Low molecular weight siderophores are used by many living organisms to scavenge scarcely available ferric iron. Presence of at least a single siderophore-based iron acquisition system is usually acknowledged as a virulence-associated trait and a pre-requisite to become an efficient and successful pathogen. Currently, it is assumed that yersiniabactin (Ybt) is the solely functional endogenous siderophore iron uptake system in highly virulent Yersinia (Yersinia pestis, Y. pseudotuberculosis, and Y. enterocolitica biotype 1B). Genes responsible for biosynthesis, transport, and regulation of the yersiniabactin (ybt) production are clustered on a mobile genetic element, the High-Pathogenicity Island (HPI) that is responsible for broad dissemination of the ybt genes in Enterobacteriaceae. However, the ybt gene cluster is absent from nearly half of Y. pseudotuberculosis O3 isolates and epidemic Y. pseudotuberculosis O1 isolates responsible for the Far East Scarlet-like Fever. Several potential siderophore-mediated iron uptake gene clusters are documented in Yersinia genomes, however, neither of them have been proven to be functional. It has been suggested that at least two siderophores alternative to Ybt may operate in the highly virulent Yersinia pestis/Y. pseudotuberculosis group, and are referred to as pseudochelin (Pch) and yersiniachelin (Ych). Furthermore, most sporadic Y. pseudotuberculosis O1 strains possess gene clusters encoding all three iron scavenging systems. Thus, the Ybt system appears not to be the sole endogenous siderophore iron uptake system in the highly virulent yersiniae and may be efficiently substituted and/or supplemented by alternative iron siderophore scavenging systems.
Insertion sequence IS100 was localized on a 9.5-kb plasmid of Yersinia pestis and was shown to be specific for Y. pestis and serotype I strains of Y. pseudotuberculosis. The nucleotide sequence of IS100 isolated from this plasmid was determined. The element, which was flanked by 5-bp direct repeats, contained 1953 bp including imperfect inverted terminal repeats of 52 and 61 bp long (43 bp were identical). Two open reading frames encoding potential polypeptides of 340 and 252 amino acids were identified on one DNA strand. Nucleotide sequence as well as deduced polypeptide sequences of IS100 were homologous to those of IS21, IS232 and IS640.

МИКРОБИОЛОГИЯ 852017, issue 3 известно, что структурный компонент клеточ-ной стенки грамотрицательных бактерий -липопо-лисахарид (лпс) -является основным патогенети-ческим фактором чумного микроба [1,11,12]. лпс относится к биологически активным веществам опо-средованного действия. для проявления его токсиче-ских свойств необходимо отделение лпс от внеш-ней мембраны бактерий и представление рецепто-рам иммунокомпетентных клеток макроорганизма в свободной функционально-активной форме [11]. по современным представлениям источником свобод-ной формы лпс в условиях макроорганизма явля-ются не разрушенные, а живые бактерии, способные выделять лпс во внешнюю среду, подобно секре-ции экзотоксинов белковой природы. примером мо-жет служить возбудитель Klebsiella pneumonia [13]. вирулентные штаммы этого патогена продуцируют экстрацеллюлярный комплекс капсульного вещества, в состав которого входит токсический компонент -лпс. Формирование этого комплекса происходит на всех фазах роста бактерий и является естественным продуктом их жизнедеятельности. что касается воз-будителя чумы, известно, что большинство факторов, определяющих его вирулентные свойства, кодирует-ся тремя резидентными плазмидами -pMT1, pCD1 и pPCP1 [2,4, 9]. как правило, это белки, которые экс-понируются на поверхности мембраны бактерий или же секретируются за ее пределы. так, плазмида pCD1 Пробл. особо опасных инф. 2017; 3:85-89 (pCD1), Y. pestis EV76 (pPCP1). о присутствии внеклеточной формы лпс в среде икубации клеток Y. pestis EV76 судили по токсичности супернатантов для биопробных животных и по реакции LAL-теста. результаты и выводы. установлено, что экстрацеллюлярную форму лпс образуют 37-градусные культуры Y. pestis EV76 полноценного штамма и вариантов, содержащих pMT1 или pCD1 плазмиды. культуры, лишенные плазмид, и вариант, содержащий плазмиду pPCP1, такой способностью не обладают. по результатам LAL-теста процесс отделения лпс от мембраны клеточной стенки во внешнюю среду сопряжен с транслокацией белков, кодируе-мых плазмидами pMT1и pCD1, и является естественной формой жизнедеятельности клеток чумного микроба. участие плазмиды pCD1 в реализации токсического потенциала лпс Y. pestis установлено впервые.Ключевые слова: Yersinia pestis, плазмиды, экстрацеллюлярная форма липополисахарида, токсигенность. Pathogenic bacteria use low-molecular-weight iron chelators – siderophores – to assimilate iron in the host body. Being recognized as virulence factors, these molecules, differing in structural and functional properties, are the subject of the most intensive research in medical microbiology. The present study is devoted to the investigation of yersiniachelin siderophore (Ych) found in the causative agent of plague, Yersinia pestis. The aim of the work was to clarify the role of Ych in the physiology of Y. pestis by comparing the properties of three strains of the plague microbe, differing in Ych production. Materials and methods. Three variants of Y. pestis EV76 strain were used in the experiments: parent strain Y. pestis EV76, its mutant that does not produce Ych due to deletion of three siderophore biosynthesis genes (analogues of ypo1530–1532 in Y. pestis CO92 strain) and a complemented mutant that was transformed by a recombinant pSC-A-5EV plasmid containing Ych biosynthesis genes cloned into the high-copy plasmid vector pSC-A-amp/kan. Comparative analysis of the three strains was carried out in terms of colony morphology, siderophore activity, growth rate, and sensitivity to hydrogen peroxide. Results and discussion. The comparison of these strains has revealed that the secretion of Ych by bacteria at 26 °С ensures the assimilation of iron. At 37 °С, Ych is not secreted into the medium and protects bacteria from the bactericidal action of reactive oxygen compounds. Thus, the study shows that yersiniachelin is able to stimulate the assimilation of iron by bacteria under iron-deficit conditions and has antioxidant properties.
This review systematizes and analyzes the data published over the past decade, devoted to the study of low-molecular-weight high affinity iron chelators – siderophores. Siderophores, which are found in bacteria, fungi and mammals, are able to extract iron from insoluble inorganic compounds, and in the host organism – from complexes with proteins that perform the function of nonspecific protection of mammals from infections. The extracted iron is delivered to cells through surface protein receptors specific for each siderophore, as well as various protein transport systems that make up membranes. Siderophores play an important role in virulence in pathogenic bacteria, performing many functions in the host organism, in addition to providing microbes with iron and other biological metals. They participate in the storage of excess iron, toxic to cells, protect bacteria from reactive oxygen compounds, compete for iron with phagocytes, and have a harmful effect on host cells, acting as secreted bacterial toxin in some cases. Bacterial siderophores perform a signaling function and regulate both, their own synthesis and the synthesis of other virulence factors. Many pathogenic bacteria produce several siderophores that are active under different conditions, against various sources of iron in the host organism and at different stages of infectious process. The review presents the results of the experimental studies aimed at elucidating the structure and diverse functions of bacterial siderophores, the mechanisms of their biosynthesis and regulation of expression, as well as the role of these molecules in the physiology and virulence of pathogenic bacteria. Special emphasis is put on siderophores of bacteria causing particularly dangerous infections.
Aim. Elucidation of the role of extrachromosomal elements of heredity in manifestations of toxic properties of Yersinia pestis. Materials and methods. The study was carried out in vaccine strain Y. pestis EV76 (pMTl, pCDl, pPCPl) and non-plasmid variants of vaccine EV76 (pMTl\ pCDl', pPCPl') and virulent 231 (рМТГ, pCDl’, pPCPl') strains of Y. pestis. Presence of functionally active form of lipopolysaccharide (LPS) in the incubation medium of the bacteria was evaluated via toxicity of supernatant of Y. pestis for intact animals (infection-toxic shock) and mice sensitized by D-GalN. Results. 37°C cultures of Y. pestis EV76 containing a full amount of plasmids were established to release LPS into the environment. Non-plasmid variants of both vaccine and virulent strains of Y. pestis pMTl', pCD Г, рРСР 1 do not have this ability. Separation of LPS from cell wall was detected in live bacteria of plague infectious agent. This process is assumed to be coupled with translocation of proteins coded by pMTl, pCDl, pPCPl plasmids from the cell into the environment. Conclusion. Functional interconnection between extrachromosomal elements of heredity and toxic activity of Y. pestis LPS is established for the first time.
