Species delimitation and genetic diversity analysis in Salvia with the use of ISSR molecular markers

Abstract: -Thirty-nine plant specimens of six Salvia species were collected from different localities of the Alborz mountain region in Iran and studied for morphological and genetic variability and species relationship. Inter simple sequence repeats (ISSR) molecular markers showed a high degree of within-species and interspecifi c genetic variability in Salvia. Analysis of molecular variance and Hickory tests showed significant molecular difference among the studied populations. A principal coordinate analysis plot of m… Show more

“…So far, there have been few attempts to study the genetic variation in Salvia species using different molecular markers [19][20][21]. However, no studies have been conducted to assess the genotypic differences and relationships in Salvia species using gene-targeted markers, and this is the novelty of the current work.…”

“…Different clustering and ordination methods like, UPGMA (Unweighted paired group using average), and Ward (Minimum spherical variance) clustering as well as principal coordinate analysis (PCoA) were used for grouping of the species. To identify the most variable morphological characters, Principal components analysis (PCA), was used (PODANI, 2000;SAFAEI et al, 2016).…”

“…(FREELAND et al, 2011). ISSRs have been successfully used to estimate population genetic diversity and structure in different plant species (SHEIDAI et al, 2014a;KOOHDAR et al, 2015;MINAEIFAR et al, 2016;MOSAFERI et al, 2015), and to delimit the species (SAFAEI et al, 2016), and identify infra-specific taxonomic forms (SHEIDAI et al, 2014b).…”

“…Plant species delimitation and interspecific genetic diversity are two important areas of investigation in phylogenetic systematics, evolution, biogeography and biodiversity studies. Data obtained can help to understand the patterns and mechanisms of speciation and hybridisation (Minaeifar et al 2016, Safaei et al 2016. They can reveal the pattern of gene flow between closely related phylogenetic species versus isolation by distance and identify the evolutionary process by which new biological species arise (Freeland et al 2011.…”

“…These include both neutral multilocus markers (e.g., SSRs (simple sequence repeats), ISSRs (inter-simple sequence repeats) and AFLPs (amplified fragments length polymorphism)), etc. as well as DNA sequences (e.g., ITS (internal transcribed sequences) of ribosomal DNA and chloroplast DNA) (see for example, Sheidai et al 2012, Koohdar et al 2015, Mosaferi et al 2015, Minaeifar et al 2016, Safaei et al 2016.…”
