chips to physical, chemical, and organoleptic characteristics.  The substitution of defatted rice bran increased content of water, ash, protein and fiber, but fat, carbohydrate and energy decreased compare with control simulated chips (</span><span style="font-family: Symbol;" lang="en-us" xml:lang="en-us">a</span><span lang="en-us" xml:lang="en-us"> = 0.05).  Organoleptic test of simulated chips with some levels of defatted rice bran flour substitution showed that acceptance of panelist  to color has mode ranged from not like to like; mode of aroma ranged from neutral to like; and both of taste and crispiness has mode ranged from not like to like.  Percentage of panelist that accepted simulated chips color in some level of defatted rice bran flour substitution ranged from 20% to 96.7%, aroma 76.7% to 90%, taste 50% to 100% and crispiness 53.3% to 100%.  The substitution of defatted rice bran flour will decrease acceptance of panelist to color, aroma, taste and crispiness (</span><span style="font-family: Symbol;" lang="en-us" xml:lang="en-us">a</span><span lang="en-us" xml:lang="en-us"> = 0,05).  Simulated chips substituted by defatted rice bran flour and whole rice bran flour shows that the content of water, ashes, carbohydrate, energy, insoluble dietary fiber and total dietary fiber of simulated chips were not significantly difference (</span><span style="font-family: Symbol;" lang="en-us" xml:lang="en-us">a</span><span lang="en-us" xml:lang="en-us"> = 0.05), but there was a significantly difference for fat and soluble dietary fiber. The organoleptic test showed that the color, aroma and taste of simulated chips were not significantly difference, but there was a significantly difference for crispiness (</span><span style="font-family: Symbol;" lang="en-us" xml:lang="en-us">a</span><span lang="en-us" xml:lang="en-us"> = 0,05).</span></p>

The epithelium of the chicken embryonic glandular stomach (proventriculus) differentiates into both a glandular and a luminal epithelium, the cells of which express specific marker genes. The subsequent formation and differentiation of the glands then proceed under the influence of the mesenchyme. To search for possible candidates for the mesenchymal factors involved, we have now investigated the expression and function of Wnt5a in this process. Our current results show that Wnt5a is expressed in the mesenchyme during active gland formation and that overexpression of this gene in ovo results in the increased and ectopic expression of some of the marker genes of the luminal and glandular epithelia. In particular, the overexpression of Wnt5a markedly enhances the expression of the embryonic chicken pepsinogen gene, a marker of the glandular epithelium, indicating its role as a mesenchymal factor that regulates the differentiation of the proventricular epithelium.

Background The identification of banana cultivars genome is needed to provide a valid identity from the accession of bananas which are used as basic data in the management of in situ and ex situ banana conservation as well as for further breeding of banana cultivars. Results The size of PCR-amplified matK ranged from 844 to 860 bp and showed a high variability. The haplotype diversity was 0.9048 with nine haplotypes. Haplotype distribution map revealed the lineage patterns of banana cultivars from Java. Reconstruction of genetic relationships using the maximum likelihood, maximum parsimony and Bayesian inference algorithms produces tree topologies and classifications that are grouped according to their genomic groups, into three main clades, i.e., AA/AAA, AAB and ABB. Based on the previously derived age constraints and fossil data, we estimate (Musaceae) that genetic divergence times of all samples occurred during the Eocene (95% HPD: 51.9 Mya), Musa acuminata group (AA, AAA, AAB) with Musa balbisiana group (BB and ABB) occurred during the Oligocene (95% HPD: 26 Mya), and the separation on each banana cultivars occurred during the Middle Miocene to Pliocene (95% HPD: 16.5–2.5 Mya). Conclusions From this study, we conclude that all studied cultivars are closely related according to its genomic groups with high variation. Genetic variation among those cultivars creates nine haplotypes. The development of variety which leads to the formation of different banana cultivars had suggested to be occurred long ago along with human migration and domestication.

Chili pepper is widely used and cultivated by Indonesian people. There are three species of chili pepper, i.e. Capsicum annuum, Capsicum frutescens, and Capsicum violaceum. C. frutescens has a higher economic value due to its pungency and carotenoid content. C. frutescens has several cultivars, one of those is C. frutescens cv. Cakra Hijau. This cultivar is resistant against pest and disease and has very high pungency. This special character of chili pepper is capsaicin, its secondary metabolic. Moreover, capsaicin also serves as defense mechanism, antiarthritis, analgesic, and anticancer. This study was aimed to isolate acyltransferase (AT3) gene which encoded capsaicin synthase (CS) enzyme. AT3 gene was isolated through PCR using forward primer 5'-ATG GCT TTT GCA TTA CCA TCA-3' and reverse primer 5'-CCT TCA CAA TTA TTC GCC CA-3'. Data were analyzed using DNA Baser, BLAST, and ClustalX. This study has successfully isolated 404 bp fragments of AT3 gene. These fragments are located at 1918-1434 bp referred to AT3 gene from C. frutescens cv. Shuanla. The isolation of upstream and downstream fragments of AT3 gene from C. frutescens cv. Cakra Hijau is undergoing.

Frog is an amphibian which is widely spread around the world. Indonesia houses 450 species which represent 11% of frog species in the world. In Java Island alone, there live 42 species of frogs and toads. Frogs can be used as an environment indicator in that the presence of frog in a particular place indicates that the place stays natural and unpolluted. The 1 st Campus of State University of Malang, which is located in the heart of Malang District, has been developing rapidly currently. Thus, it requires the construction of new various facilities to support its huge activities. Extensive construction can be destructive even damaging to the habitat of frog, which potentially threats the frog's life, if it does not take the environmental impact into careful consideration. This study is aimed to identify the species of frog which survives at State University of Malang with, particularly the frog species foun d in 1995. Species identification was conducted by observing the morphological character. This study found that there were four species with three species remained survived in 1995; those were Duttaphrynus melanostictus, Polypedates leucomystax, and Kaloula baleta; and one new species called Rana chalconota. This study also revealed that there were four species which were extinct; those were Fejervarya cancrivora, Fejervarya limnocharis, Ingerophrynus biporcatus, and Occidoziga lima. This situation shows the decreasing amount of species from 7 to 4 within the last 17 years. This result indicates that there is a serious environmental degradation which causes the losing of frog habitats. Further research is needed to study the ecological condition changing in order to save the frog species.

Padi (Oryza sativa L.) adalah tanaman pangan utama masyarakat Indonesia. Masalah kekeringan dalam upaya penanaman padi adalah hal yang biasa di kalangan petani. Penelitian tahan cekaman kekeringan pada padi perlu dilakukan untuk mengatasi masalah ketersediaan pangan. Sifat tahan cekaman kekeringan pada tanaman salah satunya dikendalikan oleh gen DREB2A. Varietas padi lokal Indonesia untuk tahan cekaman kekeringan belum banyak diteliti. Penelitian ini bertujuan untuk mempelajari varietas padi lokal Jawa Timur yang tahan terhadap cekaman kekeringan menggunakan gen DREB2A, yang diisolasi menggunakan primer forward 5 '-CCT CAT TGG GTC AGG AAG AA-3 ' dan primer reverse 5 '-GGA TCT CAG CCA CCC ACT TA-3 '. Penelitian ini menggunakan tiga varietas padi lokal, yaitu Berlian dan SOJ A3, keduanya dari Banyuwangi, dan varietas Jawa dari Malang. Isolasi DNA total dilakukan mengikuti protokol Kit Macherey-Nagel. Amplifikasi gen DREB2A dilakukan menggunakan metode Polymerase Chain Reaction (PCR). Fragmen gen DREB2A berhasil diamplifikasi sepanjang 239 bp (varietas Berlian), 239 bp (varietas SOJ A3), dan 240 bp (varietas Jawa). Analisis sampel gen DREB2A varietas padi lokal Jawa Timur menunjukkan hilangnya kemampuan gen DREB2A dari varietas Jawa untuk mengikat cis-regulator elemen promotor gen target. Anggota Famili Poeciliidae merupakan ikan introduksi dengan karakteristik berukuran kecil, live-bearers, dimorfi seksual dari segi ukuran tubuh, pola warna dan struktur gonopodium sebagai ciri diagnostik pada ikan jantan. Ikan ini berperan sebagai pengontrol perkembangan nyamuk malaria dan model pembelajaran biologi. Sejauh ini kajian taksonomi dan penelusuran hubungan kekerabatan didalam famili Poeciliidae belum ada. Tujuan penelitian ini adalah mengidentifikasi jenis, kekerabatan dan pengelompokkan berdasarkan karakter morfologi (morfometrik, meristrik, gonopodium dan ciri diagnostiknya). Jenis penelitian ini adalah deskriptif eksploratif, dengan pendekatan survey. Pada penelitian ini ditemukan tiga spesies ikan Poeciliidae yaitu Xiphoporus helleri, Poecilia mexicana dan Poecilia reticulata. Dendogram hasil analisis morfologi menggunakan metode UPGMA diperoleh dua kelompok percabangan apomorfi yaitu kelompok Genus Xiphoporus dan Genus Poecilia. Sub Clade automorfi antara Xiphoporus helerri A dan B dengan nilai kesamaan 92%. Pengelompokkan ikan Poeciliidae dibedakan berdasarkan lima karakter utama terpilih yaitu: SL (standart length), Hdl (Panjang kepala), Panjang gonopodium, panjang sirip ekor dan PreDL (Panjang sebelum sirip dorsal).Kejelasan identifikasi ikan introduksi khas Telaga Sari menjadi modal utama dalam aquaculture, manajemen, konservasi, biodiversitas dan perkembangan ilmu pengetahuan.
