Biodiversitas 20: 3705-3714. Sweet sorghum (Sorghum bicolor (L.) Moench) belongs to cereal plants that have prospects for commercial development in Indonesia, since it can adapt widely in sub-optimal land, and has many functions. However, there is still limited sweet sorghum variety that has been released in Indonesia. The study aimed to evaluate the influence of genetic x environment interaction on agronomic characters and yield components of sweet sorghum mutant strain. Adaptation test was held in 8 locations in Indonesia, namely Pekalongan, Bantul, Gunung Kidul, Lampung, Malang, East Lombok, Bontobili and Maros, from May to November 2017. The material tested was 10 M7 mutant strains and two check varieties (Numbu and Super 1). The experiment was arranged in a randomized complete block design (RCBD). There were three replications in form of blocks and each block consisted of 40 sorghum plants. The result showed that genetic x environment interaction affected the agronomic characters and yield components of sweet sorghum. Several characters such as plant height, panicle diameter, seed production, sugar brix and stem juice volume of sweet sorghum mutants were greater than check varieties. The highest seed yield was found in MB-1 strain of sweet sorghum mutant that cultivated in Pekalongan site.

Molecular Characteristics of Cassava Carvita 25 Somaclonal Variant Using SSR Marker

Cassava is one of the most important food commodities besides rice and corn. Carvita 25 is a somaclonal variation that was induced from Adira IV cassava variety. Our study aimed to analyze the genetic differences of Carvita 25 compared to Adira IV by using SSR markers. Two cassava varieties, Carvita 25 and Adira IV, were used as plant materials and eleven SSRY primers were used to amplifythe fragments of cassava DNA. The results showed that SSRY 151 primer produced the highest polymorphic band (85.71%) where 6 out of 7 alels were polymoprphics with the length size from 120 to 600 bp. Of the total 56 of polymorphic bands, 26 alels were previously present in Adira IV but then it can not be found in Carvita 25, while 30 other bands were new fragments that were previously not present in Adira IV but then were present in Carvita 25. These genetic differencesof both Adira IV and Carvita 25 were also strengthened by the Jacard similarity value. The Jaccard similarity between Carvita 25 and Adira IV were 0.40-0.50, while the similarity between plants of Carvita 25 were 0.79-0.87, and in plants of Adira IV were 0.98- 1.These values showed thewide genetic difference between Adira IV and a somaclonal variation of Carvita 25.Keywords: cassava, Carvita 25, polymorphic, somaclonal variation, SSR Marker.

Rice is a staple food crop in Indonesia, while the need is increasing due to high rice consumption as well as population increase. The problems can be solved through increase of national rice production. Productivity of lowland and upland should be increased intensively and other potential dry area outside Java and Bali Islands should be considered for extending the area of production. Recently, high yielding variety such as semi dwarf variety, hybrid rice, and new plant type of rice were being developed by Indonesian breeders. However, new method is needed to complement conventional breeding method in order to accelerate rice breeding. Anther culture is one of in vitro culture techniques that can be used to accelerate the obtainment of pure lines through doubled-haploids (DHs) regenerated at first generation of culture for less than one year. Thus, application of anther culture in conventional breeding will increase the efficiency of selection process as well as reducing the cost for labour, land and breeder’s time. The obtainment of green plantlets derived from anther culture of indica rice subspecies has been improved by the addition of 1 mM putrescine into induction and regeneration media. Recently, several upland rice lines tolerant to abiotic stresses (i.e. low light intensity and aluminum toxicity) and biotic stresses (i.e. leaf and neck blast), several lowland rice/paddy lines tolerant to biotic stresses (i.e. bacterial leaf blight and blast), and several hybrid parental lines (i.e. male sterile, maintainer and restorer) were obtained in 2-3 years from several rice breeding program involving anther culture. However, potential use anther culture to provide unique genetic material for mapping populations for use in functional genomics and molecular breeding has not been explored. The results indicated that anther culture is a feasible technology that can be used for accelerating rice breeding program in Indonesia.
