This study was concerned with the green synthesis of gold nanoflowers (AuNFs) using the bioactive constituents of Rosmarinus officinalis (rosemary) and Helichrysum italicum (immortelle) extracts, as reducer and stabilizer agents along with the determination of their antibacterial and antibiofilm activity against E. coli, S. aureus, and S. epidermidis. The AuNFs were characterized using STEM, UV–Vis, DLS, ZETA, FESEM-EDX, and FTIR techniques. The antibacterial and antibiofilm activity of the AuNFs were evaluated by microdilution broth and microtiter plate (MTP) tests, respectively. STEM and DLS analysis confirmed the flower-like morphology of gold nanoparticle clusters of R. officinalis-AuNFs (R-AuNFs) and H. italicum-AuNFs (H-AuNFs) with a size of 20–130 nm and 15–90 nm, respectively. The MICs of R-AuNFs were found to be 40 µg/mL for E. coli and S. epidermidis and 160 µg/mL for S. aureus. The MICs of H-AuNFs against all bacterial strains were 20 µg/mL. All tested AuNFs exhibited a strong dose-dependent antibiofilm activity against the test strains, and H-AuNFs was more effective than R-AuNFs. The green synthesis of AuNFs from the rosemary and immortelle extracts can be applied as a potential agent to overcome the growth of biofilm-producing microorganisms in food industries.
Aliarcobacter spp. have has been isolated from numerous food products at retail and from animal carcasses and feces at slaughter. The objectives of this study were: i) to isolate Aliarcobacter species (Aliarcobacter butzleri, Aliarcobacter skirrowii, Aliarcobacter cryaerophilus) from different slaughterhouses’ samples, ii) to detect genetic diversity, antibiotic resistance, biofilm ability and putative virulence gene profiles of the isolates. The antibiotic susceptibility to antimicrobials was examined using the Kirby-Bauer disc diffusion method. A molecular investigation of antibiotic resistance and virulence factors was also conducted using polymerase chain reaction (PCR). A total of 22 (14.6%) Aliarcobacter spp. isolates were obtained, with varying levels of antibiotic resistance observed. The genes tetO, tetW, and gyrA were detected in 0%, 31.8%, and 27.2% of the isolates, respectively. All isolates were resistant to ampicillin, rifampin, and erythromycin, while tetracycline was found to be the most effective antibiotic, with 81.8% of the isolates showing susceptibility to it. In terms of virulence factors, all isolates (100%) harbored more than one of the nine putative virulence genes tested, with 18.1% of isolates carrying more than three. Regarding biofilm formation, 7 (31.8%) and 4 (18.1%) isolates were found to form strong and moderate biofilms, respectively, while one (4.5%) isolate was classified as a weak biofilm producer. ERIC-PCR band patterns suggested that the isolated Aliarcobacter spp. from slaughterhouses had different sources of contamination. These findings highlight the potential risk posed by pathogenic and multidrug-resistant Aliarcobacter spp. in food, and the need for control measures throughout the food chain to prevent the spread of these strains. The results indicate that foods of animal origin and cattle slaughterhouses are significant sources of antimicrobial resistant Aliarcobacters.
Bu çalışmada, Kayseri ilinde mevcut olan birinci sınıf bir sığır mezbahasında kesim hattı boyunca alınan örneklerde biyofilm oluşturma yeteneğine sahip E. faecalis varlığı ve izolatlar da biyofilm ile ilişkili virülans faktör genlerinin araştırılması amaçlandı. Çalışmada, kesimhaneden alınan toplam 300 adet örnekte (180 karkas, 102 ekipman ve yüzey ve 18 mezbaha atık suyu) etkenin varlığı konvansiyonel yöntem ve PZR ile analiz edildi. İzolatların biyofilm oluşturma yeteneklerini belirlemede Kongo kırmızısı agar ve mikroplaka testi ve biyofilm ile ilişkili virülans genlerinin (gelE ve esp) tespitinde PZR kullanıldı. Analiz edilen 300 örneğin 40’ından (%13.3) E. faecalis izole edildi ve bu izolatların 35 (%87.5) biyofilm oluşturma yeteneğinde idi. Biyofilm pozitif olan izolatların 33’ünde (%82.5) gelE geni belirlendi, ayrıca bu izolatların bir tanesini (% 2.5) esp genini de içeriyordu. Sonuç olarak, Kayseri ilindeki kesimhaneden alınan tüm örnek türlerinde mezbahanın hijyenik kalitesini yansıtan biyofilm oluşturma yeteneğine sahip E. faecalis’in tespit edilmesi ve izolatların biyofilm oluşumunun virülans faktör genler ile ilişkisi tespit edilmesi gıda güvenliği açısından önemlidir.
Bu çalışmada, daha önceki çalışmalarla mezbaha ve mandıralardan izole edilen ve halk sağlığı açısından risk oluşturan önemli patojenlerden Staphylococcus aureus, Listeria spp., Escherichia coli ve Salmonella spp. izolatlarının biyofilm oluşturma yeteneklerinin belirlenmesinde Kongo Kırmızısı Agar (KKA) ve Mikroplak (MP) yöntemlerinin etkinliğinin ve duyarlılıklarının karşılaştırılması amaçlandı. Çalışma sonucunda 135 izolatın %51.1’i KKA’da, %53’ü ise MP’de biyofilm üreticisi olarak belirlendi. Analiz edilen izolatlar arasında KKA yönteminin duyarlılığının en yüksek olduğu izolat S. aureus idi (%97) bunu Listeria spp. (%59), Salmonella spp (%17). ve E. coli (%13) izledi. Listeria spp., Salmonella spp .ve E. coli seçicilik oranları ise sırasıyla %39, %59 ve %89 idi. Ancak S. aureus izolatlarının tamamı MP testinde biyofilm pozitif sonuç verdiği için KKA’nın seçiciliği bu izolat için belirlenemedi. Sonuç olarak, KKA yönteminin duyarlılığı analiz edilen izolatlardan sadece S. aureus için yüksekti, diğer izolatlarda ise bu yöntemin seçiciliğinin iyi olduğu görüldü. Gıda ve gıda işleme ortamlarında halk sağlığı için risk oluşturabilen patojen suşlarına ait biyofilm yeteneklerinin doğru tespit edilmesi önem arz etmektedir. Bu nedenle, KKA’nın MP yöntemi ile tamamlayıcı bir şekilde uygulanması sonuçların güvenirliği ve biyofilmlerin tespiti için önemli olduğu sonucuna varıldı.
