Zhihengliuella alba sp. nov., and emended description of the genus Zhihengliuella

Abstract: A Gram-positive, non-motile, white-pigmented, short rod actinobacterium, designated YIM 90734 T , was isolated from a saline soil sample collected from Ganjiahu Suosuo Forest National Nature Reserve in Xinjiang province, north-west China, and its taxonomic position was investigated by using a polyphasic approach. Strain YIM 90734 T grew optimally at 28-37 6C and pH 6.0-8.0 and in 5 % (w/v) NaCl. The peptidoglycan type was A4a, L-Lys-L-Ala-L-Glu and tyvelose and mannose were the major cell-wall sugars. The pred… Show more

“…The amino acid content of the cell wall was determined according to Staneck & Roberts (1974) and the sugars of the whole-cell hydrolysate were analysed as described by Tang et al (2009). For other chemotaxonomic analyses, strain MBRL 353 T was grown in tryptic soy broth shake flasks for 1 week at 28 u C. Cell biomass was harvested by centrifugation, washed with distilled water and lyophilized.…”

“…Analyses of the diaminopimelic acid isomer and whole-cell sugars were performed as described by Hasegawa et al (1983) and Tang et al (2009), respectively. Polar lipids were extracted and separated by two-dimensional TLC (Minnikin et al, 1979) and analysed using the procedures described by Collins & Jones (1980).…”

“…Purified cell-walls were obtained by disruption of cells with glass beads and subsequent trypsin digestion, according to the method of Schleifer (1985). Amino acids and peptides in the cell-wall hydrolysates were analysed by using twodimensional TLC on cellulose plates with the solvent systems described by Schleifer & Kandler (1972) and the HPLC system described by Tang et al (2009). Sugars from whole-cell hydrolysates were analysed as described by Staneck & Roberts (1974).…”
