ýêñïåðèìåíòàõ íà êðûñàõ ïîêàçàíî, ÷òî íîâîå ïðîèçâîäíîå 3-ãèäðîêñèïèðèäèíà (3-ÃÏ) 2-ýòèë-6-ìåòèë-3-ãèäðîêñèïèðèäèíà L-àñïàðàãèíàò (3-ÃÏÀ) â íåáîëüøèõ äîçàõ (1 è 5 ìã/êã) ïîâûøàåò ôèçè÷åñêóþ ðàáîòîñïîñîáíîñòü aeèâîòíûõ ïî òåñòàì áåãà â òðåòáàíå è ïëàâàíèÿ â áàññåéíå, ïðåâîñõîäÿ èëè íå óñòóïàÿ ïî âûðàaeåííîñòè äåéñòâèÿ ïðåïàðàòàì ñðàâíåíèÿ àêòîïðîòåêòîðàì ìåòàïðîòó è ëàäàñòåíó, ïðèìåíÿåìûì â çíà÷è-òåëüíî áîëüøèõ äîçàõ. Ïðè ýòîì èíãèáèòîð ãëþêîíåîãåíåçà òðèïòîôàí â çíà÷èòåëüíîé ñòåïåíè -íà (74 ± 5) % (p < 0,01) ïðåäîòâðàùàë ñòèìóëèðóþùåå âëèÿíèå 3-ÃÏÀ íà ôèçè÷åñêóþ ðàáîòîñïîñîáíîñòü êðûñ ïî òåñòó áåãà â òðåòáàíå. Íà ðàçíûõ ìîäåëÿõ àì-íåçèè ó ìûøåé 3-ÃÏÀ â áîëåå âûñîêîé äîçå (30 ìã/êã) îêàçûâàë âûðàaeåííîå àíòèàì-íåñòè÷åñêîå äåéñòâèå, ïðåâîñõîäÿ â ýòîì îòíîøåíèè ïðåïàðàòû ñðàâíåíèÿ ìåêñèäîë (äðóãîå ïðîèçâîäíîå 3-ÃÏ) â äîçå 30 ìã/êã è íîîòðîï ïèðàöåòàì â äîçå 200 ìã/êã è íå óñòóïàÿ èì â äîçàõ 50 è 800 ìã/êã ñîîòâåòñòâåííî. Íà ìîäåëè èøåìè÷åñêîãî èíñóëüòà ó êðûñ 3-ÃÏÀ (30 ìã/êã/ñóò) îêàçûâàë îò÷åòëèâîå íåéðîïðîòåêòîðíîå äåéñòâèå, ïðå-âîñõîäÿ ìåêñèäîë (50 ìã/êã/ñóò), â îòíîøåíèè óìåíüøåíèÿ âûðàaeåííîñòè íåâðîëîãè-÷åñêîãî äåôèöèòà.Êëþ÷åâûå ñëîâà: íîâîå ïðîèçâîäíîå 3-ãèäðîêñèïèðèäèíà; ôèçè÷åñêàÿ ðàáîòîñïî-ñîáíîñòü; àíòèàìíåñòè÷åñêèå è íåéðîïðîòåêòîðíûå ñâîéñòâà; êðûñû; ìûøè. ÂÂÅÄÅÍÈÅ ðåçóëüòàòå ìíîãîëåòíèõ èññëåäîâàíèé áûëî ñîç-äàíî íåñêîëüêî îðèãèíàëüíûõ îòå÷åñòâåííûõ ëåêàðñò-âåííûõ âåùåñòâ (ËÂ), ïðèíàäëåaeàùèõ ê êëàññó 3-ãèä-ðîêñèïèðèäèíà (3-ÃÏ). Ïåðâûì Ë ÿâèëñÿ ìåòèëýòèë-ïèðèäèíîë (2-ýòèë-6-ìåòèë-3-ÃÏ ãèäðîõëîðèä), äåéñò-âóþùåå íà÷àëî ëåêàðñòâåííîãî ïðåïàðàòà ýìîêñèïèí, îáëàäàþùèé ìåìáðàíîïðîòåêòîðíûìè, àíòèîêñèäàíò-íûìè, ïðîòèâîãèïîêñè÷åñêèìè, àíãèîïðîòåêòîðíûìè, àíòèàãðåãàíòíûìè, ðåòèíîïðîòåêòîðíûìè è äðóãèìè ñâîéñòâàìè [6]. Ýìîêñèïèí ñåãîäíÿ, ïîìèìî îôòàëü-ìîëîãèè, ïðèìåíÿþò â êëèíèêå âíóòðåííèõ áîëåçíåé, íåâðîëîãèè, íåéðîõèðóðãèè, èñïîëüçóþò â êîìïëåêñ-íîé ôàðìàêîëîãè÷åñêîé çàùèòå ïî÷êè îò èøåìè÷åñêî-ãî è îïåðàöèîííîãî ñòðåññà, à òàêaeå è â äðóãèõ îáëàñ-òÿõ ìåäèöèíû.Âòîðûì Ë ñòàë ýòèëìåòèëãèäðîêñèïèðèäèíà ñóê-öèíàò (2-ýòèë-6-ìåòèë-3-ÃÏ ñóêöèíàò) -äåéñòâóþùåå íà÷àëî ëåêàðñòâåííûõ ïðåïàðàòîâ ìåêñèäîë, ìåêñè-êîð, íåéðîêñ è äð. Îí èìååò ìåìáðàíîïðîòåêòîðíûé è àíòèîêñèäàíòíûé ìåõàíèçì äåéñòâèÿ, îáëàäàåò îïðå-äåëåííûì ñïåêòðîì ôàðìàêîëîãè÷åñêèõ ñâîéñòâ.  ÷à-ñòíîñòè, îí îêàçûâàåò íîîòðîïíîå, íåéðî-, êàðäèî-, ýíäîòåëèî-è ãåïàòîïðîòåêòîðíîå, àíêñèîëèòè÷åñêîå, ïðîòèâîãèïîêñè÷åñêîå, àíòèñòðåññîðíîå, àíòèàëêî-ãîëüíîå, ïðîòèâîïàðêèíñîíè÷åñêîå, ïðîòèâîñóäîðîae-íîå, îáåçáîëèâàþùåå, ìèåëîïðîòåêòîðíîå, íåôðîïðî-òåêòîðíîå è âåãåòîòðîïíîå äåéñòâèå, óëó÷øàåò ìîçãî-âîå êðîâîîáðàùåíèå, óìåíüøàåò àãðåãàöèþ òðîìáîöè-òîâ, ñíèaeàåò óðîâåíü ãëèêåìèè è îáùåå ñîäåðaeàíèå õîëåñòåðèíà, îáëàäàåò àíòèàòåðîñêëåðîòè÷åñêèì ýô-ôåêòîì, óñòðàíÿåò äåôèöèò ôóíêöèé ìîçãà ïðè ñòàðå-íèè, à òàêaeå ñïîñîáåí óñèëèâàòü äåéñòâèå èçâåñòíûõ ïðåïàðàòîâ è äð. [3,4,6,12].  íàñòîÿùåå âðåìÿ ëåêàð-ñòâåííûå ïðåïàðàòû, ñîçäàííûå íà îñíîâå ýòîãî ËÂ, øèðîêî è óñïåøíî ïðèìåíÿþò â ðàçëè÷íûõ îáëàñòÿõ ìåäèöèíû: íåâðîëîãèè, êëèíèêå âíóòðåííèõ áîëåçíåé, ïñèõèàòðèè, íàðêîëî...
Íà ìîäåëè èøåìè÷åñêîãî èíñóëüòà ó êðûñ îáíàðóaeåíî, ÷òî íîâàÿ ñìåñü òðèïåïòèäîâ (ÍÑÒ -H-Lys-Asp-Glu-OH, H-Asp-Glu-Pro-OH, H-Asp-Glu-Arg-OH) â äîçàõ 150 è 300 ìêã/êã/ñóò îêàçûâàåò îò÷åòëèâîå íåéðîïðîòåêòîðíîå äåéñòâèå, ïðåâîñõîäÿ â 1,1 ðàçà ïðåïàðàò ñðàâíåíèÿ ñåìàêñ â îòíîøåíèè óìåíüøåíèÿ íåâðîëîãè÷åñêîãî äåôèöèòà. Ó ìûøåé â ãåðìî-è áàðîêàìåðå ÍÑÒ (10, 50 è 150 ìêã/êã) îáëàäàåò âûðàaeåííûìè ïðîòèâîãèïîêñè÷åñêèìè ñâîéñòâàìè, ïðåâîñõîäÿ ñåìàêñ â ãåðìîêàìåðå â äîçàõ 10 è 50 ìêã/êã â 1,3 è 1,5 ðàçà, ñîîòâåòñòâåííî, à â áàðîêàìåðå -â äîçå 150 ìêã/êã â 1,9 ðàçà. Íà ìîäåëè ñêîïîëàìèíîâîé àìíåçèè ó êðûñ ÍÑÒ (50 è 150 ìêã/êã) äàåò âûðàaeåííûé àíòèàìíåñòè÷åñêèé ýôôåêò, ïðåâîñõîäÿ ñåìàêñ â àíàëîãè÷íûõ äîçàõ ïî âëèÿíèþ íà ëàòåíòíûé ïåðèîä çàõîäà â òåìíóþ êàìåðó â 1,5 è 1,4 ðàçà, ñîîòâåòñòâåííî. Íà ìîäåëÿõ àìíåçèè ó ìûøåé, âûçâàííîé ýëåêòðîñóäîðîaeíûì øîêîì è êîìïëåêñíûì ýêñòðåìàëüíûì âîçäåéñòâèåì, ÍÑÒ (50 è 150 ìêã/êã) òàêaeå îêàçûâàåò âûðàaeåííîå àíòèàì-íåñòè÷åñêîå äåéñòâèå, ïðåâîñõîäÿ ñåìàêñ íà ïåðâîé ìîäåëè â îáåèõ äîçàõ â 4 ðàçà, à íà âòîðîé -â äîçå 150 ìêã/êã â 2,9 ðàçà. ÍÑÒ (50 ìêã/êã) ó êðûñ óâåëè÷èâàåò àìïëèòóäó òðàíñêàëëîçàëüíûõ âûçâàííûõ ïîòåíöèàëîâ ñåíñîìîòîðíîé êîðû áîëüøîãî ìîçãà íà 69 %, ïðåâîñõîäÿ ñåìàêñ ïî âûðàaeåííîñòè (â 1,3 ðàçà) è äëèòåëüíîñòè (â 1,4 ðàçà) äåéñòâèÿ. Ñëåäîâàòåëüíî, ÍÑÒ ÿâëÿåòñÿ ïåðñïåêòèâíûì íåéðîòðîïíûì ñðåäñòâîì c íåéðîïðîòåêòîðíîé, ïðîòèâîãèïîêñè÷åñêîé è àíòèàìíåñòè÷åñêîé àêòèâíîñòüþ. NMT (10, 50 and 150 mg/kg) had marked antihypoxic effect on mice in hermetic and altitude chamber. NMT in doses of 10 and 50 mg/kg was more effective than semax in hermetic chamber (1.3 and 1.5 times, respectively) and in a dose of 150 mg/kg in altitude chamber (1.9 times). NMT (50 and 150 mg/kg) had also marked antiamnesic effect on model amnesia caused by scopolamine in rats and was more effective (1.5 and 1.4 times, respectively) than semax in equal doses. NMT (50 and 150 mg/kg) also had marked antiamnesic effect on model amnesia caused by maximal electroshock and complex extreme factors in mice and in both doses was 4 times more effective than semax on the first model and in a dose of 150 mg/kg was 2.9 times more effective on the second model. В экспериментах на бодрствующих необездвиженных кроликах новый отечественный лекарственный препарат эмопаг (при пневмомикроинъекции) оказывал прямое влияние на большинство (71 %) нейронов сенсомоторной коры большого мозга: он вызывал как тормозные (у 40 % клеток), так и активационные (у 31 % клеток) реакции, в отличие от мексидола, который, главным образом, угнетал спонтанную активность нейронов (у 56 % клеток). На фоне действия специфического блокатора глутаматных AMPA-рецепторов NBQX возбуждающее влияние эмопага на спонтанную активность у 93 % клеток полностью предотвращалось или существенно ослаблялось. У тех же кроликов, находящихся в условиях общей анестезии барбитуратами, эмопаг оказывал прямое влияние на большинство (87 %) клеток Пуркинье, вызывая как возбуждение (у 49 % нейронов), так и их угнетение (у 38 %). На фоне действия NBQX возбуждающее влияние эмопага на спонтанную активность у 100 % нейронов полностью предотвращалось или существенно ослаблялось. Следовательно, и сенсомоторная кора большого мозга, и кора мозжечка играют важную роль в реализации центрального действия эмопага у животных. При этом его возбуждающий эффект обусловлен активацией AMPA-рецепторов.
На переживающих срезах гиппокампа крыс эмопаг и мексидол в концентрации 4 мМ подавляли ортодромные популяционные ответы в поле СA1 на 58 ± 4 % и 43 ± 3 % соответственно. Специфический неконкурентный антагонист NMDA-рецепторного комплекса МК-801 ослаблял угнетающее действие эмопага на 38 ± 4 %, а мексидола — на 73 ± 6 %, действуя в отношении первого в 1,9 раза менее выраженно. Специфический блокатор глутаматных AMPA-рецепторов CNQX практически полностью блокировал депрессирующий эффект эмопага либо ослаблял его на 54 ± 5 %, действуя по крайней мере в 1,4 раза более выраженно, чем МК-801. В пирамидных нейронах поля СA1 гиппокампа крысят эмопаг (2 мМ) угнетал возбуждающий постсинаптический ток, вызванный раздражением коллатералей Шаффера, на 92 ± 3 %, а на фоне действия специфического блокатора глутаматных AMPA-рецепторов NBQX его депрессирующий эффект практически полностью ослаблялся на 95 ± 4 %. Следовательно, можно предполагать, что гиппокамп играет существенную роль в реализации центрального действия эмопага. При этом его эффект реализуется через активацию AMPA-рецепторов.
В экспериментах на аутбредных мышах-самцах исследована антигипоксическая активность новых производных бензотиазола. Установлено, что проявление указанного действия напрямую определяется не только типом заместителя в гетероцикле, но и фрагментом органической кислоты в составе молекулы. Наиболее выраженный эффект наблюдается в случае присутствия в структуре молекулы фрагмента янтарной кислоты.
