In this study, Klebsiella pneumoniae BLh-1 was grown under exponential fed-batch mode in cultures using raw glycerol from biodiesel synthesis under anaerobiosis to produce value-added products. Specific growth rates of 0.035, 0.07, and 0.105 h-1 were tested and 1,3-propanediol (1,3-PDO) and ethanol were produced in all cultures, whereas the formation of 2,3-butanediol was not observed. The highest concentrations of 1,3-PDO (38.5 g L-1) and ethanol (13.2 g L-1) were achieved under the specific growth rate of 0.105 h-1. Acetic and lactic acids were also produced at high concentrations (17.3 and 7.8 g L-1 , respectively) under these conditions. The yields of 1,3-PDO and ethanol increased over time, reaching final values of 0.56 and 0.19 g g-1 , respectively. Overall productivities were 1.43 g L-1 h-1 for 1,3-PDO and 0.49 g L-1 h-1 for ethanol. Compared to batch results, fed-batch operation favored K. pneumoniae metabolism towards the production of 1,3-PDO and ethanol, being an interesting process to add value to the ever-growing availability of raw glycerol from biodiesel industry.
Efficient pigment extraction is a key factor influencing the economic feasibility of microalgal biorefineries. Ethanol, acetone, ethyl acetate, and hexane were evaluated for the batch extraction of chlorophyll a, chlorophyll b, and carotenoids from Chlorella sorokiniana and Heterochlorella luteoviridis. Solvent‐to‐biomass ratios did not affect extraction results in most cases. Ethanol showed the highest efficiency in the recovery of chlorophylls, presenting high extraction yields. Acetone was among the most efficient solvents to extract violaxanthin, lutein, zeaxanthin, and carotene from both microalgae, proving to be generally suitable to this application. Conductor‐like screening model for real solvent segment activity (COSMO‐SAC) corroborated experimental results, proving to be an important tool in the screening of solvents for extraction processes.
RESUMO -Neste trabalho foram estudadas a dinâmica de crescimento e a produção de etanol a partir de glicose pela levedura Pachysolen tannophilus ATCC-32691. A velocidade de crescimento celular máxima (μ máx ) foi calculada através da curva de crescimento e equivale a 0,34 h -1 . Cultivos foram realizados em diferentes condições de temperatura, pH e concentração de glicose com o objetivo de se obter a melhor condição para a conversão da glicose a etanol através de um planejamento composto central. Obteve-se um modelo quadrático para a conversão de glicose a etanol, o qual foi validado por experimento em triplicata nas condições ótimas obtidas de 33,6 °C, pH igual a 4,37 e concentração de glicose igual a 42,3 g•L -1 , que geraram um Y Etanol = 0,47 g•g -1 .
A enzima β-galactosidase (lactase) é utilizada na produção de alimentos lácteos. Suas características funcionais e obtenção variam de acordo com os microrganismos de origem e condições de cultivo. O objetivo deste trabalho foi analisar os efeitos de diferentes meios de cultivo no crescimento e na produção de β-galactosidase pela levedura Kluyveromyces marxianus. As células foram inoculadas em meios complexos e meios sintéticos contendo glicose ou lactose como fonte de carbono à temperatura constante, realizando-se acompanhamento do crescimento celular durante 12 horas. Os resultados foram avaliados pela densidade ótica dos cultivos, concentração residual da fonte de carbono, quantificação de proteínas totais e da atividade enzimática específica. As curvas de crescimento e o consumo de açúcares pela leveduraK. marxianus foram semelhantes em todos os meios testados, observando-se uma estagnação do crescimento celular associada com a queda da concentração dos açúcares nos meios de cultivo. Ainda, obteve-se uma maior atividade enzimática específica (3,19 ± 0,01 μmol/mg/min) quando utilizado o meio sintético na presença de lactose. A utilização do meio sintético possibilitou a obtenção da enzima β-galactosidase com maior atividade, sendo considerado apropriado para o escalonamento do cultivo da levedura com o objetivo da produção da enzima de interesse.
