Genetic Diversity Analysis of 14 Potato Genotypes Based on Morphological Characters and SSR Markers

Abstract:
Potato is one of important carbohydrate sources used as an alternative crop in Indonesia. The challenges in national potato breeding program included low productivity, less tolerance to environmental stresses, and narrow genetic diversity. The purpose of this study was to analyze genetic diversity of 14 potato genotypes based on morphological characters and SSR
markers newly developed from genome sequences of Indonesian potato genotypes. Principal component analysis of morphological data wa… Show more

“…We use SSR as one of the most widely used marker suitable for MAS [20]. The use of SSR markers on potato has been done on Nugroho et al [13,22] research, and its SSR primer can be open accessed on Indonesian Agricultural Genome Center (Pusat Genom Pertanian Indonesia) [23]. We hope through this present study, in the future, screening and identification of Granola L. variety on potato seed production can be carried out at the upstream level (in vitro planlets).…”

“…Unbalancing consumption demand and production of potatoes will force the government to continue importing potatoes. Therefore, the development of superior potato varieties with high productivity, disease resistance, and suitability for cultivation in Indonesia needs to be implemented as a current breeding strategy [5].…”

“…SSR markers can efficiently facilitate the establishment of genetic linkages due to their high polymorphism level, codominant inheritance, high allele diversity level, randomly wide distribution in the genome, multi-allele, and experimentally reproducible and transferable among related species [9], [10]. Some molecular markers have been applied to the genetic diversity study on potatoes in Indonesia [5], [11]- [15], but only few reported with SSR markers. Therefore, this present study was aimed to quantify the genetic diversity among 29 elite potato varieties in Indonesia and 4 clones that originated from the United States using the 12 SSR markers and to identify the potential genotypes for potato breeding programs.…”
