Glucose-6-phosphate dehydrogenase (G6PD) deficiency is an enzymopathy in which reduced NADPH concentrations are not maintained, resulting in oxidative damage. We evaluated G6PD activity, oxidative stress levels and Trolox equivalent antioxidant capacity in individuals with the A-(202G>A) mutation for G6PD deficiency. Five hundred and forty-four peripheral blood samples were screened for G6PD deficiency; we also analyzed lipid peroxidation products measured as thiobarbituric acid reactive species and Trolox equivalent antioxidant capacity. Men with the A-(202G>A) mutation had lower G6PD activity than women with the same mutation. Individuals with the A-(202G>A) mutation also differed in mean Trolox equivalent antioxidant capacity values but not for thiobarbituric acid reactive species values. We concluded that A-(202G>A) mutation is associated with reduced G6PD activity and increased Trolox equivalent antioxidant capacity.

A adenosina trifosfato (ATP), energia calórica, e a nicotinamida adenina dinucleotídeo reduzida (NADH) e nicotinamida adenina dinucleotídeo fosfato reduzida (NADPH), energia redutora, são obtidas a partir da oxidação anaeróbica da glicose, via das pentoses e ciclo da glutationa. 1,2A via das pentoses é responsável pela oxidação de cerca de 10% da glicose consumida pelo glóbulo vermelho. A ação de duas enzimas seqüenciais, a glicose-6-fosfato desidrogenase (G6PD) e a 6-fosfogliconato desidrogenase (6PGD) reduzem a coenzima nicotinamida adenina dinucleotídeo fosfato (NADP) à nicotinamida adenina dinucleotídeo fosfato reduzida (NADPH), sendo, praticamente, toda fonte geradora de NADPH. Ao manter a glutationa no estado reduzido (GSH), através da glutationa redutase, essa via desempenha importante papel na proteção do glóbulo vermelho frente aos processos oxidativos, que são responsáveis pela redução da vida média do eritrócito.

The other n-6 and n-3 fatty acids can be synthesized from LA and LNA or be obtained through the diet.Of the n-6 and n-3 series fatty acids of nutritional value, arachidonic (AA, 20:4 n-6), eicosapentaenoic (EPA, 20:5 n-3) and docosahexaenoic (DHA, 22:6 n-3) acids are the most important.As in the human body, LA, an n-6 fatty acid, must be converted into AA, and LNA, an n-3 fatty acid, into EPA and DHA in order for them to exercise their functions, many kinetic studies are performed to measure these conversions from diets with different n-6/n-3 ratios.( 1) The first step in the process to assess the fatty acid composition of different foods and biological materials is to extract the lipids. Extraction methods are crucial to accurately measure fatty acid concentrations, as methods may degrade some fatty acids, especially polyunsaturated fatty acids and thus overestimate the true values.Accordingly, in this issue of the Journal of Hematology and Hemotherapy, a study by Morais et al. Bligh and Dyer, (4) Rose-Gottlieb, (5) and Gerber (6) and an alternative technique, proposed by the authors, with the objective of comparing the efficiency of lipid extraction and to evaluate the fatty acid composition in total lipids from human plasma.
A anemia falciforme é uma hemoglobinopatia genética, caracterizada por uma alteração na estrutura da molécula de hemoglobina (HbA1), denominada hemoglobina S (HbS), que provoca uma distorção da estrutura dos eritrócitos, passando de forma esférica para forma de "foice" (sicke-cell). Este fenômeno é conhecido como falcização e é responsável por um quadro de anemia grave em indivíduos homozigotos e por quadros esporádicos de dor em indivíduos heterozigotos (portadores de traço falciforme), de acordo com a patofisiologia, em situações de baixa concentração de oxigênio. O teste mais específico e mais sensível usado para diagnosticar anemia é o teste eletroforese de hemoglobina, que é, contudo, difícil de ser incorporado em laboratórios de rotina, devido ao seu alto custo. O objetivo deste trabalho foi estimar a prevalência do traço falciforme na população da cidade de Araraquara-São Paulo. A prevalência de indivíduos assintomáticos com hemoglobina S (traço falciforme) na região de Araraquara-SP foi de 1,5%. Separandose o estudo de prevalência por raça, observamos 0,6 % de traço falciforme em indivíduos da raça branca e 15,4 % em indivíduos da raça negra.
