Sensitivity-dependent Hierarchical Receptor Codes for Odors

In order to comprehend the strategy of odor encoding by odorant receptors, we isolated 2740 mouse receptor neurons from four olfactory epithelial zones and classified them in terms of their sensitivities and tuning specificities to a chiral pair of odorants, S(+)-carvone (caraway-like odor) and R(-)-carvone (spearmint-like odor). Our approach revealed that the majority of receptors at the lowest effective stimulus concentration represented the principal odor qualities characteristic of each enantiomer by means of the principal odor qualities of the odorants for which the receptors were most sensitive. The chiral-non-discriminating receptors were newly recruited 3.7 times of R(-)-carvone-sensitive receptors and totally became 2.8 times (39/14) of R(-)carvone-sensitive receptors in the subpopulations when the stimulus concentration was increased 10-fold [corrected]. More than 80% of the responsive receptors (an estimated 70 +/- alpha types) exhibited overlapping sensitivities between the enantiomers. The signals from the non-discriminating receptors may be reduced to decode the characteristic odor identity for R(-)-carvone in the brain over an adequate range of stimulus strengths. The information processing of odors appears to involve the selective weighting of the signals from the most sensitive receptors. An analysis of the overall receptor codes to carvones indicated that the system employs hierarchical receptor codes: principal odor qualities are encoded by the most sensitive receptors and lower-ranked odor qualities by less sensitive receptors.

Das Geruchsphanomen theoretisch zu erfassen, ist bisher weder von der physiologischen noch von der chemischen Seite her befriedigend gelungen. Nach einer Ubersicht uber die bisherigen Theorien des Riechvorganges wird versucht, das vorliegende Material einheitlich zu deuten. Es scheint, als ob eine Moglichkeit besteht, durch Kombination der Wirkung einer osmophoren Gruppe und dem EinfluR des Mumlichen Aufbaus der Molekeln zu einem Verstandnis des Geruchsphanomens zu gelangen. EinleitungIm Zusammenhang rnit der Synthese einiger Di-n-alkylacetaldehyde') haben wir versucht, uns rnit dem Problem ,,Konstitution und Geruch" grundstitzlich auseinanderzusetzen. Dabei ist die vielleicht gr66te Schwierigkeit die, daB es bisher k e i n e M a B e i n h e i t fur die Art des Geruchseindruckes gibt. Es ist deshalb nicht verwunderlich, da6 von Parfiimeuren immer wieder Vorschllge zur Geruchsklassifizierung gemacht worden sind, die aber stets mit sehr vagen Begriffen wie aromatisch, brenzlig, terpenoid usw. arbeiten. Fur eine exakte Untersuchung sind sie nicht verwertbar. Die wichtigsten derartigen G e r u c h s k l a s s if i z i e r u n g e n sind diejenigen von Linnt! und Zwaardemaker2), Keller3), Crocker und Henderson4# 6), Kru$), Bollmann7), RoberF) und BillotB). Sehr viele Substanzen sind durch ihren Geruch bereits in Konzentrationen nachweisbar, bei denen jede andere analytische Reaktion versagt (Jonon z. B. ist nach Thorl~lornson~~) noch in der Konzentration von 1,0 . 10-7 mg/l Luft wahrnehmbar); die noch eben einen Reiz ausubenden (minimum perceptibile) und erkennbaren (Erkennungsschwelle) Konzentrationen an Riechstoffen konnen mit mehr oder weniger komplizierten Apparaturen bestimmt werden. Solche Bestimmungen wurden u. a. von Zwaardemaker21 ' l ) , Komuso12), F . B . Hofmann und Kohlrausch18), Morimura14) und Redgrove's) beschrieben. Vor kurzem hat aber Le Magnenls) darauf aufmerksam gemacht, da6 die Geruchsempfindlichkeit von Mannern und Frauen speziell fur Exaltolid verschieden ist. Er fand, da6 die Reizschwelle fur Manner zeitlich unabhlngig ist; bei Frauen dagegenbei absolut vie1 gro-Berer Empfindlichkeit gegen Exaltolidzeigt sich ein rhythmisches Schwanken der Hohe der Reizschwelle in Abhingigkeit vom Menstruationscyclus und einer eventuellen Schwangerschaft. Mechanismus und Theorien des GeruchesZunachst sind bei der Frage nach dem Mechanismus des Riechens drei VorgBnge zu unterscheiden, l. die Reizung des Riechnerven, des Olfactorius, an seinen Enden, 2. die Reizung etwaiger benachbarter Nerven, wodurch eine Reeinflussung des Geruchseindruckes entstehen kann, und 3. die Weiterleitung des Nervenreizes an das Gehirn. Beim letzten Vorgang handelt es sich wahrscheinlich um einen elektrochemischen Mechanismus (PranckI7). Fur den Mechanismus der Reizung des Olfactorius und damit
