Somatic embryo proliferation, maturation and germination in Catharanthus roseus

Abstract: In the present study an efficient somatic embryogenesis method has been developed in Catharanthus roseus. Friable embryogenic callus was induced from hypocotyl of in vitro germinated seeds on Murashige and Skoog basal nutrient media supplemented with various auxins particularly 2,4-D (1.0 mg l )1 ). However, only NAA (1.0 mg l )1 ) produced somatic embryos in cultures. Embryo proliferation was even high on the same medium added with BAP. Cotyledonary somatic embryo germinated and converted into plantlets in BA… Show more

“…1f). Similar to our observation, germination of somatic embryos on BAP containing medium has also been reported in Arachis hypogaea (Venkatachalam et al 1999), Quassia amara (Martin and Madassery 2005) and Catharanthus roseus (Junaid et al 2006). In the present study, precocious germination of somatic embryos and profuse rooting was observed during germination of embryos (Fig.…”

“…Moreover, satisfactory embryo germination was observed on MS with a low (0.5 µM) concentration of NAA (data not shown in Table 2 desiccation caused by the increased osmoticum in the solid medium. Junaid et al (2006) reported that somatic embryo maturation and germination in Catharanthus roseaus occurred when 3.0% sucrose and 2.22 μM BAP supplement MS was used. In our study, SEs that were induced in a 2,4-D-containing medium developed only shoots but failed to produce any roots.…”

“…Means followed by the same letter within columns are not significantly different (P = 0.05) by Tukey's test A Number of somatic embryos at early cotyledonary stage however, immature/juvenile plant parts are generally reported to be more responsive. Hypocotyl segments, excised from axenic seedlings, have been used effectively by many workers to achieve indirect SE in several woody and herbaceous plant species (Sunnichan et al 1998;Ashok Kumar et al 2002;Junaid et al 2006). In P. marsupium, callus was induced from hypocotyl explants by the application of 2,4-D and BA either alone or in combination.…”
