This study was aimed to produce recombinant growth hormone (rGH) from giant grouper (Epinephelus lanceolatus), giant gouramy (Osphronemus gouramy) and common carp (Cyprinus carpio) and compare their bioactivity potential by means of inducing the growth hormone of juvenile Nile tilapia (Oreochromis niloticus) as the model. DNA fragment encoding mature GH protein of giant grouper (El-mGH), giant gouramy (Og-mGH) and common carp (Cc-mGH) was amplified by PCR method. The purified PCR products were ligated to pCold-1 to generate pCold/El-mGH, pCold/OgmGH, and pCold/Cc-mGH protein expression vector, respectively. Each of the expression vectors was transformed into the Escherichia coli BL21. E. coli BL21 was cultured using 2xYT medium and protein production was induced by cold shock at 15±1 o C for overnight. The inclusion bodies of E. coli transformants containing protein expression vector were isolated by sonication method, and rGH production was analyzed by SDS-PAGE. Juvenile of Nile tilapia of average body weight of 12.41±3.28 g was intramuscularly injected once a week for 4 weeks with 1 μg inclusion body containing rGH per gram fish body weight. The result showed that rGH in molecular weight of about 25 kDa was obtained. Fish injected with rGH of El-mGH, Cc-mGH and Og-mGH grew 20.94%, 18.09%, and 16.99% faster, respectively, compared with the control. This result indicated that the three rGH produced in E. coli possessed biological activity when tested on Nile tilapia and further research is needed to find its effect on the growth of other aquaculture fish species. <p class="BasicParagraph" align="center"><strong>ABSTRACT</strong></p><p class="BasicParagraph" align="center"><strong> </strong></p><p class="Pa2">This research was conducted to evaluate the performance of eel production at stocking density of 2 g/L, 3 g/L, and 4 g/L reared in recirculating system for 60 days. The experimental design used was completely randomized design. Fish were fed on sinking pellet given according to body weight. Collected data were survival, specific growth rate, biomass growth, feed convertion ratio, weight variance, blood profile, blood glucose, and cortisol levels.The results showed that the best performance was found in 4 g/L density with specific growth rate and biomass growth were 0.99±0.30 and 7.50±4.13 g/day, feed conversion ratio of 1.22±0.58, and weight variance of 21.72±3.60%. Stress responses parameters which consisted of cortisol and blood glucose levels showed no significant difference at 22.45± 8.59 nm/L and 31.92± 4.29 mg/dL, respectively. Blood profile in form of hemoglobin, hematocrit, red blood cell and differential leukocytes showed no significant differences except for the leukocyte parameters with the highest rate was achieved at the stocking density of 4 g/L (2.70±0.001x105 sel/mm3). Rancangan percobaan yang digunakan adalah rancangan acak lengkap. Pakan berupa pelet tenggelam diberikan sesuai bobot tubuh. Data yang diambil meliputi kelangsungan hidup, laju pertumbuhan spesifik, laju pertumbuhan biomassa, konversi pakan, koefisien keragaman bobot, gambaran darah, kadar glukosa darah, dan kadar kortisol. Hasil penelitian menunjukkan bahwa padat tebar berpengaruh nyata (P<0,05) terhadap laju pertumbuhan biomassa; pertumbuhan biomassa terbaik dicapai pada kepadatan 4 g/L dengan laju pertumbuhan spesifik dan laju pertumbuhan biomassa sebesar 0,99±0,30 dan 7,50±4,13, konversi pakan sebesar 1,22±0,58, dan koefisien keragaman sebesar 21,72±3,60%. Parameter respons stres yaitu kadar kortisol dan glukosa darah tidak menunjukkan perbedaan yang nyata, yaitu masing-masing sebesar 22,45±8,59 nm/L serta 31,92±4,29mg/dL. Gambaran darah berupa hemoglobin, hematokrit, jumlah sel darah merah dan diferensial leukosit tidak menunjukkan perbedaan yang nyata (P>0,05), kecuali pada parameter leukosit dengan nilai terbaik pada perlakuan 4 g/L (2,70±0,001 x105 sel/mm3). Vertical profiles of water quality were surveyed before and after mass mortality of fish cultured in cages in Lake Cirata, a man made lake in Indonesia. The first survey was performed 4 months before the mass mortality and the second survey was carried out 4 days after the mass mortality. In the first survey, the conditions below 8 m depth were reductive and anoxic, although at 8 and 10 m depth, the water became oxidative in spite of low oxygen concentration in the second survey. These changes were probably caused by strong mixing of surface water with deeper water. Eight months after the mass mortality, when the redox potential at 8 m became reductive again, exposure tests of fish were performed in different layers in the water column around the cage. During the exposure tests the redox potential of the water was less than -100 mV and 93% of carp died within 6 h of exposure at 8 m depth. These results indicated that the mass mortality was caused by the strong mixing of surface water with reductive water in deeper layers.
This study aims to analyze the hormone profile of FSH, LH and E2 to the eel after exogenous hormone stimulation and blood glucose levels. Six formulated treatment applied with combination of dopamine antagonize 10 mg.mL–1 (A), estradiol (E2) 3 mg.mL–1 + A (EA), metyltestosteron (MT) 3 mg.mL–1 + A (MTA), hCG 2 mg.mL–1 + EA (hEA) and hCG 2 mg.mL–1 + MTA (hMTA), with (F) physiologis 0.9% NaCl to control. Six group eel (200±15g) reared in a concrete tank with a capacity of 3,400 liters and filled with sea water of 35 mg L–1 as much as 2000 liters. Eels injected 1 mL.kg–1 hormone by intramusculary, were feed to apparent satiation daily for 10 weeks. The study used Completely Randomized Design with one treatment factor, namely hormonal factor and its combination. Fish blood that ware directly concentration to FSH, LH and E2, the enhanced significantly high in the blood plasma on treatment hMTA and hEA P<0.05. Glucose concentration in the blood palsma is high enough in a row on a formula hMTA 67.33 mg.dL–1 and significantly different to P<0.05. The result indicates that induction of exogenous hormone (hMTA) improve FSH, LH and E2. FSH and LH profiles show permutation patterns during the development of eel gonad, beginning with the increase of FSH in the early phases of gonadal development. LH profile moves in line with the increase in E2 during gonadal maturation process eels (A. bicolor bicolor), with blood glucose levels in the normal range. This research is based on a phenomenon that occurs in State Detention Centers in Indonesia. It attempts to test the relation among proactive personality (PP), proactive work behavior (PWB), job satisfaction (JS) and role breadth self-efficacy (RBSE) variables. With a quantitative approach using AMOS, this study took data from 455 respondents from Detention Centers in Indonesia. The results show that PP, JS and RBSE have direct effect on PWB. Further findings will be discussed. The results are expected to increase the understanding of PWB and can be the basis for the human resource management team to decide better approach to build PWB in the organization and eventually implement appropriate policy.
In oviparous animals, the qualities of offspring are determined by the materials deposited in the developing oocytes that are synthesized and produced by the hepatocytes under the control of estrogen. This study was designed to evaluate the effect of turmeric powder supplementation on liver performance to support the reproduction performances of catfish (Pangasianodon hypophthalmus). The study used 60 catfish with average body weight of ± 2 kg. The experimental catfish were assigned into a completely randomized design with 4 treatments and each treatment consisted of 15 replications. The treatment was the dose of turmeric powder supplementation in the commercial feed i.e., 0, 240, 480, and 960 mg/100 g feed. The experimental catfish were fed two times a day in the morning and in the evening as much as 3% of body weight. The results showed that turmeric powder supplementation at a dose of 240 mg/100 g feed could inhibit liver cell destruction in the catfish as was indicated by the increase in body weight, liver weight, and DNA concentrations of liver, as well as the decrease in the liver malondialdehyde, and serum glutamic piruvic transaminase, serum glutamic oxaloacetic transaminase concentrations. Turmeric powder supplementation can be used in oviparous animals and fish to increase liver function and capacity to synthesize vitellogenin to be deposited in the developing follicles during reproduction phase.
Identification of growth hormone (GH) gene in a target fish is the first step in the construction of “all fish genes transfer vector” to generate transgenic fish. The research was done to identify and characterize the GH gene of Pangasionodon hypophthalmus. There were several activities performed in identifying the GH gene: RNA extraction, cDNA synthesis, PCR amplification, and DNA fragment isolation. The characterizations were done using the nucleotide sequencing engine ABIPRISM 3100. The results were then analyzed using BLASTN/P and GENETYX version 7 program. The full-length GH gene of P. hypophthalmus was 1151 bp in length, coding for an open reading frame (ORF) of 603 bp. The 5’ and 3’ untranslated regions of the GH gene were 22 bp and 526 bp long, respectively. The GH gene of P. hypophthalmus had some common characteristics that are owned by GH genes, such as single tryptophan residue (W) on the 104th amino acid, 5 cysteine residues (C) on the amino acid 71, 135, 173, 190, and 198 and a motif of Asn-Xaa-Thr on C terminus which is the potential location for N-linked glycosilation. Polyadenylation signal (aataaa) was on the 14 bp at the upstream of polyadenylation location. Growth hormone of P. hypophthalmus consisted of over 200 amino acids from GH cDNA deduction. The highest proportion of amino acid composition was leusin (14%) while the lowest was tryptophan (0.5%).
The success of transgenic fish production has been achieved through eggs fertilization using electroporated sperms carrying exogenous DNA. This study was conducted in order to obtain the optimal electroporation condition for stripped catfish sperm. A plasmid containing green fluorescent protein (GFP) gene driven by carp β-actin promoter was transferred into sperm using electrophoresis method towards transgenic stripped catfish (Pangasionodon hypophthalmus) production. Electroporation was carried out using square wave shock with pulse length of 30 ms and pulse interval of 0.1 sec. Treatments are combination between voltage (50 V, 75 V, and 100 V) and pulse number (1 and 3). Exogenous DNA concentration used was 10 μg/mL of Tris-EDTA. Results showed that increasing the voltage from 50 to 100 decreased sperm motility, while pulse number did not affect sperm motility. Voltage of 50 gave the best motility of sperm, although sperm viability relatively similar between treatments and control except at 100 V with 3 pulses number. Further, electroporation-treated sperms were able to fertilize eggs. Higher hatching rate of eggs was obtained in electroporation treatment at 50 V with pulse number of 1 and 3. The persistence of transferred GFP was detected in electroporated and incubated sperms (control). However, GFP was only detected in larvae from eggs that were fertilized by electroporated sperm. Thus, electroporation could be applied to produce transgenic stripped catfish.
