PCR‐RFLP and DAMD‐PCR genotyping for Salvia species

Abstract: BACKGROUND: Identification of genotypes in Salvia is complicated owing to the morphological similarity and common occurrence of natural hybridisation within Salvia species. Species-and genotype-specific DNA markers are very useful for plant identification, breeding and preservation programmes and can also provide a general overview on the prediction of plant essential oil yield.

“…RFLP has been used to evaluate the variability among species-or genotype-specific sequences (Nalini et al, 2001;Van Droogenbroeck et al, 2004;Karaca et al, 2008). In the present study, RFLP using three restriction enzymes (AluI, RsaI and Sau3AI) revealed distinctive RFLP profiles among the ITS1-5.8S-ITS-2 regions of the three species (Fig.…”

“…As a result, various molecular methods such as RAPD (Boehm et al, 1999;Kim and Choi, 2003), AFLP (Kim et al, 2005), RFLP (Karaca et al, 2008), ITS/NTS (Ngan et al, 1999;Cui et al, 2003;Xu et al, 2009), SSR (Kim et al, 2007) and SCAR (Warude et al, 2006;Choi et al, 2007;Choo et al, 2009), have been utilized to authenticate and distinguish medicinal plants from their relatives and adulterants. However, with the exception of, C. officinale (diploid, 2n=2x=22) and L. chuanxiong (triploid, 2n=3x=33), the cytological background of medicinal plants rarely show cytogenetic relationships (To, 1969(To, , 1971.…”

“…However, molecular biological data may provide a Deans, 1999 solution to the taxonomic problems both in intraspecific level and among the thyme species. The use of DNA marker techniques was proven to be important in the case of polymorph species with limited reliable taxonomic characters (Karaca et al, 2008;Ince et al, 2009). Echeverrigaray et al (2001) studied the essential oil composition and genetic variability of six commercial Thymus vulgaris cultivars.…”
